Description
Verfahren zur Herstellung von Polyenaldehyden
Gegenstand der Erfindung ist ein Ver i'ahren zur Herstellung von 4- [2', 6', 6'-Tri methyl-cyclohexyliden]-2-methyl-buten- (2)-al (1) und von 4- [2', 6', 6'-Trimethyl-cyclohexen (2')-yliden]-2-methyl-buten- (2)-al.- (l), die im j'olgenden Iso-C14-aldehyd und Retrodehydro- C-aldehyd genannt werden.
Das Verfahren ist dadurch gekennzeichnet, dass man den Aldehyd der Formel R=CII-CHO, worin R den 2, 6, 6 Trimethyl-eyclohexyliden-oder den 2, 6, 6-Tri methvl-eyclohexen- (2)-yliden-Rest bedeutet, aeetalisiert, an das erhaltene Acetal in Gegen- wart eines sauren Mittels einen Propenyläther anlagert und das erhaltene Anlagerungsprodukt mit sauer reagierenden Stoffen behandelt.
Der ais Ausgangsmaterial benotigte 2, 6, 6 Trimethyl-eyelohexyliden-bzw. 2, 6, 6-Tri methyl-eyelohexen- (2)-yliden-acetaldehyd (Isobzw. Retrodehydro-C11-aldehyd) kann wie -folgt gewonnen werden :
Das durch Kondensation von Athoxyace- tylen mit 2,2,6-Trimethyl-cyclohexanon-(1) bzw. zw'', 6, 6-Trimethyl-cyclohexen- (2)-on- (1) er haltene sithoxy-acetylencarbinol wird in an n sich bekannter Weise an der Dreifachbindung partiell hydriert und mit SÏure behandelt. V.-Absorptionsmaximum bei 237-238 m, cc (in Petrolätherlosung). Der aus Trimethyl cyclohexenon gewonnene 2, 6, 6-Trimethyl-cyclo hexen-(2)-yliden-acetaldehyd ist ein gelbliches öl vom Kp13: 115¯ C ; n25 = 1, 531 ; U. V.-Absorptionsmaximum bei 278 ma (in Petroläther losung).
Die erste Stufe des erfindungsgemϯen Verfahrens besteht in der Acetalisierung des Iso-bzw. Retrodehydro-C11-aldehyds zum 2, 6, 6 Trimethyl-cyclohexyliden- bzw. 2, 6, 6-Trime thyl-cyclohexen- (2)-yliden-acetaldehydaeetal (Iso-bzw. Retrodehydro-C11-acetal). Man acetalisiert beispielsweise mittels eines Orthoesters in Gegenwart eines sauren Kondensationsmittels, wie BortrifluoridÏtherat, Zinkchlorid, Ammoniumnitrat, PhosphorsÏure, p Tolnolsulfosäure usw. Hierzn eignen sich besonders die Orthoester niederer aliphatiseher Säuren mit niederen aliphatischen Alkoholen, vorzugsweise der Orthoameisensäuremethyl-, -äthyl-oder-n-butylester. Die erhaltenen Acetale sind farblose Öle.
Die Iso-Cl1-acetale weisen im Ultraviolettspektrum ber 225 mu kein Absorptionsmaximum auf, wogegen f r die Retrodehydro-C11-acetale ein solches bei 240 mu u charakteristiseh ist. F r die weitere Verarbeitung ist eine besondere Reinigung der Acetale, z. B. durch Destillation, nicht erfor derlich.
In der zweiten Stufe des Verfahrens wird an das Iso-bzw. Retrodehydro-Cll-acetal ein Propenyläther in Gegenwart eines sauren Mittels angelagert, wobei 4- [2', 6', 6'-Trimethvl- cyclohexyliden]-bzw. 4- [2', 6', 6'-Trimethyl- cyclohexen- (2')-yliden]-2-methyl-butan-äther- (3)-acetal-(1) (Iso- bzw. Retrodehydro-C14 ätheracetal) entsteht. Als saures Mittel eignen sich BortrifluoridÏtherat, Zinkchlorid, Titantetrachlorid, Aluminiumtrichlorid, Ferrichlorid, Zinntetrachlorid usw. Man verwendet zweckmässigerweise den Propenylather des gleichen Alkohols, mit dem der Iso-bzw.
