Den foreliggende opfindelse angår hidtil ukendte 1-cyclopropy l-5-amino-6,8-dif luor-7-piperazinyl-l, 4-dihydro- 4-oxo-quinolin-3-carboxylsyre-derivater med den i krav 1 angivne formel (I) med meget høj antibakteriel virkning, 5 fremgangsmåder til fremstilling af disse forbindelser samt et farmaceutisk præparat indeholdende dem.
I JP offentliggørelsesskrift nr. 58-174.367 (hvoraf der foreligger et sammendrag i Derwent World Patent Index, Accession No. 83-823272) er det beskrevet, at forbindelser 10 repræsenteret ved den almene formel NH 0 (10) ,= R-N^

Disse forbindelser har imidlertid ingen aminogruppe i 5-stillingen i quinolinringen, og forbindelserne ifølge den foreliggende opfindelse har højere anti-mycoplasma-akti-vitet end de kendte forbindelser (18).
15 Det fremgår af de i det følgende angivne resultater af sammenligningsforsøg, at repræsentative forbindelser kendt fra de ovenfor omtalte EP offentliggørelsesskrifter nr. 113.093, nr. 132.845 og nr. 160.578 har en væsentligt lavere antibakteriel aktivitet end repræsentative forbindel-20 ser ifølge den foreliggende opfindelse. Det fremgår desuden af de også i det følgende angivne resultater af sammenligningsforsøg, at en typisk forbindelse ifølge JP-A-58.174.367 og typisk forbindelser ifølge EP-A-126.355 har væsentligt lavere antibakteriel aktivitet end forbindelser ifølge opfin-25 delsen; det fremgår også, at anti-mycoplasma-virkningen af forbindelser ifølge opfindelsen er væsentligt højere end virkningen af forbindelser kendt fra det sidstnævnte EP-skrift.
Med den foreliggende opfindelse tilvejebringes der 30 således hidtil ukendte quinolinderivater med høj antibakteriel aktivitet mod såvel gram-positive som gram-negative bakterier, visse estere af disse derivater og farmaceutisk acceptable salte af disse forbindelser, ligesom der tilvejebringes en fremgangsmåde til fremstilling af forbindel-35 serne samt et farmaceutisk præparat indeholdende dem.
De her omhandlede quinolinderivater er ejendommelige * DK 170593 B1 5 ved, at de har formlen

De her omhandlede forbindelser har asymmetriske car-bonatomer på piperazinringen ved 7-stillingen og kan derfor 10 forekomme i optisk aktive former. De omfatter således D-isomere, L-isomere og blandinger deraf.
En af forbindelserne ifølge opfindelsen har to asymmetriske carbonatomer på piperazinringen ved 7-stillingen og kan derfor forekomme som stereoisomere med forskellige 15 konfigurationer (cis eller trans). Disse stereisomere og deres blandinger er også omfattet af de her omhandlede forbindelser.

Forbindelserne ifølge opfindelsen udviser fremragende antibakteriel virkning og et bredt antibakterielt spektrum 10 i in vitro-forsøg. Endvidere udviser disse forbindelser en fremragende infektions-forsvarende virkning in vivo mod topiske eller systemiske infektioner forårsaget af grampositive og gram-negative bakterier.
De her omhandlede forbindelser har også fremragende 15 anti-mycoplasma-virkning.
Fremgangsmåderne til fremstilling af de her omhandlede forbindelser fremgår af krav 3's kendetegnende del og beskrives nærmere i det følgende: A. Substitutionsreaktion med pioerazinderivater 20 De her omhandlede forbindelser kan fremstilles ved at omsætte en carboxylsyre eller dens ester (fortrinsvis en C1_5-alkylester), som er repræsenteret ved følgende almene formel 9- NH2 o P Λ 30 i hvilken X er et halogenatom, og Y er et hydrogenatom eller Ci_5-alkyl, med en forbindelse med den almene formel h3c ) \

