Abstract
Es wird ein Verfahren zur Erzeugung einer groszen Anzahl von Analoga eines gewaehlten Peptids beschrieben. Das Verfahren umfaszt die folgenden Schritte: Synthetisierung von zwei unterschiedlichen Gemischen von gleichlangen Oligonucleotiden, die an einigen bestimmten Positionen unterschiedliche Basen und am 3-Ende einige identische Basen besitzen, Mischen der beiden Gemische zur Bildung eines Gemischs von partiell doppelstraengigen, gleichlangen DNA-Sequenzen, die danach enzymatisch in vollstaendig doppelstraengige DNA-Sequenzen umgewandelt werden, Schneiden des zuletzt genannten Gemischs mit zwei verschiedenen Enzymen zur Erzeugung von DNA-Sequenzen, die an einen in aehnlicher Weise geschnittenen Vektor ligiert werden koennen, Inserieren der DNA-Sequenzen in den Vektor durch Ligation, um ein Gemisch von Vektoren zu erzeugen, die dann in einer an sich bekannten Art und Weise in einen Wirt transformiert werden, Vermehrung des Wirts zur Bildung von Kolonien, die nacheinander analysiert werden, um die DNA-Sequenzen festzustellen, die fuer ein einzelnes Protein codieren, unter Expressionsbedingungen separate Vermehrung der groszen Anzahl von Wirten zur Erzeugung einer groszen Anzahl von Analoga des gewaehlten Peptids. Auszerdem werden neue Analoga von VIP und ein Plasmid, das ein Gen enthaelt, das fuer eines der neuen Analoga von VIP codiert, offenbart.{Verfahren; Herstellung; Peptid; Oligonucleotide; Basen; 3-Ende; DNA-Sequenzen; Enzyme; Vektor}