Re trodehydro-Cll-aldehyd acetalisiert wurde, z. B. Methylpropenyläther, Athylpropenyl- äther oder n-Butylpropenyläther. Die Reak- tion erfolgt am besten bei einer möglichst tiefen Temperatur, wodurch man unerwünschte Nebenreaktionen, wie Polymerisation und Won- densation des gebildeten Iso-bzw. Retrodehydro-C14-Ïtheracetals mit PropenylÏther, vermeiden kann. Die optimale Reaktionstem- peratur liegt je nach der Wahl des sauren Mittels, des Aeetals und des Propenyläthers zwischen 15 und 50 C.
Bei der bevorzugten Ausführungsform lässt man etwa molare Mengen Iso-bzw. Retrodehydro-Cl i-aeetal und Propenyläther bei 20 bis 40 C in Gegenwart von Zinkehlorid aufeinander einwirken. Man erhält so weitgehend reine Iso-bzw. Retrode hydro-Cl4-ätheraeetale inbeinahe quantitativer Ausbeute. Es sind farblose Ole. Im Ultraviolettspektrum weisen die Iso-Cl4-ätheraee- tale über 225 m, u kein Absorptionsmaximum auf, wogegen für die Retrodehydro-Cn-äther- acetale ein solches bei 235 mÁ.charakteristisch ist. Für die weitere Verarbeitmg ist eine besondere Reinigung, z. B. durch Destillation, nicht erforderlieh.
Als dritte Stufe des erfindungsgemässen Verfahrens werden die Iso-bzw. Retrodehydro Cl4-ätheracetale mit sauer reagierenden Stoffen behandelt. Diese Behandlung wird mit Vorteil so durchgeführt, z. B. durch Erwär- men, dass die Abspaltung des Alkohols aus der Stellung 2, 3 und die Freilegung der Aldehyd- gruppe gleichzeitig erfolgt. Als sauer reagie- rende Stoffe werden vorteilhaft wasserlösliche, organische oder anorganische Säuren, wie p Toluolsulfosäure, Essigsäure, Propionsäure, Oxalsäure, Schwefelsäure, Phosphorsäure oder sauer reagierende, wasserlösliche Salze, wie Zinkehlorid und Natriumbisulfat verwendet.
Bei der Reaktion wird mit Vorteil Sauerstoff ausgeschlossen und ein Antioxydans, z. B.
IIydroehinon, zugefügt. Man arbeitet zweekmässig unter Bedingungen, bei welchen der entstehende Alkohol fortlaufend aus der Reak tionsmischung entfernt wird. Man kann dem Reaktionsgemiseh ein mit Wasser mischbares Losungsmittel, wie beispielsweise Dioxan, Tetrahydrofuran, Athylenglykoldimethyläther usw. zugeben, um ein homogenes Reaktions- gemisch zu erhalten. Vorzugsweise wird das Iso-bzw. Retrodehydro-Cl4-ätheraeetal mit verdünnter Phosphorsäure in Gegenwart eines mit Wasser mischbaren L¯sungsmittels oder mit Essigsäure unter Zusatz eines Alkaliaeetates und etwas Wasser auf etwa 100 C erwärmt. Beim Verdünnen des Reaktions- gemisches mit Wasser fällt der ölige Iso-bzw.
Retrodehydro-Cl4-aldehyd aus, welcher durch Destillation gereinigt werden kann.
Zur Vermeidung von Substanzverlusten durch Polymerisation und Zersetzung empfiehlt es sich, während des gesamten Verfahrens Temperaturen ber 120¯ C zu vermeiden und insbesondere Zwischenprodukte, nämlieh Iso-bzw. Retrodehydro-C11-aceta, und Isobzw. Retrodehydro-C14-Ïtheracetal, undestilliert weiter zu verarbeiten.
Der nach vorliegendem Verfahren erhaltene Iso-CI4-aldehyd siedet bei 82¯ C/0, 05 mm.
Er weist im U. V.-Spektrum ein Absorptionsmaximum bei 287 bis 288 mÁ auf, E11: 1400 (in Petrolätherlösung).
Der Retrodehydro-C14-aldehyd siedet bei 94 C/0, 05 mm. Er weist im U. V.-Spektrum ein Absorptionsmaximum bei 318 m, u auf, El : 1670 (in Petrolätherlösung).
Der Iso-Cl4-aldehyd und der Retrodehydro C14-aldehyd sind wertvolle Zwisehenprodukte f r die Synthese von Vitamin A bzw. von Vitamin A2 und von ss-Carotin bzw. von Carotinoiden. Ferner können sie als Riech- stoffe mit fixierenden Eigenschaften und mit einem an p-Ionen erinnernden Geruch verwendet werden.
Beispiel 1 Retrodehydro-C11-acetal
Eine Losmg von 16 Gewichtsteilen 2, 6, 6 Trimethyl-eyelohexen- (2)-yliden-aeetaldehyd in 18 Raumteilen OrthoameisensÏureÏthylester wird mit einer Lösung von 0, 3 Raumteilen Orthophosphorsäure in 3 Raumteilen absolutem Aithanol versetzt und 15 Stunden bei 20 bis 251, C stehengelassen. Dann f gt man n 5 Raumteile Pyridin zu und giesst auf eine Mischung von 20 Gewiehtsteilen 5% iger Natriumbiearbonatlösung und 10 Gewiehtsteilen
Eis. Man nimmt in Petroläther auf, schüttelt mit Natriumbicarbonatlosung und troeknet iiber Pottasche.
Nach dem Einengen der Petrolätherlösung wird der Rüekstand im Va kuum bei 70 C von übersehüssigem Ortho ameisensäureäthylester und entstandenem Ameisensäureäthylester befreit. Man erhält 22 Gewichtsteile rohes 2, 6, 6-Trimethyl-cyelo- hexen- (2-yliden-acetaldehyd-diäthylacetal, das f r die weitere Verarbeitung rein genug ist. Dureh Destillation kann man ein farbloses Produkt erhalten ; Kpl3 : 130 bis 132 C ; n23 = 1, 4805 ; U. V.-Absorptionsmaximum bei 240 mÁ (in PetrolÏtherl¯sung).
Retrodehydro-Cl4-atheracetal 20 Gewichtsteile 2, 6, 6-Trimethyl-cyclohexen-(2)-yliden-acetaldehyd-diÏthylacetal werden mit 1 Raumteil einer 10% igen L¯sung von Zinkehlorid in Essigester versetzt. Man gibt dann unter Rühren bei 25 bis 35 C im Verlaufe von 2 Stunden gleichzeitig 8 Raumteile Propenyläthyläther und 6 Raumteile einer 10% igen Lösung von Zinkehlorid in Essigester zu und lässt anschliessend noch 15 Stunden bei Raumtemperatur stehen.
Man nimmt dann in Petroläther auf, wäscht mit verd nnter Natronlauge und troeknet über Pottasche. Nach dem Abdestillieren des L¯ sungsmittels erhält man 28 Gewiehtsteile rohes 4- [2', 6', 6'-Trimethyl-cyclohexen- (2')-yliden]-2 methyl-butanäther- (3)-diäthylacetal- (1), das ohne weitere Reinigung weiter umgesetzt werden kann. Durch Destillation kann ein farbloses Produkt vom KPO, 3 : 120 bis 125 C erhalten werden ; n22 = 1, 4715 ; U. V.-Absorptionsmaximum bei 235 m, (in Petroläther- losung).
Retrodehydro-C14-aldehyd
28 Gewichtsteile 4- [2', 6', 6'-Trimethyl-cyclohexen- (2')-yliden]-2-methyl-butan-äther- (3) diäthylacetal- (l) werden mit 60 Raumteilen Eisessig, 6 Gewichtsteilen Natriumacetat und 3 Raumteilen Wasser versetzt und nach Zugabe einer Spur Hydrochinon während 6 Stunden in Stickstoffatmosphäre auf 95 C erhitzt. Anschliessend kühlt man auf 30 bis 40 C ab und giesst auf 50 Gewiehtsteile Eis und 50 Raumteile Wasser. Man nimmt das ¯lige Reaktionsprodukt in Petroläther auf, wäscht mit Natriumbicarbonatlösung und mit Wasser und trocknet über Natriumsulfat. Nach dem Einengen der Petrolätherlosung wird der Rückstand im Hochvakuum destilliert.
Man erhält 14 Gewichtsteile 4- [2', 6', 6'-Trimethyl- eyelohexen- (2')-yliden]-2-methyl-buten- (2)-al (1) vom Kpoo5 : 94 C ; nD = 1, 610 ; U. V.-Absorptionsmaximum bei 318 mÁ E11= 1670 (in Petrolätherlosung).
Beispiel 2 Iso-C14-aZdehyd
Eine Lösung von 110 Gewichtsteilen 2, 6, 6 Trimethyl-cyclohexyliden-acetaldehyd in 120 Raumteilen Orthoameisensäureäthylester wird mit einer Losung von 2 Raumteilen Orthophosphorsäure in 18 Raumteilen absolutem Äthanol versetzt und 15 Stunden bei 20 bis 25 C stehen gelasse. Dann f gt man 20 Raumteile Pyridin zu und giesst auf eine Mischung von 200 Gewichtsteilen 5%iger Natriumbicarbonatlosung und 100 Gewichtsteilen Eis. Man nimmt in Petroläther auf, schüttelt mit Natrium bicarbonatlosung und trocknet über Pottasche.
Nach dem Einengen der Petrolätherlosung wird der Rüekstand im Vakuum bei 70 C von überschüssigem Orthoameisensäureäthylester und entstandenem Ameisensäureäthylester befreit. Der Rückstand besteht aus 150 Ge wichtsteilen 2, 6, 6'-Trimethyl-cyclohexyliden acetaldehyddiäthylacetal vom n23 = 1, 464, das im LTltraviolettspektrum kein Absorptionsmaximum ber 225 mÁ aufweist. Dieses Produkt wird ohne weitere Reinigung mit Propenyläther kondensiert.
Zu diesem Zwecke fügt man 9 Raumteile einer 10% igen L¯sung von Zinkehlorid in Essigester zu, gibt dann unter Rühren bei 30 bis 35 C im Verlaufe von 2 Stunden gleichzeitig 65 Raumteile Propenyl- äthyläther und 43 Raumteile einer 10 igen Lösung von Zinkchlorid in Essigester zu und rührt anschliessend noch 20 Stunden bei Raumtemperatur.
Das so erhaltene 4- [2', 6', 6'-Tri methyl-cyclohexyliden]-2-methyl-butan-äther- (3)-diäthylaeetal (l) vom nD3 = 1, 459 wird zwecks Verseifung und Abspaltung von Alkohol zu einer Mischung von 450 Raumteilen Eisessig, 45 Gewiehtsteilen Natriumacetat und 22 Raumteilen Wasser zugefügt und wahrend 6 Stunden in Stiekstoffatmosphäre auf 95 C erhitzt. Anschlie¯end kühlt man auf 30 bis 40 C ab und giesst auf 400 Gewichtsteile Eis und 400 Raumteile Wasser. Man nimmt das ölige Reaktionsprodukt in Petroläther auf, wäscht mit 5% iger Natriumbiearbonatlösung und mit Wasser und trocknet ber Natrumsulfat.
Nach dem Einengen der Petroläther- lösung wird der Rüekstand im Hochvakuum destilliert. Man erhält 90 Gewichtsteile 4 [2', 6', 6'-Trimethyl-cyclohexyliden]-2-methylbuten-(2)-al-(1) vom Kp0,05 : 82 bis 84¯ C ; nD3 = 1, 547 ; U. V.-Absorptionsmaximum bei 288 m, u, E 11= 1400 (in Petrolätherlösung).
Claims (1)
- PATENTANSPRUCH Verfahren zur Herstellung eines Polyen- aldehyds, dadurch gekennzeichnet, dass man einen Aldehyd der Formel R=CFI-CHO, worin R den 2, 6, 6-Trimethyl-cyclohexylidenoder den 2, 6, 6-Trimethyl-cyclohexen-(2)-ylidenrest bedeutet, acetalisiert, an das erhaltene Aceta in Gegenwart eines sauren Mittels einen Propenyläther anlagert und das erhaltene An lagerungsprodukt mit sauer reagierenden Stoffen behandelt.UNTERANSPRUCHE 1. Verfahren nach Patentanspruch, da- durch gekennzeichnet, daB man die Ausgangsverbindung mit einem niederen Alkohol aeetalisiert und an das erhaltene Aeetal den Pro penyläther des gleichen niederen Alkohols an- lagert.2. Verfahren nach Patentanspruch und lTnteransprueh 1, dadureh gekennzeiehnet, dass man die äquivalente Menge Propenyläther bei 15 bis 50 C in Gegenwart von Zinkehlorid auf das Aeetal einwirken lϯt.3. Verfahren nach Patentansprueh und Unteransprüchenlund2,dadurchgekenn- zeichnet, dass man das erhaltene Anlagerungsprodukt mit Essigsa. ure in Gegenwart von Na triumacetat erwärmt.4. Verfahren naeh Patentansprueh und Unteransprüchen 1 bis 3, dadurch gekennzeichnet, da¯ man die Zwischenprodukte undestilliert weiterverarbeitet.
