Abstract
Description
도 2a 및 도 2b: 표시된 돌연변이를 갖고 Fc-이종이합체의 한 측의 C 말단에 융합된 IL-2 뮤테인을 T 세포에서 STAT5 인산화를 자극하는 이의 능력에 대해 시험하였다. 이 뮤테인은 또한 CD25에 대해 고친화도를 부여하는 3개의 돌연변이(V69A, N71R, Q74P)를 함유하였다. 이의 활성은 Fc 융합이 없는 3개의 형태의 IL-2(빈 기호)와 비교되었다: WT IL-2, HaWT(CD25에 대한 고친화도)(N29S, Y31H, K35R, T37A, K48E, V69A, N71R, Q74P) 및 HaD(CD25에 대한 고친화도 및 감소된 신호전달 활성)(N29S, Y31H, K35R, T37A, K48E, V69A, N71R, Q74P, N88D). 게이팅된 FOXP3+CD4+ 및 FOXP3-CD4+ T 세포에 대한 포스포-STAT5 반응이 도시되어 있다.
