1. Campo de la invencion
En la presente memoria se proporcionan derivados de quinazolinona 5-sustituidos. Tambien se desvelan composiciones farmaceuticas que comprenden los compuestos y metodos para tratar, prevenir y controlar diversos trastornos.
2. Antecedentes de la invencion
2.1 Patobiologia del cancer y otras enfermedades
El cancer se caracteriza principalmente por un incremento en el numero de celulas anomalas derivadas de un tejido normal determinado, la invasion de tejidos adyacentes por estas celulas anomalas o la extension linfatica o transportada por la sangre de celulas malignas a nodulos linfaticos regionales y a zonas distales (metastasis). Datos clmicos y estudios de biologfa molecular indican que el cancer es un proceso multietapa que comienza con pequenos cambios preneoplasicos, que en ciertas condiciones pueden avanzar hasta neoplasia. La lesion neoplasica puede progresar clonalmente y desarrollar una capacidad en aumento para la invasion, crecimiento, metastasis y heterogeneidad, especialmente en condiciones en las que celulas neoplasicas escapan de la vigilancia inmunitaria del huesped. En la bibliograffa medica existe una gran variedad de canceres descritos con detalle. Los ejemplos incluyen cancer de pulmon, colon, recto, prostata, mama, cerebro e intestino. La incidencia del cancer continua ascendiendo con el envejecimiento general de la poblacion, el desarrollo de nuevos canceres y el crecimiento de poblaciones susceptibles (por ejemplo, personas infectadas con SIDA o excesivamente expuestas a la luz solar). Sin embargo, las opciones para el tratamiento del cancer son limitadas. Por ejemplo, en el caso de canceres de sangre (por ejemplo, mieloma multiple), existen pocas opciones de tratamiento disponibles, especialmente cuando falla la quimioterapia convencional y el trasplante de medula osea no es una opcion. Como alternativa, las celulas tumorales pueden liberar peptidos angiogenicos a traves de la produccion de proteasas y la ruptura posterior de la matriz extracelular donde se almacenan algunas citocinas (por ejemplo, b-FGF). La angiogenesis tambien puede inducirse indirectamente a traves del reclutamiento de celulas inflamatorias (particularmente macrofagos) y su posterior liberacion de citocinas angiogenicas (por ejemplo, TNF-a, b-FGF).
Otras diversas enfermedades y trastornos tambien se asocian con, o se caracterizan por, una angiogenesis no deseada. Por ejemplo, la angiogenesis aumentada o mal regulada se ha relacionado con diversas enfermedades y afecciones medicas que incluyen, pero sin limitacion, enfermedades neovasculares oculares, enfermedades neovasculares coroidales, enfermedades neovasculares retinales, rubeosis (neovascularizacion del angulo), enfermedades virales, enfermedades geneticas, enfermedades inflamatorias, enfermedades alergicas y enfermedades autoinmunes. Los ejemplos de tales enfermedades y afecciones incluyen, pero sin limitacion: retinopatfa diabetica; retinopatfa del prematuro; rechazo de injerto corneal; glaucoma neovascular; fibroplasia retrolental; artritis y vitreorrenopatfa proliferativa.
Por consiguiente, compuestos que puedan controlar la angiogenesis o inhibir la produccion de determinadas citocinas, incluyendo TNFa, pueden ser utiles en el tratamiento y prevencion de diversas enfermedades y afecciones.
2.2 Metodo para tratar el cancer
La terapia actual contra el cancer puede implicar cirugfa, quimioterapia, terapia hormonal y/o tratamiento por radiacion para erradicar celulas neoplasicas en un paciente (vease, por ejemplo, Stockdale, 1998, Medicine, vol. 3, Rubonstein y Federman, eds., Capftulo 12, Seccion IV). Recientemente, la terapia contra el cancer podna tambien implicar terapia biologica o inmunoterapia. Todos estos planteamientos poseen inconvenientes significativos para el paciente. celulas cancerosas. Las terapias biologicas y las inmunoterapias estan limitadas en numero y pueden producir efectos secundarios tales como erupciones o inflamaciones, smtomas similares a gripe, incluyendo fiebre, escalofnos y fatiga, problemas del tracto digestivo o reacciones alergicas.
Con respecto a la quimioterapia, existe una diversidad de agentes quimioterapeuticos disponibles para el tratamiento del cancer. o una sal, solvato o estereoisomero farmaceuticamente aceptable del mismo, para su uso en un metodo para el tratamiento, control o prevencion de un cancer de piel, pulmon, ovario, prostata, colon, recto, cerebro, cabeza y cuello, garganta, pancreas, huesos, hngado o vejiga, en donde:
Rd es:
hidrogeno;
alquilo (Ci-Ca), opcionalmente sustituido con uno o mas halo;
-C(O)-alquilo (C1-C8), en donde el alquilo esta opcionalmente sustituido con uno o mas halo; -C(O)-(CH2)n-(cicloalquilo C3-C10);
-C(O)-(CH2)n-NReRf, en donde Re y Rf son cada uno independientemente: hidrogeno;
alquilo (C1-Ca), opcionalmente sustituido con uno o mas halo; o alcoxi (C1-Ca), opcionalmente sustituido con uno o mas halo; o -C(O)-(CH2)n-O-alquilo (C1-Ca).
R7 es: hidrogeno; -(CH2)nOH; fenilo; -O-alquilo (C1-Ca); o alquilo (C1-Ca), opcionalmente sustituido con uno o mas halo;
R8 es: hidrogeno; o alquilo (C1-Ca), opcionalmente sustituido con uno o mas halo; y n es 0, 1 o 2.
5. El compuesto para el uso de la reivindicacion 4, o una sal, solvato o estereoisomero farmaceuticamente aceptable del mismo,
en donde R7 es metilo; o en donde Rd es -C(O)-alquilo (C1-C6); o 5 en donde Rd es -C(O)-CH2-O-alquilo (C1-C6).

