Der er et tvingende behov for hidtil ukendte lægemidler til behandling af mentale lidelser, hvilke lægemidler er mere virkningsfulde og har færre bivirkninger end de lægemidler, der i dag anvendes klinisk.
De for tiden anvendte antipsykotiske lægemidler fremkalder en række 5 plagsomme ekstrapyramidale bevægelseslidelser (fx akutte dystoniske reaktioner og tardiv dyskinesi), og de er dårlige til at bedre de negative symptomer (fx begrænset eller sløvet emotionel vækkelse) på skizofreni. Hovedulempen ved de antidepressive midler er, at de ikke kan lette depression hos 30-40% af patienterne. Anxiolytiske midler 10 er sædvanligvis forbundet med vanedannende egenskaber.
Inden for teknikken kendes forskellige pyridyl- og pyrimidyl-pipera-zinderivater, der er farmakologisk aktive i centralnervesystemet. Der kan nævnes nogle repræsentative eksempler. Azaperon, et neuroleptisk lægemiddel af butyrophenon-klassen, er et beroligende middel til gri-15 se. Buspiron er et anxiolytisk middel. Den anxiolytiske virkning formodes at blive formidlet ved hjælp af indvirkning på 5HT-receptorerne .
-O- COCH2CH2CH2— i/ 30 azaperon 0 u buspiron I US 2.958.694 beskrives azaperon, som kan karakteriseres som en butyrophenon. Andre typer butyrophenoner, reducerede butyrophenoner 20 eller butyrophenonderivater er beskrevet i US 2.979.508, US 2.985.657 og DE 35 07 983 Al. I modsætning hertil er forbindelserne ifølge den foreliggende opfindelse diphenylbutylpiperazinderivater, der tilhører en anden klasse af forbindelser.
Det har overraskende vist sig, at pyridyl- og pyrimidyl-piperaziner, 25 der er substitueret i piperazinringens eller diazacycloheptanringens

2 4-stilling med en stærk lipofil diphenyl-butylgruppe, udviser farmakologiske egenskaber, der er overlegne i forhold til kendte forbindelser inden for teknikken.
Ifølge opfindelsen tilvejebringes hidtil ukendte 4-(bis-fluorphenyl-5 butyl-l-(pyridyl- eller pyrimidyl)-piperazin- eller -1,4-diazacyclo-heptanforbindelser med den almene formel (I) / _n i i n-a (I) V*V2 «>/>„ 10 hvor Rj^ betegner hydrogen eller fluor; R3 og R4 er ens eller forskellige og valgt blandt hydrogen og lavere alkyl; n betegner tallet 2 eller 3; A er valgt blandt de nedenstående pyrimidyl- og pyridylgrupper 15 -o <$1 -g:; hvor Rg og R7 er ens eller forskellige og valgt blandt hydrogen, halogen, lavere alkyl, lavere alkoxy, hydroxy, cyano, nitro, tri-fluormethyl, COORg, GONRgR^o eller CO-B; hvor Rg betegner hydrogen 20 eller lavere alkyl; Rg og R^o er ens eller forskellige og valgt blandt hydrogen og lavere alkyl, og B betegner morpholino, pyrrolidino eller 4-methyl-piperidino, samt farmakologisk acceptable syreadditionssalte deraf.
Anvendt i de ovenfor anførte definitioner skal udtrykket lavere alkyl 25 omfatte ligekædede og forgrenede, mættede og umættede carborihydrid-grupper med 1-5 carbonatomer; udtrykket lavere alkoxy skal omfatte ligekædede eller forgrenede, mættede eller umættede alkoxygrupper med 1-5 carbonatomer;

3 udtrykket halogen skal omfatte fluor, chlor og brom.
Hvad angår R3 og R4 foretrækkes hydrogen eller methyl, især hydrogen.
Hvad angår Rg foretrækkes hydrogen, methyl, alkoxy, amid, nitro, tri-fluormethyl, halogen eller cyano.

