Claims (1)
- 【特許請求の範囲】 1.(1)天然にはその選択可能遺伝子内に存在しないイントロンを含有する選 択可能遺伝子で真核宿主細胞をトランスフェクションする段階であって、該イン トロンが該宿主細胞中でスプライシングされて選択可能タンパク質をコード化す るメッセンジャーRNAを与えることができ、該選択可能遺伝子内の該イントロ ンの存在が該宿主細胞において該選択可能遺伝子から生産される選択可能タンパ ク質のレベルを減少させることを特徴とする段階と、(2)該選択可能タンパク 質によって付与される選択可能な表現型を有するトランスフェクタントを同定す る段階、からなる方法。 2.選択可能遺伝子が原核生物供給源から得られるものである請求項1の方法。 3.所望のタンパク質をコード化する生成物遺伝子で該宿主細胞をトランスフェ クションすることをさらに含む請求項1の方法。 4.第2の選択可能遺伝子で該宿主細胞をトランスフェクションすることをさら に含む請求項1の方法。 5.所望のタンパク質をコード化する生成物遺伝子と第2の選択可能遺伝子で該 宿主細胞をトランスフェクションすることをさらに含む請求項1の方法。 6.第2の選択可能遺伝子がハイグロマイシン耐性遺伝子またはDHFR遺伝子 である請求項4または請求項5の方法。 7.該イントロンの長さが55ないし85ヌクレオチドである請求項1の方法。 8.該イントロンの長さが65ないし75ヌクレオチドである請求項1の方法。 9.該選択可能遺伝子がハイグロマイシン耐性遺伝子またはネオマイシン耐性遺 伝子である請求項1の方法。 10.該イントロンが該選択可能遺伝子のコード領域内に存在する請求項1の方 法。 11.該イントロンが該選択可能遺伝子の転写される非コード領域内に存在する 請求項1の方法。 12.該真核細胞宿主が哺乳類細胞である請求項1の方法。 13.該哺乳類細胞がチャイニーズ・ハムスター卵巣(CHO)細胞またはシミ アンCOS細胞またはヒト胚性腎臓293細胞である請求項12の方法。 14.請求項1の方法に従って作成されるトランスフェクタント細胞。 15.選択可能遺伝子とその選択可能遺伝子の転写される領域内のイントロンか らなる単離された核酸であって、該イントロンが該選択可能遺伝子内に天然には 存在せず、該イントロンが宿主細胞において該選択可能遺伝子から生産される選 択可能タンパク質のレベルを減少させるに足る長さであることを特徴とする単離 された核酸。 16.(1)真核宿主細胞を、 (a)その選択可能遺伝子内に天然には存在しないイントロンを含有する選択可 能遺伝子であって、該イントロンが該宿主細胞中でスプライシングされて選択可 能タンパク質をコード化するメッセンジャーRNAを与えることができ、該選択 可能遺伝子中の該イントロンの存在が、該宿主細胞において該選択可能遺伝子か ら生産される選択可能タンパク質のレベルを減少させる選択可能遺伝子、および 、 (b)所望のタンパク質をコード化する生成物遺伝子、で同時トランスフェクシ ョンし、 (2)該選択可能遺伝子によって付与される選択可能な表現型を有するトランス フェクタントを同定する、ことからなる方法。 17.同時トランスフェクションされた宿主細胞から該生成物遺伝子によってコ ード化されるポリペプチドを回収することをさらに含む請求項16の方法。
