Abstract
Un procedimienrto para la resolución de una mezcla enantiomérica de éster metílico de ácidos 1-amino-2-vinilcilciclopropil-carboxílicos, o una mezcla N-protegida de los mismos, que comprende la etapa de tratar dicha mezclacon una esterasa para obtener un enantiómero deseado de éster metílico de ácidos 1-amino-2-vinilcilciclopropilcarboxílicoso la forma N-protegida del mismo.

Tripéptidos inhibidores de la hepatitis C.
El presente invento se refiere a un procedimiento para la resolución de una mezcla enantiomérica de derivados de ácidos 5 1-aminocilciclopropil-carboxílicos según se define en las reivindicaciones.
Jimenez et al describen en Tetrahedron Letters 35/32: 5945-5948 (1994) una vía de síntesis estereo-controlada para obtener derivados de ácidos 1-aminociclopropano-1-carboxílicos purso en cuanto a los enantiómeros.
10 Fliche et al describen en Synthetic Communications 24 (20): 2873-2876 (1994) la síntesis de ácidos (1R,2S) y (1S,2S) dehidrocoronámicos utilizando sucesivamente dos esterasas con diferentes regioselectividades y una química convencional.
Yee et al describen en J Org. Chem 57: 3525-3527 (1992) un método para resolver enzimáticamente ésteres de ácidos 15 carboxílicos a-sustituidos.
Spero y Kaqpadia describen en J Org. Chem 61: 7398-7401 (1996) una síntesis enantioselectiva de derivados de aminoácidos a,a-sustituidos a través de resolución enzimática.

Con respecto a los ésteres antes descritos, a menos que se especifique otra cosa distinta, cualquier resto alquilo presente contiene ventajosamente de 1 a 16 átomos de carbono, particularmente de 1 a 6 átomos de carbono. Cualquier resto arilo presente en tales ésteres constituye ventajosamente un grupo fenilo.
En particular, los ésteres pueden ser un éster de alquilo C1-16, un éster de bencilo sin sustituir o un éster de bencilo sustituido con al menos un halógeno, alquilo C1-6, alcoxi C1-6, nitro o trifluorometilo.

5 a) Dicho brevemente, el malonato di-protegido VIa y el 1,2-dihalo-alcano VIb o el sulfato cíclico VIc (sintetizado de acuerdo con K. Burgess y Chun-Yen KE (Synthesis, (1996), 1463-1467) se hacen reaccionar en condiciones básicas para dar el diéster VId.
b) Una hidrólisis regio-selectiva del éster menos impedido estéricamente se realiza para dar el ácido VIe.
10 c) Este ácido VIe es sometido a una transposición de Curtius para dar una mezcla racémica de derivados de ácido 1amino-ciclopropilcarboxílico VIf, estando R1 en "sin" con respecto al grupo carboxilo. Una realización específica de esta síntesis se presenta en el Ejemplo 9.
15 d, e) Alternativamente, la formación selectiva de los ésteres a partir del ácido VIe con un apropiado haluro (P*Cl) o alcohol (P*OH) forma el diéster VIg en que el P* éster es compatible con la hidrólisis selectiva del P éster. La hidrólisis del P éster proporciona el ácido VIh.
f) Una transposición de Curtius en VIh da una mezcla racémica de derivados de ácidos 1-amino-ciclopropil-carboxílicos VIi, estando el grupo R1 en "anti" con respecto al grupo carboxilo. Una realización específica para esta síntesis se presenta en el Ejemplo 14.
Una síntesis alternativa para la preparación de derivados VIIf (cuando R1 es vinilo y está en "sin" con respecto al grupo carboxilo) se describe seguidamente.
