Description
Claims (16)
- 퓨린계 물질 생산능을 갖고, 프럭토스-비스포스파타제의 효소 활성이 저하되도록 변형된 바실러스(Bacillus)속 세균.
- 제1항에 있어서, 퓨린계 물질이 이노신, 크산토신, 구아노신 및 아데노신으로 이루어지는 그룹으로부터 선택되는 퓨린 뉴클레오사이드인 바실러스속 세균.
- 제1항에 있어서, 퓨린계 물질이 이노신산, 크산틸산, 구아닐산 및 아데닐산으로 이루어지는 그룹으로부터 선택되는 퓨린 뉴클레오타이드인 바실러스속 세균.
- 제1항 내지 제3항 중의 어느 한 항에 있어서, 프럭토스-비스포스파타제를 암호화하는 유전자를 파괴함, 또는 프럭토스-비스포스파타제를 암호화하는 유전자의 발현량을 저하시킴에 의해, 프럭토스-비스포스파타제 활성이 저하된 바실러스속 세균.
- 제4항에 있어서, 프럭토스-비스포스파타제를 암호화하는 유전자가 하기 (A) 또는 (B)의 단백질을 암호화하는 유전자인 바실러스속 세균.
(A) 서열번호 1의 아미노산 서열을 포함하는 단백질,
(B) 1개 또는 수개의 아미노산 잔기의 치환, 결실, 삽입, 부가 또는 역위를 포함하는 서열번호 1의 아미노산 서열을 포함하고, 프럭토스-비스포스파타제 활성을 갖는 단백질. - 제1항 내지 제5항 중의 어느 한 항에 있어서, 포스포리보실 피로포스페이트 신테타제 활성이 상승하도록 추가로 변형된 바실러스속 세균.
- 제1항 내지 제6항 중의 어느 한 항에 있어서, 퓨린 오페론의 발현량이 상승하도록 추가로 변형된 바실러스속 세균.
- 제7항에 있어서, 퓨린 오페론의 리프레서(repressor)를 암호화하는 유전자인 purR 유전자를 파괴함에 의해, 퓨린 오페론의 발현량이 상승된 바실러스속 세균.
- 제1항 내지 제8항 중의 어느 한 항에 있어서, 퓨린 뉴클레오사이드 포스포릴라제 활성이 저하되도록 추가로 변형된 바실러스속 세균.
- 제1항 내지 제9항 중의 어느 한 항에 있어서, IMP 데하이드로게나제 활성이 저하되도록 추가로 변형된 바실러스속 세균.
- 제1항 내지 제10항 중의 어느 한 항에 있어서, 바실러스속 세균이 바실러스 서브틸리스(Bacillus subtilis)인 바실러스속 세균.
- 제1항 내지 제11항 중의 어느 한 항에 따르는 바실러스속 세균을 배지에 배양하여, 상기 세균의 세포내 또는 배지중에 퓨린계 물질을 축적시키고,
상기 세포 또는 배지로부터 퓨린계 물질을 회수함
을 포함하는, 퓨린계 물질의 제조법. - 제12항에 있어서, 퓨린계 물질이 퓨린 뉴클레오사이드 또는 퓨린 뉴클레오타이드인, 퓨린계 물질의 제조법.
- 제13항에 있어서, 퓨린계 물질이 이노신, 크산토신, 구아노신 및 아데노신으로 이루어지는 그룹으로부터 선택되는 퓨린 뉴클레오사이드인, 퓨린계 물질의 제조법.
- 제13항에 있어서, 퓨린계 물질이 이노신산, 크산틸산, 구아닐산 및 아데닐산으로 이루어지는 그룹으로부터 선택되는 퓨린 뉴클레오타이드인, 퓨린계 물질의 제조법.
- 제14항에 따르는 제조법에 의해 퓨린 뉴클레오사이드를 제조하고,
상기 퓨린 뉴클레오사이드에, 폴리인산, 페닐 포스페이트 및 카르바밀 포스페이트로 이루어지는 그룹으로부터 선택된 포스페이트 공여체와, 뉴클레오사이드-5'-인산 에스테르를 생성하는 능력을 갖는 미생물 또는 산성 포스파타제를 작용시켜 퓨린 뉴클레오타이드를 생성시키고,
상기 퓨린 뉴클레오타이드를 회수함
을 포함하는, 퓨린 뉴클레오타이드의 제조법.
