Claims (1)
- 【特許請求の範囲】 1.患者におけるアレルギー性疾患の診断法であって、 (a)放出混合物中への薬理学的伝達物質放出を遮断放出混合物中への薬理学 的伝達物質放出と比較すること、ただしここに、放出混合物および遮断放出混合 物はおのおのナイーブ提供者からの単一組織標本の一部を含み、およびここに、 放出混合物および遮断放出混合物はおのおのさらに、血液標本中の目的アレルゲ ンに特異的なIgEの存在または不在が未知な、患者からの単一血液標本の一部 を含み、およびここに、遮断放出混合物はさらにIgE拮抗剤を含み、およびこ こに、放出混合物および遮断放出混合物双方をアレルゲンと混合するものとする 、 (b)段階(a)での比較に基づいて、その血清標本中にそのアレルゲンに特 異的なIgEの存在または不在を決定すること を含む診断法。 2.薬理学的伝達物質がヒスタミンである請求項1の方法。 3.組織標本が血液標本である請求項1の方法。 4.IgE拮抗剤が抗IgE抗体またはその断片である請求項1の方法。 5.抗IgE抗体がモノクローナル抗IgE抗体である請求項4の方法。 6.モノクローナル抗IgE抗体がrhuMAbE25である請求項5の方法。 7.IgE拮抗剤がIgE変異体またはその断片である請求項1の方法。 8.IgE拮抗剤が可溶性FcεRI受容体である請求項1の方法。 9.IgE拮抗剤がIgE結合性ペプチドまたはFcεRI受容体結合性ペプチ ドである請求項1の方法。 10.遮断放出混合物中の組織標本を最初にIgE拮抗剤と混合し、次に患者か らの血清標本と混合する請求項1の方法。 11.遮断放出混合物中の血清標本を最初にIgE拮抗剤と混合し、次に組織標 本と混合する請求項1の方法。 12.好塩基球のIgE誘導感化を遮断する薬剤の生物活性検定法であって、 (a)放出混合物中への薬理学的伝達物質放出を遮断放出混合物中への薬理学 的伝達物質放出と比較すること、ただしここに、放出混合物および遮断放出混合 物はおのおのナイーブ提供者から得た単一全血標本の一部を含み、およびここに 、放出混合物および遮断放出混合物はおのおのさらにアレルゲンに特異的なIg Eを含む単一血清標本の一部を含み、およびここに、遮断放出混合物はさらに薬 剤を含み、およびここに、放出混合物および遮断放出混合物双方をアレルゲンと 混合するものとする、 (b)段階(a)での比較に基づいて、好塩基球のIgE誘導感化を遮断する 薬剤の生物活性を決定すること を含む薬剤の生物活性検定法。 13.薬剤が抗アレルギー性治療剤候補である請求項12の方法。 14.抗アレルギー性治療剤候補が抗IgE抗体またはその断片、IgE変異体 またはその断片、可溶性FcεRI受容体、IgE結合性ペプチドおよびFcε RI受容体結合性ペプチドから構成される群から選択される請求項13の方法。 15.薬剤がIgE拮抗剤候補である請求項12の方法。 16.IgE拮抗剤候補が抗IgE抗体またはその断片、IgE変異体またはそ の断片、可溶性FcεRI受容体、IgE結合性ペプチドおよびFcεRI受容 体結合性ペプチドから構成される群から選択される請求項15の方法。 17.遮断放出混合物中の全血標本を最初に薬剤と混合し、次に血清標本と混合 する請求項12の方法。 18.遮断放出混合物中の血清標本を最初に薬剤と混合し、次に全血標本と混合 する請求項12の方法。 19.患者におけるアレルギー性疾患の診断法であって、 (a)反応混合物中への薬理学的伝達物質放出を遮断反応混合物中への薬理学 的伝達物質放出と比較すること、ただしここに、 (1)反応混合物および遮断反応混合物は患者血清IgEおよびアレルゲンに よる誘導に際して宿主細胞の薬理学的伝達物質放出を媒介することのできるFcε RIαサブユニットの表面発現を示すように遺伝子工学的に処理した肥満細胞 宿主のいずれか、および患者血清IgEおよびアレルゲンによる誘導に際して宿 主細胞の薬理学的伝達物質放出を媒介することのできるFcεRIαサブユニッ トの表面発現を示すように遺伝子工学的に処理した好塩基球細胞宿主のいずれか から構成される群から選択した共通の親細胞の子孫である遺伝子工学的に処理し た細胞を含み、 (2)反応混合物および遮断反応混合物はおのおのさらに血清標本中の目的ア レルゲンに特異的なIgEの存在または不在が未知な、患者からの単一血清標本 の一部を含み、 (3)遮断反応混合物はさらにIgE拮抗剤を含み、および (4)放出混合物および遮断放出混合物双方をアレルゲンとを混合するものと する、 (b)段階(a)での比較に基づいて、その血清標本中にそのアレルゲンに特 異的なIgEの存在または不在を決定すること を含む診断法。 20.遺伝子工学的に処理した細胞が遺伝子工学的に処理した肥満細胞である請 求項19の方法。 21.遺伝子工学的に処理した肥満細胞が遺伝子工学的に処理した齧歯類肥満細 胞である請求項20の方法。 22.遺伝子工学的に処理した齧歯類肥満細胞が遺伝子工学的に処理したラット 肥満細胞である請求項21の方法。 23.遺伝子工学的に処理したラット肥満細胞がRBL48細胞である請求項2 2の方法。 24.FcεRIαサブユニットが患者の動物種から誘導される請求項19の方 法。 25.患者がヒトである請求項19の方法。 26.FcεRIαサブユニットをヒト起源から誘導する請求項19の方法。 27.FcεRIαサブユニットが遺伝子工学的に処理した細胞に対して外因性 である請求項19の方法。 28.IgE拮抗剤が抗IgE抗体またはその断片である請求項19の方法。 29.抗IgE抗体がモノクローナル抗IgE抗体である請求項28の方法。 30.抗IgE抗体がrhuMAbE25である請求項29の方法。 31.IgE拮抗剤がIgE変異体またはその断片である請求項19の方法。 32.IgE拮抗剤が可溶性FcεRI受容体である請求項19の方法。 33.IgE拮抗剤がIgE結合性ペプチドまたはFcεRI受容体結合性ペプ チドである請求項19の方法。 34.遮断反応混合物中の遺伝子工学的に処理した細胞を最初にIgE拮抗剤と 混合し、次に患者からの血清標本と混合する請求項19の方法。 35.遮断反応混合物中の血清標本を最初にIgE拮抗剤と混合し、次に遺伝子 工学的に処理した細胞と混合する請求項19の方法。 36.薬理学的伝達物質がヒスタミンである請求項19の方法。
