Description

【発明の詳細な説明】
［産業上の利用分野］ 本発明は高脂血症予防治療剤に関する。 ［従来の技術］ 血液中の脂質としてはコレステロール、トリグ
リセライド、燐脂質、遊離脂肪酸などが存在して
いる。代謝異常により、コレステロールなどの血
液中への遊離が増加すると、高脂血症を呈する。 高脂血症を予防治療するために、種々の薬剤が
使用されている。 しかしながら、これらの薬剤は副作用を有する
ものが多く、副作用のない薬剤が切望されてい
る。 ［発明の目的］ 本発明の目的は、副作用のない高脂血症予防治
療剤を提供することにある。 ［発明の目的を達成するための手段］ 本発明は上記目的を達成するためになされたも
のであり、本発明の高脂血症予防治療剤は、プロ
ナーゼを有効成分として含有することを特徴とす
るものである。 本発明の高脂血症予防治療剤は、通常の製剤技
術により、例えば錠剤、カプセル剤、散剤、顆粒
剤等に調製して経口的に、あるいは注射剤、坐
剤、軟膏剤等に調製して非経口的に投与できる。
本剤は、特に腸溶経口製剤とすることが好まし
い。 更に本剤は、他の薬剤、例えば他の肝臓薬、腎
臓薬、抗生物質、免疫賦活剤、抗腫瘍剤、降圧剤
等と併用できる。 投与量は疾病度、投与法、剤型等によつても異
なるが、経口投与の場合は、成人１人につき１日
当りプロナーゼの量で１〜5000mg投与することに
より、目的を達成することができる。 また、投与量をプロナーゼの酵素単位で表した
場合には、腸溶製剤にて１万〜50万チロジン単位
の経口投与により目的を達成することができる。 ［実施例］ 以下本発明の実施例について説明する。 製剤例 １ プロナーゼ20ｇ、乳糖40ｇを混合し、ヒドロキ
シプロピルメチルセルロース10ｇを加え顆粒に成
型したのち、酢酸フタル酸セルロースを用い均等
に被覆し、腸溶性顆粒とする。 製剤例 ２ 製剤例１で得たプロナーゼ腸溶性顆粒をカプセ
ルに充填し、カプセル剤とする。 臨床例 １ ［肝疾患を有する高脂血症に対するプロナーゼ
の効果］ ●患者：男性、昭和16年生 ●診断名：高脂血症、慢性肝炎、慢性腎炎 ●血液検査：脂質検査、肝機能及び腎機能とも異
常値を示した。 ●プロナーゼによる治療：エンピナースＰ（商品
名、科研製薬(株)製・１錠中プロナーゼ9000チ
ロジン単位含有）を１日12錠内服させた。第
１表に示すように49日後の血液検査にては、
肝機能指標のAL−Ｐとγ−GTP及び腎機能
指標のクレアチニンのみが異常値を示し、脂
肪値は全項目とも正常値を示した。なお72日
後、肝機能、腎機能及び脂肪値全て正常値を
示した。副作用は全く見られなかつた。

統計的観察１ ［肝疾患を有する高脂血症に対す
るプロナーゼの効果］ 肝疾患を有する高脂血症14例について、エンピ
ナースＰ（商品名、科研製薬(株)製・１錠中プロナ
ーゼ9000チロジン単位含有）を１日12錠内服させ
た。投薬後３ケ月以内に改善した各検査項目数を
第２表に示す。なお高脂血症14例中10例は３ケ月
以内に脂肪値全項目が正常となつた。副作用は全
く見られなかつた。
【表】 臨床例 ２ ［肝疾患を有しない高脂血症に対するプロナー
ゼの効果］ ●患者：女性、昭和６年生 ●診断名：高脂血症、高血圧症 ●血液検査：肝機能及び腎機能とも正常値を示
す。脂肪値に異常が見られる。 ●プロナーゼによる治療：エンピナースＰ（商品
名、科研製薬(株)製・１錠中プロナーゼ9000チ
ロジン単位含有）を１日12錠内服させた。第
３表に示すように約３ケ月後には脂肪値は正
常に復した。副作用は全く見られなかつた。

統計的観察２ ［肝疾患を有しない高脂血症に対
するプロナーゼの効果］ 肝疾患を有しない高脂血症13例ついて、エンピ
ーナスＰ（商品名、科研製薬(株)製、１錠中プロナ
ーゼ9000チロジン単位含有）を１日12錠内服させ
た。投薬後３ケ月以内に改善した各検査項目数を
第４表に示す。なお高脂血症13例中11例は３ケ月
以内に脂肪値全項目が正常となつた。副作用は全
く見られなかつた。

【表】 ［発明の効果］ 以上説明したように、プロナーゼを有効成分と
して含有する本発明の高脂血症予防治療剤は、高
脂血症に著効を示し、かつ副作用は全く見られな
いという顕著な効果を有する。