ESQUEMA VII
10 El tratamiento de iminas VIIa comercialmente disponibles o fácilmente obtenibles con 1,4-dihalo-buteno VIIb en presencia de una base produce, después de hidrólisis de la imina resultante VIIc, el compuesto VIId que tiene el sustituyente alilo en "sin" con respecto al grupo carboxilo. A) Síntesis de una mezcla de (1R,2R)/(1S,2S)-ácidos 1-amino-2-etil-ciclopropil-carboxílicos
a) A una suspensión de cloruro de bencil-trietil-amonio (21,0 g, 92,19 mmol) en una solución acuosa al 50% de NaOH (92,4 g en 185 mL de H2O) se le añadieron consecutivamente malonato de di-terc.-butilo (20,0 g, 92,47 mmol) y 1,2dibromo-butano (30,0 g, 138,93 mmol). La mezcla de reacción se agitó enérgicamente durante una noche a TA, luego se añadió una mezcla de hielo y agua. El producto bruto se extrajo con CH2Cl2 (3x) y se lavó consecutivamente con agua
10 (3x) y salmuera. La capa orgánica se secó (sobre MgSO4), se filtró y se concentró. El residuo se cromatografió con resolución rápida (7 cm, 2 a 4% de Et2O en hexano) para proporcionar el deseado derivado de ciclopropano 9c (19,1 g, 70,7 mmol, rendimiento 76%). 1H NMR (CDCl3): 5 1,78-1,70 (m, 1H), 1,47 (s, 9H), 1,46 (s, 9H), 1,44-1,39 (m, 1H), 1,261,64 (m, 3H), 1,02 (t, 3H, J= 7,6 Hz).
15 b) A una suspensión de terc.-butóxido de potasio (6,71 g, 59,79 mmol, 4,4 eq.) en éter seco (100 ml) a 0ºC se le añadió H2O (270 IL, 15,0 mmol, 1,1 eq.). Después de 5 min, se añadió a la suspensión el diéster 9c (3,675 g, 13,59 mmol) en éter (10 mL). La mezcla de reacción se agitó durante una noche a TA, luego se vertió en una mezcla de hielo y agua y se lavó con éter (3x). La capa acuosa se acidificó con una solución acuosa al 10% de ácido cítrico a 0ºC y se extrajo con AcOEt (3x). La capa orgánica combinada se lavó consecutivamente con agua (2x) y salmuera. Después del tratamiento
20 usual (con Na2SO4, filtración y concentración), el deseado ácido 9d se aisló como un aceite de color amarillo pálido (1,86 g, 8,68 mmol, rendimiento 64%). 1H NMR (CDCl3): 5 2,09-2,01 (m, 1H), 1,98 (dd, J = 3,8, 9,2 Hz, 1H), 1,81-1,70 (m, 1H), 1,66 (dd, J = 3,0, J = 8,2 Hz, 1H), 1,63-1,56 (m, 1H), 1,51 (s, 9H), 1,0 (t, J= 7,3 Hz, 3H).
c) Al ácido 9d (2,017 g, 9,414 mmol) en benceno seco (32 mL) se le añadieron consecutivamente Et3N (1,50 mL, 10,76

EJEMPLO 10
Resolución química de (1R,2R)/(1S,2S)-1-amino-2-etil-ciclopropil-carboxilatos de t-butilo (procedentes del Ejemplo 9):
El compuesto 9e procedente del Ejemplo 9 (8,50 g, 25,86 mmol) se trató con TBAF/THF 1 M (26 mL) a reflujo durante 45
10 min. La mezcla de reacción enfriada se diluyó con EtOAc, se lavó con agua (3x) y con salmuera (1x), luego se secó (sobre MgSO4), se filtró y se evaporó para proporcionar la amina libre como un aceite de color amarillo claro. La amina libre se disolvió en CH2Cl2 anhidro (120 mL), NMM (8,5 mL, 77,57 mmol), se añadieron consecutivamente el compuesto 4 (Ejemplo 4) (10,08 g, 27,15 mmol) y HATU (11,79 g, 31,03 mmol). La mezcla de reacción se agitó a TA durante una noche, luego se trató tal como se ha descrito con anterioridad. La mezcla diastereoisómera bruta se separó por
15 cromatografía con resolución rápida (eluyente - mezcla de hexano y Et2O; 25:75), para proporcionar los dipéptidos 10a (la mancha de elución menos polar) como una espuma de color blanco (4,42 g; 64% del rendimiento teórico) y 10b (la mancha de elución más polar) como una espuma de color marfil (4 g, 57% del rendimiento teórico). En este momento, ambos isómeros se habían separado, pero todavía no se conocía la estereoquímica absoluta.