o una sal, solvato o estereoisomero farmaceuticamente aceptable del mismo, para su uso en un metodo para el tratamiento, control o prevencion de un cancer de piel, pulmon, ovario, prostata, colon, recto, cerebro, cabeza y cuello, garganta, pancreas, huesos, hngado o vejiga, en donde:

-(CH2)n-(arilo de 6 a 10 miembros);
-C(O)-(CH2)n-(arilo de 6 a 10 miembros) o -C(O)-(CH2)n-(heteroarilo de 6 a 10 miembros), en donde el arilo o heteroarilo esta opcionalmente sustituido con uno o mas de: halo; -SCF3; alquilo (C-i-Ca), el mismo opcionalmente sustituido con uno o mas halo; o alcoxi (C-rCa), el mismo opcionalmente sustituido con uno o mas halo;
-C(O)-(CH2)n-NHRh, en donde Rh es:
arilo de 6 a 10 miembros, opcionalmente sustituido con uno o mas de: halo; alquilo (C-rCa), el mismo opcionalmente sustituido con uno o mas halo; o alcoxi (C1-C6), el mismo opcionalmente sustituido con uno o mas halo; o
-C(O)-(CH2)n-O-(CH2)n-(arilo de 6 a 10 miembros);
R9 es: hidrogeno; -(CH2)nOH; fenilo; -O-alquilo (C1-C6); o alquilo (C1-C6), opcionalmente sustituido con uno o mas halo;
R10 es: hidrogeno; o alquilo (C1-C6), opcionalmente sustituido con uno o mas halo; y n es 0, 1 o 2.
9. El compuesto para el uso de la reivindicacion 8, en donde R9 es metilo; o
en donde Rg es -C(O)-fenilo, -C(O)-CH2-fenilo, o -C(O)-NH-fenilo; opcionalmente en donde el fenilo esta sustituido con uno o mas de metilo, -CF3 o halogeno.
10. El compuesto para el uso de la reivindicacion 8, que es:
11. El compuesto para el uso de cualquiera de las reivindicaciones 1-10, en donde el cancer de piel es melanoma.

El compuesto para el uso de cualquiera de las reivindicaciones 1-10, en donde el cancer es adenocarcinoma rectal, carcinoma colorrectal no extirpable, carcinoma hepatocelular metastasico, carcinoma peritoneal, carcinoma seroso papilar, carcinoma hepatocelular no extirpable, carcinoma tiroideo papilar, carcinoma tiroideo folicular o carcinoma tiroideo medular.
5 13. El compuesto para el uso de la reivindicacion 11, en donde el melanoma es melanoma metastasico.
14. El compuesto para el uso de cualquiera de las reivindicaciones 1-10, donde el cancer es sarcoma de Kaposi, sarcoma ginecologico, sarcoma de tejidos blandos, sarcoma de tejidos blandos de alto riesgo extirpado o leiomiosarcoma.
