Description
Claims (1)
- 【特許請求の範囲】 1. 式I [式中、R1は水素またはメチルを表し、R2およびR3は独立して、水素、メチ ル、メトキシ、ヒドロキシ、カルバモイル、スルファモイルまたはハロゲンを表 し、Xは式II: (式中、R4およびR5は非芳香族炭化水素基を表し、それらは同じでも異なって いてもよく、あわせて少なくとも3個の炭素原子を含み、R4およびR5が窒素原 子とともに環を形成してもよい。)の第三アミノ基を表す。]で表される3,3 −ジフェニルプロピルアミン類、生理学的に許容されうる酸との塩ならびに該化 合物が光学異性体となりうる場合はラセミ混合物および個々のエナンチオマー。 2. R4およびR5の各々が独立して飽和炭化水素基、特に C1〜C8アルキル、特にC1〜C6アルキルなどの飽和脂肪族炭化水素基またはア ダマンチルであり、R4およびR5があわせて少なくとも3個、好ましくは少なく とも4個の炭素原子を含むことを特徴とする請求項1に記載の3,3−ジフェニ ルプロピルアミン類。 3. R4およびR5の少なくとも一方が分岐炭素鎖を含むことを特徴とする請求 項1または2に記載の3,3−ジフェニルプロピルアミン類。 4. Xが下記a)〜h)の基のいずれかを表すことを特徴とする請求項1〜3 のいずれか一項に記載の3,3−ジフェニルプロピルアミン類。 5. HOCH2基が5位にあり、R2が水素であり、R3が水素またはヒドロキ シであって、好ましくは2位にあることを特徴とする請求項1〜4のいずれか一 項に記載の3,3−ジフェニルプロピルアミン類。 6. N,N−ジイソプロピル−3−(2−ヒドロキシ−5−ヒドロキシメチル フェニル)−3−フェニルプロピルアミン、その生理学的に許容されうる酸との 塩、ラセミ体およびその個々のエナンチオマーから選択されることを特徴とする 請求項1に記載の3,3−ジフェニルプロピルアミン類。 7. 薬剤的に活性な物質、特に抗コリン薬剤として使用するための請求項1〜 6のいずれか一項に記載の3,3−ジフェニルプロピルアミン類。 8. 請求項1〜6のいずれか一項に記載の3,3−ジフェニルプロピルアミン および好ましくは混和性のある薬剤担体を含む薬剤組成物。 9. 請求項1〜6のいずれか一項に記載の3,3−ジフェニルプロピルアミン の抗コリン薬剤を作るための使用。 10. a)式III: [式中、R1〜R3およびXは上記で定義した通りであり、R6は水素またはR7O (R7は水素、アルキル、アルケニル、アルキニルまたはアリールである。)で あり、どのヒドロキシ基もメチル化またはベンジル化などにより保護されていて もよい。]の3,3−ジフェニルプロピルアミンのR6COの基を還元するか、 b)式IV: [式中、R1〜R3は上記で定義した通りであり、どのヒドロキシ基も保護されて いてもよく、Yは脱離基である。]の反応性エステル化した3,3−ジフェニル プロパノールを式V: H−X V [式中、Xは上記で定義した通りである。]のアミンと反応させるか、 c)式VI: [式中、R1〜R3およびXは上記で定義した通りであり、どのヒドロキシ基も保 護されていてもよい。]の3,3−ジフェニルプロピオンアミドを還元するか、 d)式VII: [式中、R1〜R3およびXは上記で定義した通りであり、どのヒドロキシ基も保 護されていてもよく、ZはR4およびR5とメチルを除いて同じである。]の二級 3,3−ジフェニルプロピルアミンをN−メチル化するか、 e)式VIIIaの3,3−ジフェニルプロペンアミンまたは式VIIIbの3 ,3−ジフェニルプロピルアミン: [式中、R1〜R3およびXは上記で定義した通りであり、どのヒドロキシ基も保 護されていてもよく、Wはヒドロキシ基またはハロゲン原子を表す。]を還元す るか、 f)式IX: [式中、R1〜R3およびXは上記で定義した通りであり、Halはハロゲンであ る。]のジフェニルプロピルアミンをホルムアルデヒドまたはホルムアルデヒド 等価物と反応させるか、 g)式X: [式中、R1〜R3およびXは上記で定義した通りである。]のジフェニルプロピ ルアミンのメチル基を酸化して、 i)必要であれば、得られた化合物のヒドロキシ保護基を、所 望により一方または両方のフェニル環をモノまたはジハロゲン化した後に脱離し 、および/または ii)所望により、得られた式Iの塩基を生理学的に許容されうる酸によりその塩 に変換するか(逆も可能)、および/または iii)所望により、得られた光学異性体の混合物を個々のエナンチオマーに分割 し、および/または iv)所望により、得られた式I(R1は水素であり、および/またはR3はヒドロ キシである。)の化合物のオルト−ヒドロキシ基をメチル化する 工程を含む、請求項1〜6のいずれか一項に記載の3,3−ジフェニルプロピル アミンの製造法。
