Claims (1)
- 【特許請求の範囲】 1. TRAILポリペプチドをコードしている単離DNAであって、該TR AILポリペプチドが、配列番号:2のアミノ酸1〜281および配列番号:6 のアミノ酸1〜291から成る群より選択されるアミノ酸配列と少なくとも80 %同一であるアミノ酸配列を含み、該TRAILポリペプチドがジャーカット細 胞のアポトーシスを誘導できる上記単離DNA。 2. 前記TRAILポリペプチドが、配列番号:2のアミノ酸1〜281お よび配列番号:6のアミノ酸1〜291から成る群より選択されるアミノ酸配列 を含む請求項1に記載の単離DNA。 3. 可溶性TRAILポリペプチドをコードしている単離DNAであって、 該可溶性TRAILポリペプチドが、 (a)ヒトTRAILの細胞外ドメイン(配列番号:2のアミノ酸39〜28 1);および (b)該細胞外ドメインのフラグメント から成る群より選択される配列と少なくとも80%同一であるアミノ酸配列を含 み; 該可溶性TRAILポリペプチドがジャーカット細胞のアポトーシスを誘導でき る上記単離DNA。 4. 前記可溶性TRAILポリペプチドが、 (a)ヒトTRAILの細胞外ドメイン(配列番号:2のアミノ酸39〜28 1);および (b)該細胞外ドメインのフラグメントであって、ジャーカット細胞のアポト ーシスを誘導できる上記フラグメント から成る群より選択されるアミノ酸配列を含む請求項3に記載のDNA。 5. 前記可溶性TRAILポリペプチドが、配列番号:2のアミノ酸x〜2 81の配列を含み、ここにおいて、xは39〜95の整数である請求項4に記載 のDNA。 6. 前記可溶性TRAILポリペプチドが、配列番号:2のアミノ酸95〜 281の配列を含む請求項5に記載のDNA。 7. 前記可溶性TRAILポリペプチドが、 (a)ヒトTRAILの細胞外ドメイン(配列番号:2のアミノ酸39〜28 1);および (b)該細胞外ドメインのフラグメント から成る群より選択されるアミノ酸配列中に1個または複数の保存的置換を含み ; その保存置換されたTRAILが、ジャーカット細胞のアポトーシスを誘導で きる請求項3に記載のDNA。 8. 請求項1〜7のいずれか1項に記載のDNAを含む発現ベクター。 9. TRAILポリペプチドを製造する方法であって、請求項8に記載のベ クターで形質転換された宿主細胞を、TRAILの発現を促進する条件下で培養 し、そしてTRAILポリペプチドを回収することを含む上記方法。 10.精製TRAILポリペプチドであって、配列番号:2のアミノ酸1〜2 81および配列番号:6のアミノ酸1〜291から成る群より選択されるアミノ 酸配列と少なくとも80%同一であるアミノ酸配列を含み、該TRAILポリペ プチドがジャーカット細胞のアポトーシスを誘導できる上記精製TRAILポリ ペプチド。 11.配列番号:2のアミノ酸1〜281および配列番号:6のアミノ酸1〜 291から成る群より選択されるアミノ酸配列を含む請求項10に記載の精製T RAILポリペプチド。 12.菌株ATCC 69849 で寄託された組換えベクターのcDNAインサー トによってコードされたヒトTRAILポリペプチド。 13.精製可溶性TRAILポリペプチドであって、 (a)ヒトTRAILの細胞外ドメイン(配列番号:2のアミノ酸39〜28 1);および (b)該細胞外ドメインのフラグメント から成る群より選択される配列と少なくとも80%同一であるアミノ酸配列を含 み; 該可溶性ヒトTRAILポリペプチドがジャーカット細胞のアポトーシスを誘導 できる上記精製可溶性TRAILポリペプチド。 14.(a)ヒトTRAILの細胞外ドメイン(配列番号:2のアミノ酸39 〜281);および (b)該細胞外ドメインのフラグメントであって、ジャーカット細胞のアポト ーシスを誘導できる上記フラグメント から成る群より選択されるアミノ酸配列を含む請求項13に記載のTRAILポ リペプチド。 15.配列番号:2のアミノ酸x〜281の配列を含み、ここにおいて、xは 39〜95の整数である請求項14に記載のTRAILポリペプチド。 16.配列番号:2のアミノ酸95〜281を含む請求項15に記載のTRA ILポリペプチド。 17.前記可溶性TRAILポリペプチドが、 (a)ヒトTRAILの細胞外ドメイン(配列番号:2のアミノ酸39〜28 1);および (b)該細胞外ドメインのフラグメント から成る群より選択されるアミノ酸配列中に1個または複数の保存的置換を含み ; その保存置換されたTRAILが、ジャーカット細胞のアポトーシスを誘導で きる請求項13に記載のTRAILポリペプチド。 18.請求項13に記載の可溶性TRAILポリペプチドを2〜3個含むオリ ゴマー。 19.請求項16に記載の可溶性TRAILポリペプチドを3個含むTRAI L三量体。 20.請求項11または15に記載のTRAILタンパク質を特異的に結合す る抗体。 21.前記抗体がモノクローナル抗体である請求項20に記載の抗体。