Forbindelser, hvor A er 2-substitueret pyridyl, er af særlig interesse, især sådanne med en methoxy-, amid-, cyano- eller hydrogensub-stituent i 3-stillingen.
10 Forbindelserne med formlen (I) har basiske egenskaber og kan som følge deraf omdannes til terapeutisk aktive syreadditionssalte deraf ved behandling med hensigtsmæssige syrer; fx uorganiske syrer såsom saltsyre, brombrintesyre, svovlsyre, salpetersyre og phosphorsyre, eller organiske syrer såsom eddike-, propan-, glycol-, mælke-, malon-15 , oxal-, rav-, fumar-, vin-, citron- og pamoinsyre.
Omvendt kan saltformerne omdannes til den frie baseform ved behandling med alkali.
Forbindelser med formlen (I) og farmaceutisk acceptable syreadditionssalte deraf har værdifulde farmakologiske egenskaber, der gør 20 dem nyttige til behandling af mentale lidelser såsom psykoser, depression og angst. Stress og angst hos dyr kan også behandles.
Således kan forbindelser med formlen (I) og farmakologisk acceptable syreadditionssalte deraf ifølge opfindelsen anvendes til fremstilling af farmaceutiske præparater til behandling af mental lidelse hos et 25 levende væsen.
Sådanne farmaceutiske præparater udgør et yderligere aspekt af opfindelsen og indeholder én eller flere af forbindelserne med formlen (I) eller farmakologisk acceptable syreadditionssalte deraf som aktiv

4 bestanddel, fortrinsvis sammen med et farmaceutisk acceptabelt bærestof og, om ønsket, andre farmakologisk aktive midler.
Det er blevet formodet, at de antipsykotiske virkninger af neurolep-tiske lægemidler er uadskilleligt forbundne med deres formåen til at 5 fremkalde ekstrapyramidale bivirkninger, muligvis ved en virkning på et fælles site. Denne formodning sættes der nu spørgsmålstegn ved, eftersom antipsykotiske lægemidler i vore dage har et reduceret potentiale til at fremkalde disse bivirkninger. Således formodes det, at de to hovedvirkninger af klassiske neuroleptiske lægemidler, dvs.
10 antipsykotisk virkning og formåen til at fremkalde ekstrapyramidale syndromer, formidles ved indvirkninger på forskellige sites i hjernen. De neuroleptiske lægemidler kan fremkalde deres terapeutiske an-tipsykotiske virkninger ved en indvirkning på det mesolimbiske system. På den anden side er dette site i hjernen sædvanligvis formodet 15 at være det område, hvor de neuroleptiske lægemidler fremkalder deres ekstrapyramidale bivirkninger, eftersom hovedfunktionen af den nigro-striatale dopaminerge bane er styringen af motorisk udøvelse. Eksperimentalt måler inhibering af amphetamin-induceret hypermotilitet hos gnavere den limbiske virkning, mens det generelt accepteres, at neu-20 roleptikas formåen til at inducere catalepsi hos gnavere er godt korrelerede til risikoen for ekstrapyramidale bivirkninger.
Forbindelserne ifølge den foreliggende opfindelse udviser neuroleptiske egenskaber, eftersom de indvirker på amphetamin-induceret hypermotilitet, men udviser svag eller ingen cataleptogen aktivitet. De 25 viser en høj affinitet overfor D2- og 5-HT2 receptorer i hjernen. En høj 5HT2~receptor bindingsaffinitet kan indikere antidepressive egenskaber, eftersom mange antidepressive midler (fx amitriptylin) er potente 5ΗΪ2-blokkere. En høj 5HT2-receptor-bindingsaffinitet tyder også på anxiolytiske egenskaber. Et hidtil ukendt anxiolytisk middel, 30 ritanserin, mærker fortrinsvis 5-HT2-bindingssite.
Virkningsfulde mængder af et hvilket som helst af de ovenfor nævnte farmakologisk aktive stoffer med formlen (I) kan administreres til et menneske eller et dyr til terapeutiske formål via sædvanlige administrationsveje og i sædvanlige former såsom oralt i opløsninger, emul-35 sioner, suspensioner, piller, tabletter og kapsler, i farmaceutisk

5 acceptable bærestoffer og parenteralt i form af sterile opløsninger.
Ved den parenterale administration af det aktive stof kan bærestoffet eller excipiensen være en steril, parenteralt acceptabel væske, fx vand, eller en parenteralt acceptabel olie, fx arachidinolie.
5 Skønt meget små mængder af de aktive materialer ifølge den foreliggende opfindelse er virkningsfulde, når der er tale om mindre omfattende terapi eller i tilfælde af administration til patienter med en relativ lav kropsvægt, er enhedsdoser sædvanligvis fra 2 mg og opefter, fortrinsvis 10, 25 eller 50 mg eller endnu højere, afhængig 10 af den tilstand, der skal behandles, og af patienternes alder og vægt samt respons på den medicinske behandling.
Enhedsdosis kan være fra 0,1-100 mg, fortrinsvis fra 10-50 mg. Daglige doser bør fortrinsvis være i området fra 10 mg til 200 mg. De præcise individuelle doser samt de daglige doser vil naturligvis blive 15 fastlagt efter sædvanlige lægelige principper ved anvisning af en læge eller dyrlæge.
Forbindelserne med den almene formel (I) kan fremstilles ved konventionelle fremgangsmåder.
Opfindelsen angår også sådanne fremgangsmåder, som er ejendommelige 20 ved det i krav 8's kendetegnende del angivne.

7 EKSEMPEL 1 4-[4,4-bis(p-fluorphenyl)butyl]-1-(5-chlor-2-pyridyl)-piperazindihy-drogenchlorid 19,8 g (0,06 mol) 1-[4,4-bis(p-fluorphenyl)butyl]-piperazin og 2,96 g 5 (0,02 mol) 2,5-dichlorpyridin blev opvarmet sammen med 2 ml toluen ved 130°C (oliebadets temperatur) i 20 timer.
Efter afkøling blev reaktionsblandingen delt mellem ligroin og Me0H/H20 (3:1). MeOH/l^O-fasen blev ekstraheret yderligere en gang med ligroin, og de samlede ligroinfaser blev tørret med Na2S04· For-10 dampning af opløsningsmidlerne gav rå fri base, der blev opløst i ether, og hydrogenchloridet blev fældet med overskydende HC1 i EtOH. Omkrystallisation fra EtOAc/EtOH gav 4,3 g (42%) af titelforbindelsen (1:1), smp. 173-176°C.

Omkrystallisation fra EtOH, EtOAc + ether gav 7,3 g (65%) af titelforbindelsen (2:1), smp. 195-197°C.
EKSEMPEL 3 4-[4,4-bis(p-fluorphenyl)butyl]-1-(2-pyrimidyl)-piperazin dihydro-genchlorid 20 3,3 g (0,01 mol) l-chlor-4,4-bis(p-fluorphenyl)butan, 3,3 g (0,02 mol) 1-(2-pyridyl)-piperazin og 0,05 g Kl blev tilbagesvalet i 15 ml toluen i 48 timer. Efter afkøling og tilsætning af ether (30 ml) blev 1-(2-pyridyl)-piperazin hydrogenchlorid udfældet og frafiltreret. Efter efterfølgende adskillige ganges vask med Η£θ blev 25 den organiske fase tørret med K2CO3. Afdampning af opløsningsmidlet gav den rå base. Denne blev opløst i ether, og HCl i EtOH blev tilsat 11

for at fælde hydrogenchloridet. Omkrystallisation fra EtOAc/EtOH gav 3,2 g (67%) af titelforbindelsen (2:1), smp. 224-227°C.
EKSEMPEL 4
Dette eksempel illustrerer styrken af forbindelser med formlen (I) og 5 farmaceutisk aktive salte deraf til behandling af mentale lidelser.
Test 1 - Antagonisme af amphetamin-induceret hypermotilitet hos mus.
En lav dosis (2 mg/kg) d-amphetamin administreret til mus inducerede udtalt lokomotorisk aktivitet.
Testprocedure 10 NMRI-hunmus med en vægt på ca. 25 g blev delt i grupper på hver 4 dyr og placeret i macrolonbure. Burene blev placeret på en registrerende arena i 9 lyddæmpede kasser med konstant lys og ventilation. Efter en tilvænningstid på 90 minutter blev musene taget ud af burene og behandlet på den følgende måde; en kontrolgruppe modtog saltvand 15 i.p., en kontrolgruppe modtog d-amphetamin-sulfat, 2 mg/kg i.p., og testdyrene blev behandlet subcutant med tre forskellige doser af testforbindelsen og umiddelbart derefter med d-amphetamin 2 mg/kg i.p. Umiddelbart efter administrationen blev macrolonburene med musene atter anbragt i testkasserne, og et computertilsluttet udstyr 20 registrerer den lokomotoriske aktivitet i burene i 90 minutter.
For hver testforbindelse blev 4 bure behandlet med saltvand, 4 bure blev behandlet udelukkende med d-amphetamin, og 6 bure/dosis testforbindelse + d-amphetamin. Dette er summen af tre forskellige eksperimenter.
25 Dataanalyse
For hver behandlingsgruppe blev gennemsnittet af tællinger i løbet af 90 minutter efter administrationen beregnet.

12 ED^q-værdien (mg/kg) blev beregnet ud fra dosis-responskurven ved hjælp af lineær regression. ED5Q-værdien repræsenterer en dosis af testforbindelse, der nedsætter motiliteten til punktet midt mellem amphetamin- og saltvands-kontrollerne.
5 Test 2 - Affinitet til Dj-receptorer
Bindingsprøven udføres i det væsentlige som beskrevet af Leysen et al., (Mol. Pharmacol. 21, 301-314, 1982) ved hjælp af 3H-spiroperidol som ligand.

Den overraskende, nyttige virkning af forbindelserne ifølge opfindelsen i forhold til den narliggende, kendte forbindelse azaperon (1-(4-fluorphenyl)-4-[4-(2-pyridinyl)-1-piperazinyl]-1-butanon, kendt fra US 2.979.508) kan ses af følgende ED50-værdier (mg/kg) for ad-5 færdsmodellen "inhibering af amfetamin-inducerede stereotopier":

Flere litteratursteder fremfører betydningen af såvel limbisk som striatal dopaminneuron i medieringen af henholdsvis lokomotorisk aktivitet og stereotyp adfard. Af ovenstående tabeller fremgår det 15 tydeligt, at forbindelserne ifølge opfindelsen inhiberer amfetamininduceret lokomotorisk aktivitet, mens de ikke eller kun ubetydeligt påvirker stereotyp adfærd. Eftersom de striatale dopaminneuroner påvirker motorikken, er det formodentlig neuroleptikas virkninger herpå, som forårsager de meget svære motoriske bivirkninger.
20 Endvidere fremgår det af de ovenstående tabeller, at forbindelserne ifølge opfindelsen har et fordelagtigt 5HT2/D2-forhold. Dette tyder på, at dopamintransmissionen medieres via en 5HT2-effekt og ikke via blokering af D2-blokade. En alt for kraftig D2-blokade anses for at være en af årsagerne til de motoriske bivirkninger. En høj 5HTla-affini-tet indebærer, at forbindelserne har gode anxiolytiske egenskaber. Af ovenstående tabeller fremgår det også, at azaperon, der er en kendt forbindelse, som er nærliggende i forhold til forbindelserne ifølge 30 opfindelsen, har en kraftig virkning på striatal dopaminneuron, hvilket vil sige en kraftig bivirkning, hvilket ses ikke at være tilfældet for de repræsentative forbindelser ifølge opfindelsen.

Disse opløsninger kan også indeholde stabiliserende midler og/eller puffer og kan hensigtsmæssigt tilvejebringes i forskellige dosiserihedsampuller.
