Claims (1)
- 【特許請求の範囲】 1.式I: [式中、 Bはヌクレオ塩基であり、 XおよびYは、独立して、酸素または硫黄であり、 Yは所望により保護されていてもよい水素またはヒドロキシであり、 R1は、所望により官能基で置換されていてもよいヒドロカルビルであり、 R2は、水素または所望により官能基で置換されていてもよいヒドロカルビル であり、 Aは、化合物Iのアセタール安定性を改善し得る電子吸引性または電子供与性 基であり、 L1およびL2は、同一または異なっていてもよい炭化水素リンカーであり、 (i)基Aを介してL1に結合しているか、または(ii)L1に直接結合しているい ずれかのL2が存在する場合には、基AはリンカーL1およびL2のうちの1つに 結合しており、 Fは色素標識であり、 QはFについてのカップリング基であり、および l、mおよびnは、独立して、0または1であるが、但し、mが1である場合 にはlは1であって、nが1である場合にはlが1であってmは1である] で示される化合物またはその塩。 2.R1およびR2が、独立して、所望により1または2以上の官能基で置換さ れていてもよい第一級、第二級および第三級アルキルから選択される請求項1記 載の化合物。 3.R1およびR2がメチルおよびエチルから選択される請求項2記載の化合物 。 4.Aが鎖L1-A-L2の一部分であって、かつアミド、スルホキシ、スルホン 、カルボアルコキシ、エーテル、チオエーテルおよびアミノから選択される請求 項1-3いずれか1項記載の化合物。 5.Aが鎖L1-L2への置換基であって、シアノ、ニトロおよびハロゲンから 選択される請求項1-3いずれか1項記載の化合物。 6.L1が好ましくは10個までの炭素原子を有する直鎖脂肪族炭化水素鎖で ある請求項1-5いずれか1項記載の化合物。 7.L2が好ましくは10個までの炭素原子を有する直鎖脂肪族炭化水素鎖で ある請求項1-6いずれか1項記載の化合物。 8.Yがアルコキシである請求項1-7いずれか1項記載の化合物。 9.特にオリゴ-またはポリヌクレオチド調製用の、核酸合成反応における請 求項1-8いずれか1項記載の化合物の使用。 10.微小配列決定における請求項1-8いずれか1項記載の化合物の使用。 11.決定すべき核酸を含む一本鎖鋳型を供し、相補的核酸分子中に取り込ま れた各ヌクレオチドの同一性をその取り込みにつづいて決定するヌクレオチドの 付加による段階的一連様式で相補鎖核酸分子を少なくとも部分的に合成すること よりなり、ここに、該ヌクレオチドが請求項1-8いずれか1項定義の式Iで示 される化合物であって、3'-ブロッキング基がその取り込み後にヌクレオチドか ら除去されて核酸分子のさらなる伸長が許容されることを特徴とする核酸配列決 定方法。 12.3'-ブロッキング基が酸加水分解によって除去される請求項11記載の 方法。 13.式I中のFが蛍光標識である請求項11または12いずれか1項記載の 方法。 14.該一本鎖鋳型が固体支持体に結合している請求項11-13いずれか1 項記載の方法。 15.式II: [式中、BおよびXは請求項1の定義に同じである] で示される化合物またはその塩と、式III: [式中、R1、R2およびZは請求項1の定義に同じで、pは1ないし10、好ま しくは1ないし6の整数であって、R3はヒドロカルビルである] で示される化合物とを反応させて、式IV: [式中、B、X、Z、R1、R2、R3およびpは前記定義に同じである] で示される化合物またはその塩を得、所望により後者とジアミンH2N-(CH2) q-NH2[式中、qは1ないし10、好ましくは1ないし6の整数である]とを 反応させて式V: [式中、B、X、Z、R1、R2、pおよびqは前記定義に同じである] で示される化合物を得、ついで、所望により色素標識Fを末端アミノ基にカップ リングさせてもよいことを特徴とする式Ia: [式中、B、X、Z、R1、R2、Q、F、mおよびnは請求項1記定義に同じで あって、pおよびqは前記定義に同じである] で示される化合物またはその塩の製法。