Eksempler på halogenatomet X i formlen (II) er fluor-, chlor- eller bromatomer.
Denne reaktion kan udføres ved omrøring af udgangs-5 forbindelserne (II) og (III) ved 10-180°C i fra 10 minutter til 24 timer i et indifferent opløsningsmiddel. Eksempler på det indifferente opløsningsmiddel omfatter alkoholer, såsom ethanol, ethere, såsom dioxan, tetrahydrofuran og 1,2-dimethoxyethan, aromatiske carbonhydrider, såsom benzen, 10 toluen og xylen, acetonitril, dimethylformamid, dimethylsulf-oxid, pyridin og vand.
Generelt udføres den ovennævnte reaktion i nærværelse af en syreacceptor under anvendelse af udgangsforbindelsen med formlen (III) i en mængde, der er ækvivalent ned eller 15 er en smule i overskud i forhold til udgangsforbindelsen (II). Om ønsket kan udgangsforbindelsen (III) anvendes i overskud for at få den til at virke samtiig som syreacceptor. Eksempler på syreacceptorer er natriumhydrogencarbonat, natriumcarbonat, kaliumcarbonat, triethylamin, pyridin og 20 picolin.
Udgangsforbindelserne (II) og (III), der anvendes i denne reaktion, kan, om muligt, anvendes på beskyttede former, og efter reaktionen fjernes beskyttelsesgrupperne på gængs måde. Beskyttelsesgrupperne kan være enhver beskyttel-25 sesgruppe, som kan fjernes uden ødelæggelse af de her omhandlede, ved reaktionen dannede forbindelsers struktur. Grupper, som sædvanligvis anvendes som beskyttelsesgrupper for aminogruppen inden for det kemiske område vedrørende peptider, aminosukkere, nucleinsyrer eller /?-lactamforbin-30 delser, kan anvendes til den foreliggende opfindelses formål.
Aminobeskyttelsesgrupperne kan fraspaltes ved solvo-lyse (herunder hydrolyse) eller reduktion, afhængigt af beskyttelsesgruppernes egenskaber.
Specifikke eksempler på beskyttelsesgrupper, som kan 35 elimineres ved solvolyse, omfatter acylgrupper, såsom formyl, acetyl og trifluoracetyl, substituerede eller usubstituerede DK 170593 B1 9 alkoxycarbonylgrupper, såsom ethoxycarbonyl, tert-butoxy-carbonyl, benzyloxycarbonyl, p-methoxybenzyloxycarbonyl og β-(p-toluensulfonyl)-ethoxycarbonyl, en tritylgruppe, en trimethylsilylgruppe, en o-nitrophenylsulfenylgruppe, en 5 diphenylphosphinylgruppe og en tetrahydropyranylgruppe.
Reaktionen udføres i et opløsningsmiddel ved 0-150°C i nærværelse eller fraværelse af en katalysator, såsom en syre eller base.
Eksempler på syren er uorganiske syrer, såsom salt-10 syre, hydrogenbromidsyre, svovlsyre og phosphorsyre, organiske syrer, såsom eddikesyre, trifluoreddikesyre, myresyre og toluensulfonsyre, og Lewis-syrer, såsom bortribromid og aluminiumchlorid. Eksempler på basen er alkalimetal- og jordalkalimetalhydroxider, såsom natriumhydroxid og barium-15 hydroxid, alkalimetalcarbonater, f.eks. natrium- og kalium-carbonat, alkalimetalalkoxider, såsom natriummethoxid og natriumethoxid, og natriumacetat. Sædvanligvis anvendes vand som opløsningsmiddel. Afhængigt af forbindelsens egenskaber kan der anvendes et andet opløsningsmiddel, såsom 20 ethanol, dioxan, ethylenglycoldimethylether, benzen eller eddikesyre, eller et blandet opløsningsmiddel af et sådant opløsningsmiddel og vand.
Eksempler på beskyttelsesgrupper, som kan elimineres ved reduktion, omfatter arylsulfonylgrupper, såsom p-toluen-25 sulfonyl, en methylgruppe, som er substitueret med phenyl eller benzyloxy, såsom benzyl, trityl eller benzyloxymethyl, arylmethoxycarbonylgrupper, såsom benzyloxycarbonyl og p-methoxybenzyloxycarbonyl, og halogenethoxycarbonylgrupper, såsom /3,/3,/3-trichlorethoxycarbonyl- og /J-iodo-ethoxycarb-3 0 onylgrupper.
Ved denne reaktion anvendes forskellige reaktionsbetingelser, afhængigt af beskyttelsesgruppens egenskaber med hensyn til eliminering. Eksempelvis gennemføres denne ved behandling af forbindelsen med en hydrogenstrøm i et indif-35 ferent opløsningsmiddel ved 10-60°C i nærværelse af en katalysator, såsom platin, palladium eller Raney-nikkel, eller DK 170593 B1 10 ved at behandle den med metallisk natrium i flydende ammoniak ved -50 til -20°C, eller ved at behandle den med et metal, såsom zink i eddikesyre eller i en alkohol, såsom methanol. Eksempler på opløsningsmidlet ved den katalytiske reduktion 5 kan omfatte ethylenglycoldimethylether, dioxan, dimethylform-amid, ethanol, ethylacetat og eddikesyre.
Udgangsforbindelsen (II) kan fremstilles ved de fremgangsmåder, som er beskrevet i referenceeksemplerne 1 og 2 eller ved metoder, som i alt væsentligt er i overensstemmelse 10 hermed.
B. Aminerinosreaktion
Forbindelserne ifølge opfindelsen kan fremstilles ved at omsætte en carboxylsyre eller dens Ci_5-alkylester, som er repræsenteret ved følgende almene formel Η» / F λ (IV) 20 yj Δ

