Eksempler på egnede syrer til saltdannelse er salt-, svovl-, phosphor-, eddike-, citron-, oxal-, malon-, salicyl-, æble-, glucon-, furnar-, rav-, ascorbin-, malein-, 15 methansulfonsyre og lignende. Saltene fremstilles ved at bringe den frie baseform i kontakt med en tilstrækkelig mængde af den ønskede syre til dannelse af enten et mono^-salt, disalt osv. på konventionel måde. De frie baseformer kan regenereres ved at behandle saltformen med en 20 base. For eksempel kan der anvendes fortyndede opløsninger af vandig base. Flydende præparatformer omfatter opløsninger, suspensioner og emulsioner. Som eksempel kan nævnes vandeller vand/propylenglycol-opløsninger til parenteral in-3Q jektion. Sådanne opløsninger fremstilles således, at de er acceptable for biologiske systemer (isotonicitet, pH osv.). Flydende præparater kan også foreligge som opløsning i vandig polvethylenglycolopløsning. Vandige opløsninger egnede til oral brug kan fremstilles ved at oplø-35 se den aktive komponent i vand og tilsætte egnede farvningsmidler, smagsstoffer, stabiliseringsmidler og fortykningsmidler, som ønsket. 5 En suspension af 0,59 g (1,54 mmol) ethyl-7-brom- acetyl-l-ethyl-6-fluor-1,4-dihydro-4-oxo-3-quinolincar-boxylat i 4 ml Ν,Ν-dimethylformamid blev behandlet med 0,22 g (1,67 mmol) acetamidothioacetamid [fremstillet i-følge J.Am.Chem.Soc. 51, 1817 (1929)]. Efter omrøring 3 10 timer blev blandingen behandlet med 0,22 ml (1,58 mmol) triethylamin, omrørt 1 time og hældt i 40 ml isvand. En opløsning af 0,32 g (0,76 mmol) ethyl-7-[2-(ace-tamidomethyl)-4-thiazolyl]-l-ethyl-6-fluor-1,4-dihydro-20 4-oxo-3-quinolincarboxylat i 3 ml 6N saltsyre blev omrørt under tilbagesvaling i 3 timer. Blandingen blev omrørt 72 timer ved stuetemperatur. Til 500 mg (1,37 mmol) 7-[2-(aminomethyl)-4-thiazolyl] -l-ethyl-6,8-difluor-l,4-dihydro-4-oxo-3-quinolin-25 carboxylsyre blev der sat 15 ml 40% vandig methylamin. Blandingen blev opvarmet ved 50°C i 72 timer. Eksempel 8 l-Ethyl-6-fluor-l,4-dihydro-8-methoxy-7-[2-[(methylamino) methyl] -4-thiazolyl]-4-oxo-3-quinolincarboxylsyre.
35 Til 500 mg (1,32 mmol) l-ethyl-6,8-difluor-l,4-di- hydro-7-[2-[(methylamino)methyl]-4-thiazolyl]-4-oxo-3-quinolincarboxylsyre i 30 ml tørt methanol blev der sat 20 DK 170471 B1 300 mg (2 ækv.) kalium-t-butoxid. Efter 24 timers tilbagesvaling blev blandingen afkølet, fortyndet med vand, bragt til pH 6,0 og filtreret, hvorved vandtes 310 mg l-ethyl-6-fluor-1,4-dihydro-8-methoxy-7-[2-[(methylami-5 no)methyl]-4-thiazolyl]-4-oxo-3-quinolincarboxylsyre, smp. 164-166°C. * På tilsvarende måde blev der fremstillet 7-[2-(aminomethyl)-4-thiazolyl]-l-ethyl-6-fluor-1,4-dihydro- 8-methoxy-4-oxo-3-quinolincarboxylsyre, smp. 172-175°C, 10 ( 8a).
Eksempel 9 l-Ethyl-6-fluor-l,4-dihydro-8-hydroxy-7-[2-[(methylami-no) methyl]-4-thiazolyl]-4-oxo-3-quinolincarboxylsyre.
15 Til 700 mg (1,79 mmol) l-ethyl-6-fluor-1,4-dihydro- 8-methoxy-7-[2-[(methylamino)methyl]-4-thiazolyl]-4-oxo- 3-quinolincarboxylsyre blev der såt 8 ml hydrogenbromid-syre i eddikesyre, og blandingen blev opvarmet til 70°C.

På tilsvarende måde blev der fremstillet 7-[2-{ami-nomethyl)-4-thiazolyl]-l-ethyl-6-fluor-1,4-dihydro-8-hydroxy-4-oxo-3-quinolincarboxylsyre, smp. 275-277°C, ( 9a).