Determinación de la estereoquímica absoluta de los compuestos 10a y 10b por correlación con el (1R-amino-2Retil-ciclopropil)-carboxilato de t-butilo conocido
El Profesor A. Charette, de la Universidad de Montreal, proporcionó el compuesto 11a que tenía la estereoquímica absoluta que se muestra, la cual fue determinada por cristalografía de rayos X (J. Am. Chem. Soc., 1995, 117, 12721). El compuesto 11a (13,2 mg, 0,046 mmol) se disolvió en una mezcla de HCl 1M y EtOAc (240 IL) y se agitó durante aproximadamente 48 horas. La mezcla se evaporó hasta sequedad para proporcionar el compuesto 11b como una pasta 5 de color amarillo claro y se acopló al compuesto 4 (18 mg, 0,049 mmol) como se describe en el Ejemplo 10, utilizando NMM (20,3 IL, 0,185 mmol) y HATU (21,1 mg, 0,056 mmol) en CH2Cl2. El material bruto se purificó por cromatografía con resolución rápida (eluyente - mezcla de hexano y Et2O; 50:50) para proporcionar el dipéptido 11c en forma de un aceite (7,7 mg; 31%). EJEMPLO 12
El carbamato 9e procedente del Ejemplo 9 (2,6 g, 7,88 mmol) se agitó durante 40 min en TFA a 0ºC. Luego la mezcla se concentró y diluyó con THF (10 mL). Se añadió una solución acuosa de NaOH (700 mg, 17,5 mmol en 8,8 mL de H2O) seguido por una solución en THF (13 mL) de (Boc)2O (2,06 g, 9,44 mmol, 1,2 eq.). La mezcla de reacción se agitó 20 durante una noche a TA (el pH se mantuvo en 8 añadiendo una solución acuosa al 10% de NaOH cuando se necesitaba), luego se diluyó con H2O, se lavó con Et2O (3x) y se acidificó a 0ºC con una solución acuosa al 10% de ácido cítrico. La capa acuosa se extrajo con EtOAc (3x) y se lavó consecutivamente con H2O (2x) y con salmuera. Después del tratamiento usual (con MgSO4, filtración y concentración) se aisló el deseado aminoácido protegido por Boc (12a) (788 mg, 3,44 mmol, rendimiento 44%). 1H NMR (CDCl3): 5 5,18 (bs, 2H), 1,64-1,58 (m, 2H), 1,55-1,42 (m, 2H), 1,45 (s, 9H),

30 El derivado con Boc 12a (0,30 g, 1,31 mmol) se disolvió en Et2O (10 mL) y se trató con diazometano recientemente preparado en Et2O a 0ºC hasta que persistiese el color amarillo de un ligero exceso de diazometano. a) La mezcla de enantiómeros (1S,2S)/(1R,2R)-éster metílico de ácido 1-Boc-amino-2-etil-carboxílico del Ejemplo 10 (0,31 g, 1,27 mmol) se disolvió en acetona (3 mL) y luego se diluyó con agua (7 mL) mientras que se estaba agitando rápidamente. El pH de la solución se ajustó a 7,5 con NaOH acuoso 0,05 M antes de que se añadiese Alcalase®
10 [extracto de 2,4 L procedente de Novo Nordisk Industrials] (300 mg). Después del tratamiento usual (con MgSO4, filtración y concentración) el éster 14a deseado se aisló (1,106 g, 3,35 mmol, rendimiento 91%) como un aceite incoloro.