i hvilken R, X og Y har de ovenfor angivne betydninger, med 25 ammoniak.
Denne reaktion kan udføres ved at bringe udgangsforbindelsen (IV) i kontakt med ammoniak i 1-50 timer ved en temperatur på 50-150'C i et indifferent opløsningsmiddel, f.eks. en alkohol, såsom ethanol, pyridin, dimethylformamid 30 eller vand, fortrinsvis i et forseglet rør.
Denne reaktion gennemføres i nærværelse af en syre-acceptor under anvendelse af ammoniak i en mængde, der er ækvivalent med eller er i ringe overskud i forhold til udgangsforbindelsen (IV). Hensigtsmæssigt anvendes ammoniak i 35 overskud for også at få den til at virke som syreacceptor.
Et salt, såsom ammoniumacetat, kan bringes til at virke i DK 170593 B1 11 stedet for ammoniak.
Udgangsforbindelsen (IV), som anvendes ved denne reaktion, kan, om muligt, anvendes på en form, som er beskyttet med en sådan beskyttelsesgruppe som beskrevet ovenfor i 5 forbindelse med reaktion A, og efter reaktionen elimineres beskyttelsesgruppen på gængs måde.
Udgangsforbindelsen (IV) er hidtil ukendt og kan fremstilles ved den i referenceeksempel 3 beskrevne metode eller ved metoder, der i det væsentlige svarer dertil.
10 Når de her omhandlede forbindelser, som fås ved de ovennævnte fremgangsmåder, er estere, kan de omdannes til forbindelser med formlen (I) ved hydrolyse af esterdelen på gængs måde. Om nødvendigt, kan forbindelserne med formlen (I) esterificeres på gængs måde 15 til dannelse af estere af forbindelserne med formlen (I).
Farmaceutisk acceptable salte · af forbindelserne med formlen (I) eller deres estere kan fremstilles ved at behandle forbindelserne med formlen (I) eller estere deraf med syrer, eller ved at behandle for-20 bindeiserne (I) med baser eller metalsalte. Syrer, som er egnede til saltdannelse, omfatter f.eks. saltsyre, phosphor-syre, eddikesyre, mælkesyre, oxalsyre, ravsyre, methan-sulfonsyre, maleinsyre, malonsyre, gluconsyre, asparagin-syre og glutaminsyre. Baser eller metalsalte, som er egnede 25 til saltdannelse, omfatter f.eks. metalhydroxider, såsom natriumhydroxid og kaliumhydroxid, metalcarbonater, såsom natriumcarbonat og kaliumcarbonat, zinkchlorid, zinksulfat, zinknitrat og sølvnitrat.
De her omhandlede forbindelser, som er fremstillet 30 som anført ovenfor, isoleres og renses på gængs måde, og afhængigt af isolerings- og rensningsbetingelserne kan de fås i form af et salt eller en fri syre. De kan omdannes til hinanden til fremstilling af de her omhandlede forbindelser i de ønskede former.
35 Stereoisomerene (cis- og trans-former) af de her om handlede forbindelser kan isoleres ved en konventionel 0 DK 170593 B1 12 metode, såsom fraktioneret krystallisation eller chromato-grafi. Det er muligt at fremstille de her omhandlede forbindelser med en cis- eller trans-konfiguration ved de ovenfor beskrevne reaktioner under anvendelse af udgangs-5 forbindelserne med en cis- eller trans-konfiguration.
De optisk aktive isomere af de her omhandlede forbindelser kan adskilles ved kendte metoder.
De således fremkomne forbindelser (I), deres estere og salte af disse af alle hidtil ukendte forbindelser.
10 Især er forbindelserne (I) og deres salte værdifulde som antibakterielle midler, eftersom de har meget høj anti-bakteriel aktivitet. Forbindelserne (I) og deres salte kan ikke blot anvendes som medicin til mennesker og dyr, men også som fiskemedicin, landbrugskemikalier og næringsmid-15 delkonserveringsmidler.
Esterne af forbindelserne (I) er naturligvis værdifulde som udgangsmaterialer til syntese af forbindelserne (I). Når esterne på enkel måde kan omdannes til forbindelserne (I) in vivo, kan de udvise en tilsvarende 20 virkning og er også anvendelige som antibakterielle midler.
Når de her omhandlede forbindelser anvendes som antibakterielle midler til mennesker, anbefales det, at de indgives i en dosis på 5 mg til 5 g pr. dag én eller flere gange daglig, selvom dosen kan varieres afhængigt af 25 alder, legemsvægt og patientens symptomer, indgivelsesmåden etc. Forbindelserne kan indgives oralt eller paren-teralt.
De her omhandlede forbindelser kan indgives i den fremkomne pulverform, men de indgives sædvanligvis i form 30 af et farmaceutisk præparat sammen med farmaceutisk acceptable hjælpestoffer. Specifikke eksempler på de farmaceutiske præparater er tabletter, opløsninger, kapsler, granulater, fingranulater, pellets, pulvere, sirupper, injektionspræparater og salver. Disse farmaceutiske præparater fremstilles ved i og for sig kendte metoder. Hjælpestoffer til oral indgift er sådanne, som sædvanligvis anvendes 0 13 DK 170593 B1 inden for området af farmaceutiske præparater og ikke reagerer med de her omhandlede forbindelser, såsom stivelse, mannitol, krystallinsk cellulose, CMC-Na, vand og ethanol. Hjælpestoffer til injektionspræparater er 5 sådanne, som sædvanligvis anvendes inden for området, såsom vand, isotonisk natriumchloridopløsning, glucose-opløsning og transfusionsopløsning.
De ovennævnte flydende præparater og salver kan også anvendes til lokale behandlinger i oto-rhino-laryngologi 10 eller ophthalmologi.