En opløsning af 0,16 g (0,5 mmol) ethyl-l-ethyl-6-35 fluor-7-thiocarbamoyl-l,4-dihydro-4-oxo-3-quinolincarbo-xylat og 0,32 g (2,5 mmol) 1,3-dichloracetone i 3 ml Ν,Ν-dimethylformamid blev opvarmet 3,5 time på dampbad.

15 En opløsning af 0,87 g (2,17 mmol) ethyl-7-[4- (azidomethyl). -2-thiazolyl] -l-ethyl-6-fluor-1,4-dihydro- 4-oxo-3-quinolincarboxylat i 125 ml eddikesyre blev omrørt med 0,10 g 10% palladium-på-kul-katalysator, og luftformigt hydrogen blev boblet igennem i 1,5 time. Ef-20 ter filtrering, fordampning af opløsningsmiddel og tri-turering med ether vandtes 0,77 g ethyl-7-[4-(aminome-thyl)-2-thiazolyl]-l-ethyl-6-fluor-1,4-dihydro-4-oxo-3-quinolincarboxylat.
En opløsning af 0,70 g (1,87 mmol) ethyl-7-[4-25 (aminomethyl)-2-thiazolyl]-l-ethyl-6-fluor-1,4-dihydro- 4-oxo-3-quinolincarboxylat i 15 ml 6N saltsyre blev opvarmet på dampbad 2,25 time.Efter tilsætning af 15 ml vand blev blandingen afkølet til 0°C og filtreret. Det indsamlede faste stof blev suspenderet i 8 ml vand, op-30 løst ved pH 11 med 2N natriumhydroxid og genudfældet ved tilsætning af 2N saltsyre til pH 6. Fremstilling af Udgangsmaterialer Ethyl-7-acetyl-l-ethvl-6-fluor-l, 4-dihydro-4-oxo-3-quinolincarboxvlat 5 En opløsning af 26,4 g (0,17 mol) 3-acetyl-4-fluor- anilin og 34,6 ml (0,35 mol) 1,3-propandithiol i 2,5 liter chloroform blev afkølet til 5°C. Luftformigt hydro-genchlorid blev boblet ind i den omrørte opløsning i 15 minutter ved 5-10°C, og reaktionsblandingen blev opvar-10 met til stuetemperatur under omrøring natten over. Opløsningsmidlet blev fjernet i vakuum ved 60°C, og den faste inddampningsrest blev to gange optaget i 800 ml toluen og strippet i vakuum ved 60°C. Det faste materiale blev opløst i 1,5 liter chloroform, vasket to gange 15 med 200 ml mættet natriumbicarbonatopløsning, to gange med 200 ml vand og tørret med magnesiumsulfat. Opløsningsmidlet blev fjernet i vakuum ved 65°C, og inddampningsresten blev opløst i 2 liter chloroform, vasket godt med vand og tørret med magnesiumsulfat. Fjernelse af opløsningsmidlet i vakuum 10 gav 48,2 g af en lysebrun olie, ethyl-l-ethyl-6-fluor- 1,4-dihydro-7-(2-methyl-l,3-dithian-2-yl)-4-oxo-3-quino-lincarboxylat. Uden yderligere rensning blev dette materiale opløst i 1 liter 80% acetonitril/vand og i løbet af 30 minutter ved stuetemperatur og under nitrogen sat 15 til en godt omrørt suspension af 28,9 g (0,13 mol) mer-curioxid og 72,0 g (0,27 mol) mercurichlorid i 2 liter 80% acetonitril/vand. Reaktionsblandingen blev opvarmet til tilbagesvaling i 6 timer under nitrogen, afkølet til stuetemperatur og filtreret gennem en pude af Celite.