La mezcla de reacción se agitó durante una noche a TA, se puso a reflujo durante 30 min y luego se diluyó con AcOEt. La solución se lavó consecutivamente con agua (2x) y con salmuera. EJEMPLO 15
a) A una solución en THF (180 mL) de terc.-butóxido de potasio (4,62 g, 41,17 mmol, 1,1 eq.) a -78ºC se le añadió la imina 15a comercialmente disponible (10,0 g, 37,41 mmol) en THF (45 mL). La mezcla de reacción se calentó a 0ºC y se
40 agitó a esta temperatura durante 40 min. Luego la mezcla se enfrió de vuelta a -78ºC para la adición de 1,4-dibromobuteno 15b (8,0 g, 37,40 mmol) y luego se agitó a 0ºC durante 1 h y se enfrió de retorno a -78ºC para la adición de terc.butóxido de potasio (4,62 g, 41,17 mmol, 1,1 eq.). La mezcla de reacción se agitó finalmente durante una hora más a 0ºC y se concentró para proporcionar el compuesto 15c.
45 b, c, d) El compuesto 15c se recogió en Et2O (265 mL) y se trató con una solución acuosa 1 N de HCl (106 mL). Después de 3,5 h a TA, las capas fueron separadas y la capa acuosa fue lavada con Et2O (2x) y alcalinizada con una solución acuosa saturada de NaHCO3. EJEMPLO 16
Preparación de (1R,2S/1S,2R)-éster etílico de ácido 1-Boc-amino-2-vinil-ciclopropil-carboxílico:
10 La sal hidrocloruro 15d (1,0 g, 5,2 mmol) y (Boc)2O (1,2 g, 5,7 mmol) se disolvieron en THF (30 mL) y se trataron con DMAP (0,13 g, 1,04 mmol, 0,2 eq.) y diisopropil-etilamina (2,8 mL, 15,6 mmol). La mezcla de reacción se agitó durante 24 h antes de ser diluida con EtOAc (40 mL) y se lavó consecutivamente con una solución (acuosa) saturada de NaHCO3, con HCl acuoso al 5% y con salmuera saturada. La fase orgánica se secó (sobre MgSO4), se filtró y se concentró para

Resolución enzimática de (1R,2S)/(1S,2R) 1-amino-2-vinil-ciclopropil-carboxilato de etilo:
a) El derivado racémico 17a (0,29 g, 1,14 mmol) se disolvió en acetona (5 mL) y se diluyó con H2O (10 mL). El pH se ajustó con NaOH acuoso 0,2 N a 7,2 antes de que se añadiese Alcalase® (300 mg). Para mantener constante el pH durante la incubación, se añadió una solución de NaOH mediante un valorador con pH-estato durante 9 días hasta que se hubiera añadido la cantidad teórica de la base. Después de extracción con un ácido y con una base como se describe
30 en el Ejemplo 13, se aislaron el éster sin hidrolizar (0,15 g, 100%) y el material hidrolizado (0,139 g, 95%). El análisis del éster sin hidrolizar por HPLC utilizando una columna quiral, puso de manifiesto una relación de 43:1 del compuesto 17c deseado al que se asignó la estereoquímica (R,S) basándose en la correlación química, como se describe en los Ejemplos 10 y 11. Resolución de (1R,2S)/(1S,2R)-1-amino-2-vinil-ciclopropil-carboxilato por cristalización con ácido dibenzoil-Dtartárico
