Abstract
Un procedimiento para detectar la presencia de una molécula de ácido nucleico diana que comprende: a) formar una estructura de escisión entre un ácido nucleico diana, un primer oligonucleótido y un segundo oligonucleótido, en la que el ácido nucleico diana comprende una primera región, una segunda región y una tercera región, estando dicha primera región secuencia abajo de y contigua a dicha segunda región, y estando dicha segunda región secuencia abajo de y contigua a dicha tercera región, en la que dicho primer oligonucleótido comprende una parte 3'' y una parte 5'', conteniendo dicha parte 3'' de dicho primer oligonucleótido una secuencia de uno o más nucleótidos que es completamente complementaria a dicha tercera región de dicho ácido nucleico diana, y conteniendo dicha parte 5'' de dicho primer oligonucleótido una secuencia de uno o más nucleótidos adyacente a dicha parte 3'' de dicho primer oligonucleótido que es completamente complementaria a dicha segunda región de dicho ácido nucleico diana, en la que dicho segundo oligonucleótido comprende una parte 3'' y una parte 5'', conteniendo dicha parte 5'' una secuencia de uno o más nucleótidos que es completamente complementaria a dicha primera región de dicho ácido nucleico diana, y en la que dicha parte 3'' de dicho segundo oligonucleótido es complementaria a dicha segunda región de dicho ácido nucleico diana o comprende o está constituida por al menos un solo nucleótido que está apareado erróneamente a dicha segunda región de dicho ácido nucleico diana, siendo dicha estructura de escisión formada cuando dicho primer oligonucleótido se aparea a dicha tercera y dicha segunda región de dicho ácido nucleico diana, y en la que dicho segundo oligonucleótido está apareado a dicha primera región de dicho ácido nucleico diana tal que la parte 3'' de dicho segundo oligonucleótido se solapa con la secuencia de nucleótidos de dicha parte 5'' de dicho primer oligonucleótido apareado a dicha segunda región de dicho ácido nucleico diana; b) escindir dicha estructura de escisión con una enzima termoestable que tiene actividad nucleasa 5'' y bien carece de actividad sintética o comprende actividad sintética reducida en comparación con una ADN polimerasa nativa para generar un producto de escisión no diana, y c) detectar la escisión de dicha estructura de escisión para detectar la presencia de la molécula de ácido nucleico diana.