En opløsning af 2,45 g (8,0 mmol) ethyl-7-acetyl-l-ethyl-6-fluor-1,4-dihydro-4-oxo-3-quinolincarboxylat i 35 50 ml eddikesyre blev behandlet med 0,46 g kaliumbromat, og derefter blev der dråbevis i løbet af en halv time tilsat 3,8 ml 48% hydrogenbromidsyre. Blandingen blev 25 DK 170471 B1 omrørt 24 timer ved stuetemperatur og hældt i 200 ml isvand. Bundfaldet blev frafiltreret, vasket med vand og tørret, hvorved vandtes 2,87 g ethyl-7-bromacetyl-l-ethyl-6-fluor-l,4-dihydro-4-oxo-3-quinolincarboxylat.
5 7-Acetyl-l-ethyl-6,8-difluor-l,4-dihydro-4-oxo-3-quino-lincarboxylsyre 2.6- Difluoracetophenon 10 Til 64,19 g (455 mmol) 2,6-difluorbenzonitril i 300 ml diethylester blev der i løbet af 1 time ved -78°C sat 650 ml 1,6M methyllithium (2,0 ækv.). Blandingen blev omrørt ved -78°C i 2,5 time og derefter behandlet med 250 ml 6N saltsyre. Reaktionsblandingen blev bragt til 5°C og 15 fik lov at nå stuetemperatur i nattens løb. Lagene blev adskilt, og vandlaget blev derefter vasket med dichlor-methan. Nitreringsmidlet blev tilsat med tilstrækkelig hastighed 30 til at holde reaktionstemperaturen ved 5°C. Reaktionsblandingen blev derefter omrørt i 20 minutter og hældt over is. Blandingen blev ekstraheret med dichlormethan to gange. Til 18,1 g (90,0 iranol) 2,6-difluor-3-nitroaceto-phenon blev der sat methanol, Raney-nikkel og luftformigt hydrogen. Da blandingen havde optaget den teoreti-5 ske mængde hydrogen, blev den filtreret ind i et overskud af diethylmethylenmalonat. Methanolet blev fjernet, * og blandingen blev behandlet med toluen, der derefter blev afdestilleret indtil det halve volumen. Blandingen blev derefter koncentreret i vakuum, og inddampningsre-10 sten blev omrørt med ether:pentan, hvorved vandtes 24,4 g 3-acetyl-2,4-difluoranilinomethylenmalonat, smp.82-84°C.
Ethyl-7-acetyl-6,8-difluor-1,4-dihydro-4-oxo-3-quinolin-carboxylat
Til 380 ml tilbagesvalende Dowtherm A blev der sat ^ 14,4 g (42,2 mmol) 3-acetyl-2,4-difluoranilinomethylenmalonat i tre portioner. Reaktionsblandingen blev omrørt i 30 minutter. Efter afkøling blev den behandlet med 500 ml pentan. De faste stoffer blev frafiltreret og vasket med ether:pentan, hvorved vandtes 7,9 g ethyl-7-acetyl- 6.8- difluor-1,4-dihydro-4-oxo-3-quinolincarboxylat, smp. 267-270°C. Dette materiale anvendtes uden yderligere rensning.
Til 22,3 g (76,6 mmol) ethyl-7-acetyl-6,8-difluor- l,4-dihydro-4-oxo-3-quinolincarboxylat i 900 ml dimethyl-^ formamid blev der sat 25,6 g (2,5 ækv.) kaliumcarbonat og 22 ml (3,6 ækv.) ethyliodid. Blandingen blev omrørt ved 45°C natten over. Blandingen blev koncentreret. Ind-dampningsresten blev opløst i vand og ekstraheret over i dichlormethan. Dichlormethanet blev koncentreret, og den Q Λ som inddampningsrest vundne olie blev renset ved søjle-chromatografi på silicagel, hvorved vandtes 10,5 g ethyl-7-acetyl-l-ethyl-6,8-difluor-1,4-dihydro-4-oxo-3-quino-lincarboxylat, smp. 129-130°C.
Til 400 mg (1,23 mmol) af dette materiale blev der sat 5 ml 6N saltsyre, og suspensionen blev omrørt ved 85°C natten over. Filtrering gav 310 mg 7-acetyl-l-ethyl- 6.8- difluor-1,4-dihydro-4-oxo-3-quinolincarboxylsyre, smp. 250-251°C.

Efterfølgende hydrolyse med saltsyre gav l-ethenyl-6,7,8-trifluor-1,4-dihydro-4-oxo-3-quinolincarboxylsyre, smp. 186-187°C.
10 7-(Bromacetyl)-l-ethyl-6,8-difluor-1,4-dihydro-4-oxo-3-quinolincarboxylsyre
Til 1,12 g (3,80 mmol) 7-acetyl-l-ethyl-6,8-difluor-1 ,4-dihydro-4-oxo-3-quinolincarboxylsyre i 30 ml ed-15 dikesyre blev der sat 0,18 g kaliumbromat og 1,48 ml 48% hydrogenbromidsyre. Blandingen blev omrørt ved 50°C i 24 timer. Blandingen blev koncentreret til det halve volumen, og 20 ml vand blev tilsat. De faste stoffer blev frafiltreret, hvorved vandtes 1,3 g 7-bromacetyl-l-ethyl-20 6,8-difluor-1,4-dihydro-4-oxo-3-quinolincarboxylsyre, smp. 213-215°C.

7-Acetyl-6,8-difluor-1-(2-fluorethyl)-1,4-dihydro-4-oxo-30 3-quinolincarboxylsyre
Til 1,62 g (34,0 mmol) natriumhydrid (50% dispersion i olie, pentan-vasket) i 250 ml dimethylformamid blev der ved stuetemperatur sat 10,0 g (34 mmol) ethyl-7-acetyl-6,8-difluor-1,4-dihydro-4-oxo-3-quinolincarbo-35 xylat opløst i 100 ml dimethylformamid. Blandingen blev omrørt i 2 timer, og 13,0 g (2,4 ækv.) 1-brom-2-fluorethan blev tilsat. Blandingen blev omrørt natten over ved 5Q°C. Den blev koncentreret og fordelt mellem vand 28 DK 170471 B1 og dichlormethan. Dichlormethanet blev derefter koncentreret, og inddampningsresten blev renset ved søjlechro-matografi, hvorved vandtes 3,75 g ethyl-7-acetyl-6,8-di-fluor-1-(2-fluorethyl)-1,4-dihydro-4-oxo-3-quinolincar-5 boxylat, smp. 155-156°C. Dette materiale blev hydrolyseret med 2N saltsyre og 2-propanol som co-opløsningsmid- * del, hvorved vandtes 2,95 g 7-acetyl-6,8-difluor-l-(2-fluorethyl)-1,4-dihydro-4-oxo-3-quinolincarboxylsyre, smp. 215-216°C.

Til 35,45 g (0,230 mol) 2-hydroxymethyl-2-methyl- l,3-dioxolan,0,78 H20 i 300 ml tetrahydrofuran ved -78°C 25 blev der sat 100 ml 2,3M n-butyllithium. Opløsningen blev opvarmet til -40°C og sat til 46,35 g (0,230 mol) 2,6-difluor-3-nitro-acetophenon i 200 ml tetrahydrofuran ved 0°C. Reaktionsblandingen blev omrørt 30 minutter og derefter hældt i 1000 ml af en ethylacetat/mættet ammo-20 niumchlorid-opløsning (1:1). Opløsningen blev filtreret gennem Celite. Lagene blev adskilt, og det vandige lag blev ekstraheret med 3 x 500 ml ethylacetat. De samlede organiske ekstrakter blev vasket med brine, tørret over magnesiumsulfat, og opløsningsmidlet blev fjernet ved 25 reduceret tryk. Produktet blev chromatograferet på sili- 29 DK 170471 B1 ca med hexan:ether (4:1), hvorved vandtes 41,2 g l-[6-fluor-2-[2-(2-methyl-l,3-dioxalan-2-yl)propoxy]-3-nitro-pheny1]ethanon.
1- (2-Acetyl-3-ethanon-l-(2-acetyl-3-fluor-6-nitrophenox\)-5 2-propanon 2,2 g (7,35 mmol) l-[6-Fluor-2-[2-(2-methyl-l,3-dioxalan-2-yl)propoxy]-3-nitrophenyl]ethanon og 360 ml vand .‘saltsyre: eddikesyre (100:10:250) blev omrørt natten over. Opløsningsmidlerne blev fjernet ved reduceret tryk, 10 inddampningsresten blev optaget i dichlormethan og vasket gentagne gange med vand. Det organiske lag blev tørret over magnesiumsulfat, og opløsningsmidlet blev fjernet ved reduceret tryk. En blanding af 4,98 g (19,5 mmol) 1-(2-acetyl-3-20 fluor-6-nitrophenoxy-2-propanon, 100 ml 95% ethanol og 1 g Raney-nikkel blev rystet i en hydrogenatmosfære ved 5 4,5 x 10 Pa ved stuetemperatur i 18 timer. Reaktionsblandingen blev filtreret, og opløsningsmidlet blev fjernet ved reduceret tryk. Inddampningsresten blev chroma-25 tograferet på silica med hexan:ether (1:1), hvorved vandtes 2,76 g 1-(7-fluor-3,4-dihydro-3-methyl-2H-l,4-benzoxazin-8-yl)ethanon.
4-[(8-Acetyl-7-fluor-2,3-dihydro-2-methyl-4H-l,4-benzo-30 xazin-4-yl)methylen]-2,2-dimethyl-l,3-dioxan-4,6-dion
En blanding af 3,66 g (17,49 mmol) 1-(7-fluor-3,4-dihydro-3-methyl-2H-l,4-benzoxazin-8-yl)ethanon, 100 ml methanol og 3,91 g (21,0 mmol) 4-(methoxymethyl'en)-2,2-dimethyl-1,3-dioxan-4,6-dion blev omrørt ved stuetempe-35 ratur natten over. Reaktionsblandingen blev filtreret, og opløsningsmidlet blev fjernet ved reduceret tryk.

