Opis wynalazku
Przedmiotem wynalazku jest sposób otrzymywania chinolino-5,8-dionów oraz chinolino-5,8-diony, które znajdują zastosowanie jako produkty pośrednie, zwłaszcza w przemyśle farmaceutycznym.
Bracher (Heterocycles, 29, 2093, 1989) podawał już sposób przygotowania chinolino-5,8-dionów wychodząc z 5-amino-8-hydroksychinoliny przez utlenianie dichromianem. Ten sposób wymaga podwójnego podstawienia rdzenia chinolinowego, co pociąga za sobą konieczność zapewnienia trudno dostępnych i kosztownych surowców. S. Ghosh (J. Fluorine Chem., 1994; 67: 53-56) opisuje pochodne chinolino-5,8-dionów otrzymane przez cykloaddycję między p-benzochinonem i podstawionym dienem. 4-chinolino-5,8-dion opisany jest jako związek pośredni syntezy bardziej złożonych związków tri- lub tetracyklicznych (M. Croisy-Delsey i wsp., J. Heterocyclic Chem. 1993; 30: 55-60), podczas gdy 2-metoksy-chinolino-5,8-dion prezentowany jest jako produkt wtórny N-alkilacji 2,5,8(1H)-chinolinotrionu (C. Avedano i wsp., Synthesis 1991: 727-730). 4-hydroksy-5,8-chinolino-chinon może być syntetyzowany wychodząc z 2,5-dimetoksyaniliny i estru kwasu metyloakrylowego (P. Withopf i wsp. Tetrahedron, 1987; 43(20): 4549-4554). Chinolino-5,8-dion i 2-chloro-4-metylo-chinolino-5,8-dion są produktami wyjściowymi służącymi do syntezy pochodnych azaantrachinonu (K. T. Potts i wsp., J. Org. Chem., 1986: 2011-2021). Czynniki przeciwmalaryczne pochodzące z 6-amino-5,8-dimetoksychinolin przygotowane są wychodząc z 2-trifluoro-4-metylu lub 2-4 dimetylochinolino-5,8-dionu (C. Tempie i wsp., J. Med. Chem., 1974; 17(6): 615-619). W końcu, EP 0 433 679 opisuje pochodne chinolino-5,8-dionów jako inhibitory reakcji Maillard'a w organizmie, jest ona odpowiedzialna (przez denaturację białek przez cukry) za kilka konsekwencji cukrzycy (neuropatie, retinopatie, etc).
Istnieją ponadto sposoby przygotowania chinonów przez fotoutlenianie fenoli (na przykład, Chem. Comm., 2173, 1996), ale nigdy nie rozważano zastosowania takich sposobów do związków heterocyklicznych, a w szczególności do pochodnych chinoliny.
W efekcie, nie było oczywiste, że takim metodom nie będzie przeszkadzała obecność jądra pirydynowego, przyciągającego elektrony. Można się też było obawiać reakcji między chinonami i azotem jądra pirydynowego prowadzących do barwnych polimerów. To właśnie zaobserwowano z 5-hydroksyizochinoliną.
Obecnie stwierdzono, że szczególna grupa 8-hydroksychinolin daje przez fotoutlenianie w obecności sensybilizatora i rozkład pośredniego hydroksynadtlenku, chinolino-5,8-diony ze znaczną wydajnością.

PL 203 362 B1 w którym R1, R2 i R3 mają znaczenie podane powyż ej, utleniana jest przy pomocy tlenu pod wpływem promieniowania aktynicznego w roztworze rozpuszczalnika organicznego i w obecności katalitycznej ilości sensybilizatora, a wytworzony hydronadtlenek o wzorze:
rozkłada się do związku o wzorze I.
Reakcja utleniania przez tlen przeprowadzana jest w rozpuszczalniku organicznym, w którym rozpuszczalna jest wyjściowa 8-hydroksychinolina o wzorze II. Rozpuszczalnik jest korzystnie wybrany tak, aby był w nim także rozpuszczalny sensybilizator.
Sensybilizatorem może być w szczególności tetrafenyloporfina (lub TPP) w którym to przypadku rozpuszczalnik może być zwłaszcza dichlorometanem.
Można stosować też inne sensybilizatory i odpowiednie rozpuszczalniki, a w szczególności:
- czerwień bengalską w CH3CN, MeOH, EtOH, CHCI3 lub wodzie,
- błękit metylenowy w CH3CN, MeOH, EtOH, CHCI3 lub wodzie,
- tetra(4-pirydylo)-porfinę w CH2CI2 lub CHCl3,
- dicyjanonaftalen (DCN), dicyjanoantracen (DCA), dicyjanobenzen (DCB),
- bis-tetrafluoroboran N,N'-dimetylo-2,7-diazopirenu (DAP2+).
Reakcję przeprowadza się korzystnie w temperaturze zbliżonej do temperatury otoczenia (od 15 do 25°C) pod napromienieniem światłem widzialnym pod barbotowaniem tlenu, w czasie, który może trwać od 2 do 8 godzin.
Hydronadtlenek wytworzony z wzoru III rozkłada się spontanicznie przy mieszaniu, w szczególności w obecności Na2SO4.
Celem niniejszego wynalazku są także chinolino-5,8-diony o wzorze:
w którym R1 oznacza wodór,
R2 oznacza wodór,
R3 jest wybrany spośród wodoru, grupy alkilowej C1-C6, -CHO, -CN, -CO2R, -CONRR'-,
-CH2N-COOR, -CH2-N-COOR
CH2-COOR' CH2-AR
R i R' są wybrane spośród grup alkilowych C1-C6, a Ar jest grupą arylową C6-C14, z wyjątkiem związków, w których:
R1 = H, R2 = H i R3 jest wybrany spośród H, CH3, CN i CHO.
Korzystnie, R3 jest wybrany spośród grup -CONRR', oraz -CH2N-COOR, -CH2-N-COOR

Ogólny sposób postępowania
W fiolce z pyreksu o płaskich ścianach napromieniowywano w 20°C (chłodzenie wodą) roztwór związku o wzorze II (2 mmole) w dichlorometanie (lub innym rozpuszczalniku jeśli jest nierozpuszczalny) (20 ml), zawierającym katalityczną ilość tetrafenyloporfiny (TTP, 6 mg, lub 0,5% w molach) światłem widzialnym (łuk ksenonowy 1500 W, filtr dla promieniowania > 495 nm) pod barbotowaniem tlenu podczas 2-8 godzin (reakcję śledzono za pomocą CCM). Roztwór napromieniowany następnie wlano na Na2SO4 suchy (3 g), mieszano w temperaturze pokojowej przez 12 godzin i sączono. Rozpuszczalnik odparowano, a otrzymany surowy chinon oczyszczono za pomocą chromatografii na kolumnie z krzemionki.

w którym R1, R2 i R3 mają znaczenie podane powyż ej, utleniana jest przy pomocy tlenu pod działaniem promieniowania aktynicznego w roztworze rozpuszczalnika organicznego i w obecności katalitycznej ilości sensybilizatora, a wytworzony hydronadtlenek o wzorze:
