REVENDICATION
1. Procédé de préparation de l'acide l-(p-azidobenzoyl)-5-méthoxy-2-méthyl-3-indolylacétique et de ses sels pharmaceutique-ment acceptables, caractérisé en ce qu'on fait réagir un ester de l'acide 5-méthoxy-2-méthyl-3-indolylacétique avec un halogénure de p-azidobenzoyle, en ce qu'on hydrolyse le groupe ester du produit obtenu et en ce que, éventuellement, on transforme l'acide en un sel pharmaceutiquement acceptable.
Des acides 1-aroyl- et hétéroaroyl-3-indolylaliphatiques inférieurs sont connus et possèdent un degré élevé d'activité antiinflammatoire et sont efficaces pour prévenir et inhiber la formation de tissus de granulome. Certains de ces composés sont utiles pour lei traitement de troubles arthritiques et dermatologiques et des états analogues qui répondent à un traitement avec des agents anti-inflammatoires. Un des composés le plus actif est connu sous le nom commercial d'Indométhacine qui est l'acide 1-p-chlorobenzoyl-5-méthoxy-2-méthyl-3-indolylacétique.
La présente invention a pour objet le procédé défini dans la revendication 1, pour la préparation de l'acide l-(p-azidobenzoyl)-5-méthoxy-2-méthyl-3-indolylacétique c'est-à-dire du composé de formule:

et de ses sels pharmaceutiquement acceptables.
On a trouvé que la présence du substituant azido dans le groupe benzoyle donne le composé ayant une activité pharmacologique analogue à celle de l'Indométhacine mais, et cela est surprenant, avec une toxicité et des effets secondaires désavantageux bien moindres.
Le présent composé est une poudre cristalline jaune qui fond à 170-172' C avec décomposition. Les cristaux sont solubles dans le diméthylformamide et le diméthylsulfoxyde, légèrement solubles dans le méthanol et l'éthanol et pratiquement insolubles dans l'eau.
Le procédé part d'un ester d'acide indolylacétique de formule II:

il dans laquelle R est un groupe alcoyle inférieur, un groupe aralcoyle ou vinylique, que l'on peut obtenir par estérification de l'acide libre, qui est connu, avec l'alcool correspondant, ou directement à partir de l'ester désiré de l'acide livulinique dans la synthèse originale du noyau indole ou par la transestérification d'autres esters.
L'acylation de l'atome d'azote de l'indole est conduite de préférence en traitant un composé de formule II avec un hydrure de métal alcalin pour former un sel de métal, puis en le mettant en contact intime avec un halogénure de p-azidobenzoyle dans un s solvant anhydre comme milieu. On peut alors enlever le groupe ester de diverses manières.
L'activité anti-inflammatoire du présent composé a été évaluée chez les rats par l'essai du granulome induit par une boulette de coton (Meier R., Schuler W. et Dessaules, «Experientia», 1950, 6, io 469), en administrant le composé par voie orale chaque jour pendant 7 j.
L'activité antiœdème du composé a été évaluée en l'administrant de façon aiguë avant l'injection de carragénine dans la surface plantaire d'une patte de rat, puis en mesurant l'inhibition (Winter 15 C.A., Risley E.A. et Nuss G.W., «J. Pharmacol. Exp. Ther.», 1963, 141,369), du volume de l'œdème.
Le composé agit contre la formation d'un granulome et de l'œdème à une dose de 3 mg/kg de poids du corps par voie orale (inhibition approximative de 29% et 46% respectivement). Dans 20 l'essai du granulome, le composé était légèrement moins actif que l'Indométhacine, mais il était aussi actif que l'Indométhacine pour inhiber la formation de l'œdème.
On a évalué l'activité analgésique chez les souris en mesurant le pourcentage d'animaux protégés de l'essai de douleurs intenses 25 induit par l'injection intrapéritonéale de phénylquinone (Henders L.C., Forsaith J., «J. Pharmacol. Exp. Ther.», 1959,125, 237). La valeur DE50 orale (dose empêchant 50% des souris de se tordre de douleur) était d'environ 2,5 mg/kg de poids du corps tandis que celle de l'Indométhacine était de 1,25 mg/kg de poids du corps. 30 Les valeurs DL50 après l'administration aiguë par voie orale du composé chez des souris et des rats mâles étaient de 250 mg/kg de poids du corps et de 85 mg/kg de poids du corps respectivement, tandis que les valeurs DL50 correspondantes pour l'Indométhacine étaient de 13,5 mg/kg et de 10 mg/kg de poids du corps; les valeurs 35 DL50 chez les souris et les rats femelles étaient respectivement de 240 mg/kg et de 105 mg/kg de poids du corps pour le présent composé et de 23 mg/kg et de 17 mg/kg de poids du corps pour l'Indométhacine. Lorsqu'on l'a administré par voie orale à des rats à différentes doses, le composé s'est montré environ 10 fois moins 40 ulcérogène que l'Indométhacine.

45 A une solution de 23,3 g (0,1 mol) de 5-méthoxy-2-méthyl-3-indolylacétate de méthyle dans 50 ml de toluène sec, on ajoute 3 g d'hydrure de sodium à 80%. On agite le mélange à la température ordinaire pendant 4 h, puis on y ajoute lentement une solution de 18,16 g (0,1 mol) de chlorure de p-azidobenzoyle dans 80 ml de so toluène sec en 30 mn.
On agite le mélange réactionnel sous azote pendant 48 h à 50-60° C. Après refroidissement, on le verse dans 400 ml d'eau glacée et 15 ml d'acide acétique.
On lave la solution toluénique séparée avec une grande quantité 55 d'eau, on sèche sur du sulfate de sodium et on évapore pour obtenir un sirop qu'on dissout dans l'éther.
L'évaporation lente de cette solution dans un becher ouvert donne le l-(p-azidobenzoyl)-5-méthoxy-2-méthyl-3-indolylacétate de méthyle pratiquement pur de couleur jaune.
60 Le spectre infrarouge ne montre pas d'absorption N—H au voisinage de la région de 2,9-3,0 ia, mais un doublet fort à 4,7-4,75 ^ indique la présence d'un groupe azido, et des absorptions C=0 à 5,75 et 5,98 (j. sont caractéristiques des groupes fonctionnels ester et amide respectivement.
« B. Préparation de l'acide l-(p-azidobenzoyl)-5-mêthoxy-2-méthyl-3-indolylacétique.
On chauffe à reflux pendant 20 h une solution de 4,85 g (12,8 mmol) de l-(p-azidobenzoyl)-5-méthoxy-2-méthyl-3-indolylacétate

de méthyle dans 40 ml d'acide acétique contenant 400 mg d'acide p-toluènesulfonique, puis on concentre sous vide.
On extrait le résidu gommeux à l'acétate d'éthyle.
On filtre l'extrait pour en séparer la substance insoluble, on lave à l'eau et on sèche sur du sulfate de sodium.
La séparation du solvant sous pression réduite donne le produit désiré.
La Chromatographie sur couche mince sur du gel de silice donne une tache'dans un mélange 95:5 de chloroforme: éthanol.
On obtient un échantillon analytique par cristallisation à partir d'un mélange de méthanol et d'eau; il fond à 170-172°C avec 5 dégagement de gaz; IR (KBr) doublet à 4,69-4,76 y. (N3), 5,85 y. (acide C = O) 5,97 u. (amide C = O).
