Claims (1)
- 【特許請求の範囲】 1.複数の隣接する毛細路から輻射線の励起及び検知を行なうためのスキャナー であって、 平面に配置された複数の並んだ毛細路と、第一の波長の輻射エネルギー源と、 前記輻射エネルギーを受け、前記毛細路の平面の励起容積に焦点を合わせるため の対物レンズと、 前記励起容積が、順に反復的に前記毛細路のうちの一つの内にあって、前記毛細 路内の材料を励起し、材料に異なる波長のエネルギーを放射させるよう、前記毛 細路を移動させるための手段と、前記放射された輻射線を受け、信号を発生させ るための検知システムと、各々の前記毛細路内の励起容積にある材料を表わす出 力をもたらすため、前記信号を受信し、処理をするコンピュータ手段とを、備え 、 前記対物レンズは、前記輻射エネルギーを集め、この輻射エネルギーを、前記異 なる波長の放射された輻射エネルギーを透過させ、他の波長の輻射線を透過させ ないための共焦空間及び分光フィルター手段を含む光学システムに送る役割をす る、 ことを特徴とするスキャナー。 2.前記毛細路を移動させるための前記手段は、前記コンピュータによって制御 され、それにより、出力を毛細路と相関させることができることを特徴とする請 求項1記載のスキャナー。 3.毛細路を移動させるための手段は、連続的に移動し、それにより、各前記毛 細管におけるバンドを走査することを特徴とする請求項1記載のスキャナー。 4.前記毛細路を移動させるための前記手段は、毛細路を前記励起容積にステッ プさせることを特徴とする請求項1記載のスキャナー。 5.毛管路は、細長い円筒状の細管の部分であることを特徴とする請求項2記載 のスキャナー。 6.合焦した輻射エネルギーに晒すため、前記細管の領域を同一平面に並んだ関 係に保持するための手段を含んでいることを特徴とする請求項5記載のスキャナ ー。 7.前記細管の端は、個々に取り扱い及び負荷できるよう分離可能であることを 特徴とする請求項6記載のスキャナー。 8.前記励起輻射エネルギーは、複数の波長で輻射エネルギーを放射する材料を 励起し、前記光学システムは、異なる波長の放射された輻射エネルギーを異なる 位置に選択的に送るための複数の共焦空間及び分光フィルター手段と、各々選択 された波長の放射されたエネルギーを受け、対応する出力信号を提供する複数の 検知手段とを含んでいることを特徴とする請求項1記載のスキャナー。 9.毛細路内の試料から輻射線の励起及び検知を行なうための装置であって、同 一平面に並んだ関係にある複数の毛細路を提供する手段と、第一の波長の輻射線 で前記毛細路内の試料を励起するための輻射手段と、各毛細路内の試料を前記輻 射線で順に、反復的に励起するため、前記同一平面に並んだ複数の毛細路を移動 させるための手段と、前記試料から放射された輻射線を集めるための手段と、前 記励起輻射線に応じて前記毛細路内の試料によって放射された輻射線を検知する ための手段と、 検知された放射された輻射線を処理するため、及び前記移動手段の運動を制御し て前記毛細路内の試料を表わす二次元の出力を時間と位置の関数として提供する ためのコンピュータ手段とを、 備えていることを特徴とする装置。 10.前記毛管路は、細長い円筒状の細管の部分であることを特徴とする請求項 9記載のスキャナー。 H.合焦した輻射エネルギーに晒すため、前記細管の領域を同一平面に並んだ関 係に保持するための手段を含んでいることを特徴とする請求項10記載のスキャ ナー。 12.前記細管の端は、独立に取り扱い可能であることを特徴とする請求項9記 載のスキャナー。 13.小さな直径の細管内で分離することのできる蛍光標識した検体を検知する ためのスキャナーであって、 各々蛍光標識した検体を分離するための複数の細管と、前記標識した検体から蛍 光を励起する波長を有する輻射エネルギーのエネルギー源と、 前記輻射エネルギーを小さな容積に合焦するため、及び前記容積から放射された エネルギーを集めるためのレンズ手段と、前記複数の細管の領域を順に、反復的 に前記容積の輻射エネルギーに晒し、それにより、前記容積において蛍光標識さ れた検体の蛍光を生じさせるための手段と、 集められた蛍光輻射線を受け、前記容積にある検体を表わす出力をもたらすため の手段とを、備えていることを特徴とするスキャナー。 14.前記レンズ手段は、空間フィルターを含む共焦光学検知アセンブリの一部 をなす対物レンズを備えていることを特徴とする請求項13記載のスキャナー。 15.前記光学検知システムは、前記励起波長のエネルギーを排除するための空 間フィルターを含んでいることを特徴とする請求項14記載のスキャナー。 16.