A una solución de una mezcla racémica bruta de (1S,2R y 1R,2S)-1-amino-2-vinil-ciclopropil-carboxilatos de etilo [obtenida a partir del éster etílico de N-(difenilmetilen)-glicina (25,0 g, 93,5 mol) como se describe en el Ejemplo 15] en 10 EtOAc (800 mL) se le añadió ácido dibenzoil-D-tartárico (33,5 g, 93,5 mol). La mezcla se calentó a reflujo, se dejó a TA durante 15 min y luego se enfrió a 0ºC. Se obtuvo un sólido de color blanco después de 30 min. El sólido se filtró, se lavó con EtOAc (100 mL) y se secó en aire. El sólido se suspendió en acetona (70 mL), se sonicó y se filtró (3x). Luego el sólido se recristalizó dos veces en acetona caliente (tanda cosechada A). Las aguas madres se concentraron y el residuo se recristalizó tres veces en acetona caliente (tanda cosechada B). Las dos tandas cosechadas de los sólidos 15 amorfos de color blanco de sal de ácido dibenzoil-D-tartárico se combinaron (5,53 g) y suspendieron en una mezcla de Et2O (250 mL) y solución saturada de NaHCO3 (150 mL). Preparación de (1R,2S)/(1S,2R)-hidrocloruro de éster metílico de ácido 1-amino-2-vinil-ciclopropano-carboxílico
Preparación de la imina 19b El hidrocloruro del éster etílico de glicina 19a (1519,2 g, 10,88 mol, 10 eq.) se suspendió en terc.-butil-metil-éter (8 L). Se añadieron benzaldehído (1115 g, 10,88 mol, 1 eq.) y sulfato de sodio anhidro (773 g, 5,44 mol, 0,5 eq.) y la mezcla se
35 enfrió a 5ºC en un baño de hielo y agua. Se añadió gota a gota trietilamina (2275 mL, 16,32 mol, 1,5 eq.) en el transcurso de 15 min (con uso de 0,5 L de terc.-butil-metil-éter para enjuagados) y la mezcla se agitó durante 40 h a la temperatura ambiente. Luego la mezcla de reacción se sofocó por adición de agua enfriada con hielo (5 L) y la capa orgánica se separó. La fase acuosa se extrajo con terc.-butil-metil-éter (1 L) y las fases orgánicas combinadas se lavaron con una mezcla de NaHCO3 saturado (400 mL) y agua (1,6 L) y luego con salmuera. La solución se secó sobre MgSO4, se
40 concentró bajo presión reducida y el aceite residual de color amarillo se secó hasta peso constante en vacío. Preparación de una mezcla racémica de N-Boc-(1R,2S)/(1S,2R)-hidrocloruros de ésteres etílicos de ácidos 1-amino-2
vinil-ciclopropano-carboxílicos 19e:
Se suspendió terc.-butóxido de litio (4,203 g, 52,5 mmol, 2,1 equiv.) en tolueno seco (60 mL). La imina 19b (5,020 g, 26,3
5 mmol, 1,05 equiv.) y el dibromuro 19c (5,348 g, 25 mmol, 1 equiv.) se disolvieron en tolueno seco (30 mL) y esta solución se añadió gota a gota durante 30 min a la solución agitada de LiOtBu a la temperatura ambiente. Después de compleción, la mezcla de color rojo oscuro se agitó durante 10 min adicionales y se sofocó por adición de agua (50 mL) y terc.-butil-metil-éter (TBME, 50 mL). La fase acuosa se separó y se extrajo una segunda vez con TBME (50 mL). Las fases orgánicas se combinaron, se añadió HCl 1 N (60 mL) y la mezcla se agitó a la temperatura ambiente durante 2 h.

Preparación del compuesto del título 19f por medio de trans-esterificación del compuesto 19e:
25 El éster etílico 19e (10,807 g, 42,35 mmol) se disolvió en metanol seco (50 mL) y se añadió una solución de metóxido de sodio en MeOH (al 25% p/p, 9,7 mL, 42 mmol, 1 equivalente). La mezcla se calentó a 50ºC durante 2 h, en cuyo punto el análisis por TLC indicó una transesterificación completa (19e Rf 0,38, 19f Rf 0,34 en una mezcla 20% de EtOAc y hexano). La mezcla de reacción se enfrió a la temperatura ambiente y se acidificó a pH 4 utilizando HCl 4 N en dioxano.
