Abstract
Description
Claims (16)
- 1.下記工程: (a)好ましい励起ピークを有する初期供与体染料と、好ましい発光ピークを有 する最終受容体染料とを含み、組合せ中の各染料が励起エネルギーの有意なエネ ルギ−伝達を可能にするために充分なスペクトル重複を有する染料組合せを選択 する工程と; (b)前記染料組合せをポリマー微小粒子中に配合する工程とを含む方法によっ て製造される蛍光微小粒子。
- 2.スペクトル重複が、約90%より大きいエネルギー伝達を可能にする請求項 1記載の微小粒子。
- 3.ポリマー微小粒子がポリスチレン、臭素化ポリスチレン、ニトロセルロース 、ポリアクリル酸、ポリアクリロニトリル、ポリアクリルアミド、ポリアクロレ イン、ポリジメチルシロキサン、ポリブタジエン、ポリイソブレン、ポリウレタ ン、ポリ酢酸ビニル、ポリ塩化ビニル、ポリビニルビリジン、ポリビニルペンジ ルクロリド、ポリビニルトルエン、ポリビニリデンクロリド、又はポリジビニル ベンゼンである請求項1記載の微小粒子。
- 4.蛍光染料がポリアザインダセン、クマリン、炭化水素もしくは置換炭化水素 染料;オキサゾールもしくはオキサジアゾール;アリール置換もしくはヘテロア リール置換ポリオレフィン(C2−C5オレフィン部分);カルボシアニン;フ タロシアニン;オキサジン;カルボスチリル;ポルフィリン染料;又はこれらの 組合せである請求項1記載の微小粒子。
- 5.少なくとも1種の蛍光染料が式: ▲数式、化学式、表等があります▼ 〔式中、R1〜R6は同一又は異なる基であり、単独の又は組合せた、水素、ハ ロゲン、アルキル、アルコキシ、アルケニル、シクロアルキル、アリールアルキ ル、アシル(これらのアルキル部分はそれぞれ、炭素数約20未満である)、ア リールもしくはヘテロアリールであり;R7は窒素、メチン、又はハロゲン−、 アルキル−、アルコキシ−、アルケニル−、シクロアルキル−、アリールアルキ ルー、アシル−(これらのアルキル部分はそれぞれ、炭素数約20未満である) 、アリール−もしくはヘテロアリール−置換メチンである]で示されるポリアザ インダセン染料である請求項4記載の微小粒子。
- 6.スペクトル重複が、約95%より大きいエネルギ−伝達を可能にする請求項 5記載の微小粒子。
- 7.蛍光染料組合せが約6種未満の染料を約0.5重量%〜2重量%の総染料濃 度で含む請求項1記載の微小粒子。
- 8.初期供与体対最終受容体の比が約1:5と約10:1との間である請求項1 記載の微小粒子。
- 9.ポリマ−微小粒子がポリスチレン、ポリメチルメタクリレート、ポリアクリ ロニトリル、ポリアクリルアミド又はポリアクロレインであり;蛍光染料の組合 せが式: ▲数式、化学式、表等があります▼ [式中、R1〜R6は同一又は異なる基であり、単独の又は組合せた、水素、ハ ロゲン、アルキル、アルコキシ、アルケニル、シクロアルキル、アリールアルキ ルアシル(これらのアルキル部分はそれぞれ、炭素数約20未満である)、アリ ールもしくはヘテロアリールであり;R7は窒素、メチン、又はハロゲン−、ア ルキル−、アルコキシ−、アルケニル−、シクロアルキル−、アリールアルキル −、アシル−(これらのアルキル部分はそれぞれ、炭素数約20未満である)、 アリール−もしくはヘテロアリール−置換メチンである]で示される、種々なポ リアザインダセン染料である、約6種未満の染料を含み;総染料濃度が約0.8 〜1.2重量%であり;初期供与体染料と最終受容体染料との比が約4:1と約 6:1との間である請求項1記載の微小粒子。
- 10.請求項9記載の蛍光微小粒子の製造方法。
- 11.好ましい励起ピークを有する初期供与体染料と、好ましい発光ピークを有 する最終受容体染料とを含み、組合せ中の各染料が励起エネルギーの有意なエネ ルギ−伝達を可能にするために充分なスペクトル重複を有する蛍光染料組合せを 含み;総染料濃度が約10重量%未満になり、初期供与体対最終受容体の比が約 1:5と約10:1との間になるように、前記染料組合せがポリマ−微小粒子中 に配合される蛍光微小粒子。
- 12.微小粒子がニトロセルロース、ポリスチレン、臭素化ポリスチレン、ポリ アクリル酸、ポリアクリロニトリル、ポリアクリルアミド、ポリアクロレイン、 ポリジメチルシロキサン、ポリブタジエン、ポリイソプレン、ポリウレタンポリ 酢酸ビニル、ポリ塩化ビニル、ポリビニルビリジン、ポリビニルベンジルクロリ ド、ポリビニルトルエン、ポリビニリデンクロリド、又はポリジビニルベンゼン であり、直径約15μm未満である請求項11記載の微小粒子。
- 13.蛍光染料の組合せが式: ▲数式、化学式、表等があります▼ [式中、R1〜R6は同一又は異なる基であり、単独の又は組合せた、水素、ハ ロゲン、アルキル、アルコキシ、アルケニル、シクロアルキル、アリールアルキ ルアシル(これらのアルキル部分はそれぞれ、炭素数約20未満である)、アリ ールもしくはヘテロアリールであり;R7は窒素、メチン、又はハロゲン−、ア ルキル−、アルコキシ−、アルケニル−、シクロアルキル−、アリールアルキル −、アシル−(これらのアルキル部分はそれぞれ、炭素数約20未満である)、 アリール−もしくはヘテロアリール−置換メチンである]で示される、約6種未 満のポリアザインダセン染料を含む請求項12記載の微小粒子。
- 14.共有結合した又は不動吸着した(passively adsorbed )生体反応性物質をさらに含む請求項13記載の微小粒子。
- 15.前記生体反応性物質が生体分子である、すなわちビオチン、アビジンスト レプトアビジン、ジゴキシゲニン、又は核酸化合物である請求項14記載の微小 粒子。
- 16.核酸検出方法において、 (a)標的核酸を含むと考えられるサンプルを調製する工程と;(b)好ましい 励起ピークを有する初期供与体染料と、好ましい発光ピークを有する最終受容体 染料とを含み、組合せ中の各染料が励起エネルギーの有意なエネルギー伝達を可 能にするために充分なスペクトル重複を有する染料組合せを配合し、かつ標的核 酸を特異的に認識する核酸配列に結合しているポリマー微小粒子を各プローブが 含む、微小粒子標識プローブの懸濁液を調製する工程と;(c)微小粒子標識プ ローブの懸濁液をサンプルと一緒にする工程と;(d)結合しないプローブを除 去する工程と;(e)微小粒子の初期供与体染料の励起ピークの範囲内で発光す る励起源でプローブを照射する工程と; (f)照射されたプローブから生ずる蛍光を検出する工程とを含む方法。
