Abstract
Description
도 2a 내지 도 2c는, 인슐린에 대해 면역 염색된 세포의 영상을 나타낸다. 실시예 2에 기재된 바와 같이, 도 2a, 대조군; 도 2B, 50 ng/mL 에프린-A3으로 처리된 세포; 및 도 2c, 100 ng/mL 에프린-A3으로 처리된 세포.
도 3a 내지 도 3c는, 인슐린에 대해 면역 염색된 세포의 영상을 나타낸다. 실시예 2에 기재된 바와 같이, 도 3a, 대조군; 도 3b, 50 ng/mL 에프린-A4로 처리된 세포; 및 도 3c, 100 ng/mL 에프린-A4로 처리된 세포.
도 4a 내지 도 4d는, 스핑고신-1-포스페이트(S1P)로 처리하고 제1일(도 4a), 제7일(도 4b), 및 2가지의 상이한 배율로 제10일(도 4c 및 도 4d)에 영상화한 세포의 S6 배양물의 위상차 영상을 도시한다. 상기 영상들은, 제7일에 내분비 세포의 클러스터링의 명확한 증거가 있었고, 제10일에 췌장 내배엽 상피의 박층에 의해 클러스터들이 서로 분리되었음을 나타낸다.

Claims (13)
- 액티빈 A 또는 액티빈 C를 포함하는 배지에서 췌장 내배엽 세포를 배양함으로써 소마토스타틴의 발현이 향상된 췌장 내분비 세포를 생성하는 단계를 포함하는, 췌장 내분비 세포에서 소마토스타틴의 발현을 향상시키는 방법.
- 제1항에 있어서, 췌장 내분비 세포에서 인슐린, 글루카곤 및 그렐린(ghrelin)의 발현은 억제되는 방법.
- 제1항에 있어서, 췌장 내분비 세포가 액티빈 A 또는 액티빈 C로 처리되지 않은 췌장 내배엽 세포로부터 생성된 췌장 내분비 세포와 비교하여 더 많은 소마토스타틴을 발현하는 방법.
- 제1항에 있어서, 액티빈 A 또는 액티빈 C로 처리되지 않은 췌장 내배엽 세포로부터 생성된 췌장 내분비 세포에서의 인슐린의 발현과 비교하여 액티빈 A 또는 액티빈 C로 배양된 췌장 내배엽 세포로부터 생성된 췌장 내분비 세포에서 인슐린의 발현이 억제되는 방법.
- 제1항에 있어서, 액티빈 A 또는 액티빈 C로 처리되지 않은 췌장 내배엽 세포로부터 생성된 췌장 내분비 세포에서의 글루카곤의 발현과 비교하여 액티빈 A 또는 액티빈 C로 배양된 췌장 내배엽 세포로부터 생성된 췌장 내분비 세포에서 글루카곤의 발현이 억제되는 방법.
- 제1항에 있어서, 액티빈 A 또는 액티빈 C로 처리되지 않은 췌장 내배엽 세포로부터 생성된 췌장 내분비 세포에서의 그렐린의 발현과 비교하여 액티빈 A 또는 액티빈 C로 배양된 췌장 내배엽 세포로부터 생성된 췌장 내분비 세포에서 그렐린의 발현이 억제되는 방법.
- 제1항에 있어서, 췌장 내배엽 세포가 실질적으로 CDX2 또는 SOX2를 발현하지 않는 방법.
- 제7항에 있어서, 액티빈 A 또는 액티빈 C로 배양되는 췌장 내배엽 세포가 만능 세포의 단계적 분화에 의해 수득된 것인 방법.
- 제8항에 있어서, 만능 세포가 인간 배아 만능 세포로부터 유래된 것인 방법.
- 제1항에 있어서, 췌장 내배엽 세포가 인간 췌장 내배엽 세포인 방법.
- 제1항에 있어서, 액티빈 A를 포함하는 배지에서 췌장 내배엽 세포를 배양하는 단계를 포함하는 방법.
- 제1항에 있어서, 액티빈 C를 포함하는 배지에서 췌장 내배엽 세포를 배양하는 단계를 포함하는 방법.
- 제1항에 있어서,
만능 줄기 세포를 완성 내배엽 세포로 분화시키는 단계;
완성 내배엽 세포를 원시 장관(guttube) 세포로 분화시키는 단계;
원시 장관 세포를 전장(foregut) 세포로 분화시키는 단계;
전장 세포를 췌장 전장 전구체 세포로 분화시키는 단계; 및
췌장 전장 전구체 세포를 췌장 내배엽 세포로 분화시키는 단계를 추가로 포함하는 방법.