Den minimale inhiberingskoncentration bestemmes ved en 25 dobbelt agar-fortyndingsmetode ved anvendelse af Chanock-agar. Tre μΐ af en kultur af forsøgsorganismer i Chanock-medium inokuleres på 10 ml medikament-indeholdende agarlag i petriskåle. Væksten af Mycoplasma observeres efter inkubation ved 37°C under de nedenfor viste betingelser.

Eksempel 14
Den antibakterielle virkning af forbindelsen 1 mod Campylobacter gejuni er vist i tabel IV. Tallene i < tabellen viser de minimale inhiberingskoncentrationer 5 (MIC) (pg/ml), beregnet for fri base. Den minimale inhi-beringskoncentration bestemmes ved den dobbelteagar-fortyndingsmetode ved anvendelse af blod-indeholdende Mueller-Hinton-agar. En øskenfuld af en kultur af forsøgsorganismer i Mueller-Hinton-medium inokuleres på 10 ml 10 medikament-indeholdende agar-lag i petriskåle. Bakteriel vækst observeres efter 48 timers inkubation ved 37°C mikroaerobt. MIC defineres som den laveste medikamentkoncentration, som forhindrer synlig bakteriel vækst.

Eksempel 15 (akut toksicitet)
En opløsning indeholdende hver af forbindelserne 1 og 2 ifølge opfindelsen i forskellige koncentrationer indgives oralt til hanmus (ddY) i en dosis på 0,1 5 ml pr. 10 g legemsvægt. Antallet af døde mus tælles efter 7 dage, og værdien af den gennemsnitlige dødelige dosis (LD^Qf mg/kg) beregnes i overensstemmelse med Behrens-Kaerber-metoden. Resultaterne er vist i tabel V.