複数の細管内で電気泳動により分離されたDNA配置決定断片からの蛍光 を検知する方法であって、前記複数の細管の領域を、同一平面で並んだ関係に配 置することと、 前記細管における所定の容積を順に、反復的に、対物レンズによって所定の容積 に合焦された所定波長の光エネルギーで励起し、断片を異なる波長で蛍光発光さ せることと、 前記細管各々における前記所定の容積から蛍光放射された光を、前記対物レンズ で集めることと、 前記所定波長の光を排除し、前記放射光を通過させるため、異なる波長の前記蛍 光放射された光エネルギーを分光的及び空間的にろ過することと、前記細管各々 内の前記断片からの蛍光を表わす出力信号を発生させるため、ろ過した放射光を 検知装置に供すること、を含むことを特徴とする方法。 17.細管の一端が、配列決定断片を細管の端内に速やかに並行して負荷するこ とができるよう、分れていることを特徴とする請求項16記載の方法。 18.複数の隣接する毛細路から輻射線の励起及び検知を行なうためのスキャナ ーであって、平面に配置された複数の並んだ毛細路と、第一の波長の輻射エネル ギー源と、 前記輻射エネルギーを受け、前記毛細路の平面の励起容積に焦点を合わせるため の対物レンズと、 前記波長のエネルギーで前記励起容積を励起するため、前記輻射エネルギーを対 物レンズに送るためのビーム・スプリッターと、前記励起容積が、順に反復的に 前記毛細路のうちの一つの内にあって、前記毛細路内の材料を励起し、材料に異 なる波長のエネルギーを放射させるよう、前記毛細路を移動させるための手段と 、前記放射された輻射線を受け、信号を発生させるための検知システムと、 各々の前記毛細路内の励起容積にある材料を表わす出力をもたらすため、前記信 号を受信し、処理をするコンピュータ手段とを、備え、 前記対物レンズは、前記輻射エネルギーを集め、この輻射エネルギーを、前記異 なる波長の輻射エネルギーを通過させるビーム・スプリッターを通じて、前記異 なる波長の放射された輻射エネルギーを透過させ、他の波長の輻射線を透過させ ないための共焦空間及び分光フィルターを含む光学システムに送る役割をする、 ことを特徴とするスキャナー。 19.前記毛細路を移動させるための前記手段は、前記コンピュータによって制 御され、それにより、出力を毛細路と相関させることができることを特徴とする 請求項18記載のスキャナー。 20.毛細路を移動させるための前記手段は、連続的に移動し、それにより、各 前記毛細管におけるバンドを走査することを特徴とする請求項19記載のスキャ ナー。 21.前記毛細路を移動させるための前記手段は、毛細路を前記励起容積にステ ップさせることを特徴とする請求項18記載のスキャナー。 22.毛管路は、細長い円周状の細管の部分であることを特徴とする請求項19 記載のスキャナー。 23.合焦した輻射エネルギーに晒すため、前記細管の領域を同一平面に並んだ 関係に保持するための手段を含んでいることを特徴とする請求項22記載のスキ ャナー。 24.前記細管の端は、個々に取り扱い及び負荷できるよう分離可能であること を特徴とする請求項23記載のスキャナー。 25.前記励起輻射エネルギーは、複数の波長で輻射エネルギーを放射する材料 を励起し、前記光学システムは、複数の追加のビーム・スプリッター、異なる波 長の放射された輻射エネルギーを異なる位置に選択的に送るための共焦空間及び 分光フィルター手段、及び各々選択された波長の放射されたエネルギーを受け、 対応する出力信号を提供する複数の検知手段を含んでいることを特徴とする請求 項18記載のスキャナー。 26.複数の細管内で電気泳動により分離されたDNA配置決定断片からの蛍光 を検知する方法であって、前記複数の細管の領域を、同一平面で並んだ関係に配 置することと、 前記細管における所定の容積を順に、反復的に、対物レンズによって所定の容積 に合焦された所定波長の光エネルギーで励起し、断片を異なる波長で蛍光発光さ せることと、 前記細管各々における前記所定の容積から蛍光放射された光を、前記対物レンズ で集めることと、 前記所定波長の光及び散乱光を排除し、前記放射光を通過させるため、異なる波 長の前記蛍光放射された光エネルギーを分光的及び共焦的空間的にろ過すること と、 前記細管各々内の前記断片からの蛍光を表わす出力信号を発生させるため、ろ過 した放射光を検知装置に供すること、を含むことを特徴とする方法。 27.細管の一端が、配列決定断片を細管の端内に速やかに並行して負荷するこ とができるよう、分れていることを特徴とする請求項26記載の方法。 28.前記断片は、複数の波長で蛍光を発し、前記複数の波長の各々で前記蛍光 放射された光エネルギーを分光的及び共焦的空間的にろ過し、これを対応する検 知要素に供することを特徴とする請求項26記載の方法。
