Claims (1)
- 【特許請求の範囲】 1.インフルエンザウイルスによって感染され得、そして無血清培地の懸濁物に おける増殖に適合する、動物細胞。 2.前記細胞が脊椎動物起源である、請求項1に記載の細胞。 3.前記細胞が哺乳動物起源である、請求項2に記載の細胞。 4.前記細胞が腎臓細胞由来である、請求項1〜3のいずれか1つに記載の細胞 。 5.前記細胞がMDCK細胞(ATCC CCL34 MDCK(NBL-2))由来である、請求項4に記 載の細胞。 6.前記細胞が細胞株MDCK33016(DSM ACC2219)である、請求項5に記載の細胞 。 7.細胞培養におけるインフルエンザウイルスの複製のための方法であって、 (i)懸濁物中の無血清培地において、請求項1〜6のいずれか1つに記載の細 胞を増殖させる工程; (ii)該細胞をインフルエンザウイルスに感染させる工程; (iii)感染直前、感染と同時、または感染直後に細胞懸濁物にプロテアーゼを 添加し、赤血球凝集素[HA0]の前駆体タンパク質を切断する工程;および (iv)さらなる培養期の後、該細胞中で複製した該インフルエンザウイルスを単 離する工程; を含む、方法。 8.前記細胞の培養が灌流系において行われる、請求項7に記載の方法。 9.前記細胞の培養がバッチ法において行われる、請求項7に記載の方法。 10.工程(i)の培養培地のpHが6.6〜7.8の範囲である、請求項7〜9のいず れか1つに記載の方法。 11.前記培養培地のpHが6.8〜7.3の範囲である、請求項10に記載の方法。 12.前記インフルエンザウイルスでの感染が、前記細胞培養が約8〜25×105 細胞/ml(バッチ法)または約5〜20×105細胞/ml(灌流法)の細胞密度に達す る場合に生じる、請求項7〜11のいずれか1つに記載の方法。 13.インフルエンザウイルスでの前記細胞の感染が、前記細胞培養が約0.001 〜10のm.o.i.(感染の多重度)で生じる、請求項7〜12のいずれか1つに記載 の方法。 14.前記感染が約0.002〜0.5のm.o.i.で生じる、請求項13に記載の方法。 15.前記プロテアーゼがセリンプロテアーゼである、請求項7〜14のいずれ か1つに記載の方法。 16.前記セリンプロテアーゼがトリプシンである、請求項15に記載の方法。 17.トリプシンが前記培養培地において1〜200μg/mlの最終濃度まで添加され る、請求項16に記載の方法。 18.前記培養培地中のトリプシンの最終濃度が5〜50μg/mlの範囲である、請 求項17に記載の方法。 19.前記感染細胞が2〜10日間培養される、請求項7〜18のいずれか1つに 記載の方法。 20.前記感染細胞が3〜7日間培養される、請求項19に記載の方法。 21.前記感染細胞が30℃〜36℃で培養される、請求項7〜20のいずれか1つ に記載の方法。 22.前記感染細胞が32℃から34℃で培養される、請求項21に記載の方法。 23.前記複製したウイルスの採取および単離が、感染後2〜10日に行われる、 請求項7〜22のいずれか1つに記載の方法。 24.前記複製したウイルスの採取および単離が、感染後3〜7日に行われる、 請求項23に記載の方法。 25.請求項7〜24のいずれか1つに記載の方法によって入手可能な、インフ ルエンザウイルス。 26.適切である場合に免疫応答を増大させる物質と組合せて、請求項25に記 載のインフルエンザウイルスを含む、ワクチン。 27.前記インフルエンザウイルスがインタクトなウイルス粒子として存在する 、請求項26に記載のワクチン。 28.前記インフルエンザウイルスが弱毒化ウイルスとして存在する、請求項2 6に記載のワクチン。 29.前記インフルエンザウイルスが崩壊したウイルス粒子として存在する、請 求項26に記載のワクチン。 30.適切である場合に免疫応答を増大させる物質と組合せて、請求項25に記 載のインフルエンザウイルスの構成成分を含む、ワクチン。 31.前記ワクチンが前記インフルエンザウイルスの単離されたタンパク質を含 む、請求項30に記載のワクチン。 32.請求項25に記載のインフルエンザウイルスまたはそのようなウイルスの 構成成分を含む、診断用組成物。