Reaktionsblandingen blev afkølet og hældt i is, krystallerne blev frafiltreret og tritureret med ethylether, hvorved vandtes 3,45 g 10-acetyl-9-fluor-2,3-dihydro-3-methyl-7-15 oxo-7H-pyrido[1,2,3-de]-1,4-benzoxazin-6-carboxylsyre, smp. 258-259°C.
8-Chlor-1-cyclopropy1-6,7-difluor-1,4-dihydro-4-oxo-3-20 guinolincarboxylsyre 3-Chlor-2,4,5-trifluorbenzoesvre - En stadig strøm af chlorgas blev boblet gennem en opløsning af 8,1 g (46 mmol) 2,4,5-trifluorbenzosyre 25 (*Jpn. Kokai Tokkyo Koho JP 58, 150, 543 Sept. 7, 1983), 0,2 g (katalytisk mængde) af iod og 15 ml chlorsulfonsyre ved 60°C i fire timer. Reaktionsblandingen blev afkølet til stuetemperatur og forsigtigt hældt på is under konstant omrøring. Suspensionen blev ekstraheret med ethyl-30 acetat, og ekstrakterne blev samlet, vasket med vand og tørret over magnesiumsulfat. Opløsningsmidlet blev fordampet, hvorved vandtes 9,4 g af titelforbindelsen som et orangefarvet fast stof.
35 3-Chlor-2,4,5-trifluorbenzoylchlorid
En opløsning af 9,4 g (45 mmol) 3-chlor-2,4,5-trifluorbenzoesyre, oxalylchlorid (7,6 g, 60 mmol), 3 dråber dimethylformamid og 100 ml dichlormethan blev om- 31 DK 170471 B1 røret ved stuetemperatur i tre timer. Opløsningsmidlet og overskud af oxalylchlorid blev fordampet på dampbad, og inddampningsresten blev destilleret ved 121—123 eC ved 30 mm Hg, hvorved vandtes 2,9 g af titelforbindelsen.

Ethyl- (3-chlor-2,4,5-trif luor) -3-oxophenyl-propionat
Til en blanding af 2,64 g (20 mmol) malonsyre-monoethylester, 0,10 g (katalytisk mængde) af dipyridyl og 50 ml tørt tetrahydrofuran ved - 30°C blev der hurtigt 10 sat 13 ml n-butyllithium (1,55M, 20,2 mmol). Efter endt tilsætning blev opløsningen opvarmet til -5°C, og yderligere 13 ml n-butyllithium (1,55M, 20,2 mmol) blev tilsat dråbevis, indtil en bleg lyserød farve holdt sig i ti minutter. Suspensionen blev derefter afkølet til 15 -78eC. Til denne blanding blev der sat en opløsning af 2,3 g (10 mmol) 3-chlor-2,4,5-trifluorbenzoylchlorid i 3 ml tørt tetrahydrofuran. Reaktionsblandingen blev omrørt ved -78eC i én time, derefter opvarmet til -35°C og hældt i en blanding af is (50 g) og 6N saltsyre (4 ml).
^ Denne opløsning blev ekstraheret med dichlormethan, og ekstrakterne blev vasket med vand, 5% vandig natriumbi-carbonat, fortyndet saltsyre og vand. Reaktionsblandingen blev afkølet til stuetemperatur og koncentreret, hvorved vandtes 2,7 g af titelforbindelsen som en rød olie.

Til en opløsning af ethyl-2-(3-chlor-2,4,5-tri-fluorbenzoyl)-3-cyclopropylaminoacrylat i 40 ml tørt t-butanol blev der sat en opslæmning af 1,20 g (10,7 mmol) kalium-t-butoxid i 20 ml tørt t-butanol. Reaktionsblan-15 dingen blev omrørt ved 6°C i otte timer, derefter afkølet til stuetemperatur og koncentreret. Inddampningsre-sten blev opløst i chloroform, vasket med vand, tørret over magnesiumsulfat, filtreret og koncentreret, hvorved vandtes et blegbrunt fast stof. Råproduktet blev renset • 20 ved silicagel-chromatografi, idet der blev elueret med 80/20 chloroform/ethylacetat, hvorved vandtes titelforbindelsen (1,2 g).
8-ChlOr-1-cvclopropyl-6,7-difluor-1,4-dihydro-4-oxo-3-25 quinolincarboxylsyre