30 El NaCl precipitado se eliminó por filtración (con uso de terc.-butil-metil-éter para los lavados) y los materiales volátiles se eliminaron bajo presión reducida. Se añadió terc.-butil-metil-éter (100 mL) al residuo y los sólidos se eliminaron por filtración. EJEMPLO 20
Resolución enzimática del (1R,2S)/(1S/2R)-hidrocloruro de éster metílico de ácido 1-amino-2-vinil-ciclopropano40 carboxílico
(1R,2S)
Preparación de N-Boc-(1R,2S)-éster metílico de ácido 1-amino-2-vinil-ciclopropano-carboxílico 20a:
45 El éster racémico 19f (0,200 g, 0,83 mmol) se disolvió en acetona (3 mL) y se añadió agua (7 mL). Se añadió NaOH 0,05 M (1 gota) para llevar el pH de la solución a ~8 y luego se añadió Alcalase® 2.4L (Novo Nordisk Biochem, 0,3 g en un mL de agua). La mezcla se agitó enérgicamente a la temperatura ambiente, manteniéndose el pH de la solución en 8 utilizando un valorador automático. Al comienzo de los días 4º y 5º de agitar a pH 8, se añadió una solución adicional de enzima (2 x 0,3 g). Después de un total de 5 días, se consumió un total de 8,3 mL de NaOH 0,05 M. La mezcla de reacción se diluyó con EtOAc y agua y la fase orgánica se separó. Después de haber lavado con salmuera, el extracto orgánico se secó (sobre MgSO4) y se concentró en vacío. El compuesto 20a (0,059 g, rendimiento 30%) se obtuvo como un aceite transparente: la 1H NMR era idéntica a la del compuesto 19f. HPLC (Chiralcel ODH, 4,6 x 250 mm, mezcla
5 isocrática al 1% de EtOH en hexano, caudal 0,8 mL/min):(1R,2S)-2 Rt 19,3 min (97%); (1S,2R)-2 Rt 17,0 min (3%).
Preparación del (1R,2S)-hidrocloruro de éster metílico de ácido 1-amino-2-vinil-ciclopropano-carboxílico 20b:
El compuesto 20a (39,96 g, 165,7 mmol) se disolvió en dioxano (25 mL) y la solución se añadió gota a gota con agitación
10 a HCl 4 N en dioxano (Aldrich, 250 mL). Después de 45 min, el análisis por TLC indicó una desprotección completa. Los materiales volátiles se eliminaron bajo presión reducida y el residuo se evaporó conjuntamente dos veces con MeOH (2 x 100 mL). Se añadieron éter (300 mL) y MeOH (10 mL) al residuo oleoso de color pardo y la mezcla se agitó durante una noche a la temperatura ambiente, dando como resultado la precipitación de un material semi-sólido. Se añadió una cantidad adicional de MeOH (15 mL) y se continuó la agitación durante 6 h, en cuyo punto se recogió un sólido de color

El producto se lavó con 5% de MeOH en éter (50 mL) y con éter (2 x 50 mL), y se secó en vacío para dar el compuesto 20b como un sólido de color amarillento (22,60 g, rendimiento 76%). Los materiales filtrados (inclusive los líquidos de lavado) se evaporaron en vacío para dar una cantidad adicional del compuesto 20b como un aceite de color pardo (7,82 g, rendimiento 26%). Claims (7)
- REIVINDICACIONES1. Un procedimienrto para la resolución de una mezcla enantiomérica de éster metílico de ácidos 1-amino-2vinilcilciclopropil-carboxílicos, o una mezcla N-protegida de los mismos, que comprende la etapa de tratar dicha mezcla5 con una esterasa para obtener un enantiómero deseado de éster metílico de ácidos 1-amino-2-vinilcilciclopropilcarboxílicos o la forma N-protegida del mismo.
- 2. El procedimiento de acuerdo con la reivindicación 1, en el que dicha esterasa es Alcalase®.10 3. Un procedimiento para la resolución de una mezcla enantiomérica de (1R,2R)/(1S,2S)-Boc-1-amino-2etilciclopropil-carboxilato de metilo que tiene la fórmula:que comprende tratar la mezcla con Alcalase® para proporcionar productos de las fórmulas:
