Description

【発明の詳細な説明】
本発明は、ここでA/16686抗生物質と称する
抗生物質、分類学的にアクチノプラネス
(Actinoplanes)属の新規な菌株として特徴づけ
られる従来の文献に記載されない菌株を培養する
ことによつてそれを製造する方法、およびその抗
バクテリア剤(antibacterial agent)としての使
用に関する。 抗生物質A/16686は、酸付加塩を形成するこ
とができる塩基性を有するグリコペプチド抗生物
質である。したがつて、抗生物質A/16686の生
理学的に許容しうる酸付加塩は本発明の一部であ
る。 有用性の説明を簡単にするため、「抗生物質
A/16686」という語は、ここでは抗生物質A/
16686の遊離塩基およびその生理学的に許容しう
る酸付加塩から選ばれた抗生物質を呼ぶ。 抗生物質A/16686は、試験管内で病原性バク
テリア、殊にグラム陽性バクテリアの生育を阻止
する。その上、抗生物質A/16686の非経口的投
与は、マウスにおける実験的感染に対して高度の
保護を与える。 前述のように、抗生物質A/16686は、アクチ
ノプラネス(Actinoplanes)属の新規な菌株を培
養することにより製造される。バグハルボツド
(Vaghalbod)(インド)における土壌の試料から
単離したこの菌株の培養物は、ATCC
〔(American Type Culture Collection)―
12301Parklawn Drive、Rockville―
Maryland20852―U.S.A.〕の永久培養物コレクシ
ヨンに1979年1月30日に寄託され、受託番号
ATCC33076が付与された。また、上記の菌株
〔アクチノプラネス・エス・ピー(Actinoplanes
sp.)ATCC33076〕は茨城県筑波郡の工業技術院
微生物工業研究所に、微工研菌寄第5477号
(FERM―PNo.5477)として寄託されている。 アクチノプラネス・エス・ピー(Actinoplanes
sp.)ATCC33076の特性を次に記載する。 形態学 本菌株はオレンジ色の基質菌糸体をもち、種々
の培地上でよく生育する。それは顔料を生成しな
い。気中菌糸体は常に存在しない。顕微鏡検査に
おいて、栄養(vegetative)菌糸体は約1μmの
直径をもつ分枝した菌糸を示す。胞子嚢はジヤガ
イモ寒天上でのみ不十分に形成し、そして非常に
不規則の表面と5.0〜9.0μmの範囲の直径をもつ
球状体をもつ。胞子嚢の放出は胞子嚢壁の破裂後
に観察される。球様胞子は動くことができる。
(1.0〜1.5μmの直径)。細胞壁成分を分析する
と、メソ―ジアミノピメリン酸およびD型の砂糖
パターンが明らかになる(Lechevalier etal.、

)。 培養特性 表に、ShirlingおよびGottlieb(Intern J.
System.Bact.16、313―340、1966)が示唆する
種々の標準培地およびWaksman(The
Actinomycetes、Vol.―The Williams and
Wilkins Co.1961)が推奨する他の培地上で培養
したアクチノプラネス(Actinoplanes)
ATCC33076の培養特性を報告する。培養特性
は、30℃において接種後6〜14日において決定し
た。

Claims (1)

  1. 【特許請求の範囲】 1 塩酸塩の形態で、 (A) 224〜226℃において溶融する白色結晶性物質
    であり; (B) 水およびジメチルホルムアミド中に非常に可
    溶性であり;メタノール、エタノール、プロパ
    ノールおよびブタノール中に可溶性であるが;
    エチルエーテル、石油エーテルおよびベンゼン
    中に不溶性であり; (C) 51.73%の炭素、6.34%の水素、9.96%の窒
    素、5.84%の塩素(合計含量)、4.74%の塩素
    イオン、および1%の残渣の近似元素組成; (D) 次の観測可能な吸収極大を有する赤外吸収ス
    ペクトル(nujol);(添付図面の第1図参
    照):3290―3070、2930および2860(nujol)、
    1765、1630、1510、1455、1375(nujol)、
    1230、1175、1130、1065、1030、1015、980、
    840および820cm-1; (E) 次の吸収極大を有する紫外吸収スペクトル

    らなる群より選ばれる抗生物質。 2 抗生物質A/16686塩酸塩である特許請求の
    範囲第1項記載の抗生物質。 3 アクチノプラネス属に属する抗生物質A/
    16686生産菌を培養し、培養物から抗生物質A/
    16686を回収することを特徴とする抗生物質A/
    16686遊離塩基およびその生理学的に許容しうる
    酸付加塩からなる群より選ばれる抗生物質の製造
    方法。 4 菌株アクチノプラネス・エス・ピー
    (Actionoplanes sp.)ATCC33076を、同化可能
    な炭素源、同化可能な窒素源および無機塩類を含
    有する培地中で、深部好気性条件下に、実質的な
    量の抗生物質A/16686が生成するまで培養し、
    そして該抗生活性物質を通常の方法により回収す
    ることを特徴とする特許請求の範囲第3項記載の
    方法。 5 菌株アクチノプラネス・エス・ピー
    (Actionoplanes sp.)ATCC33076を、同化可能
    な炭素源、同化可能な窒素源および無機塩類を含
    有する培地を、深部好気性条件下に、実質的な量
    の抗生活性物質が生成するまで培養し、液体培地
    および菌糸体を低級アルカノールおよびクロロホ
    ルムから選ばれる有機溶媒で別々に抽出して該抗
    生活性物質を回収し、抽出溶媒から回収した抗生
    物質をクロロホルム:エタノール:水の4:7:
    2の混合物で処理し、分離した油状生成物を水中
    に注ぎ入れ、固化した生成物を分離し、さらにそ
    れを溶解液としてアセトニトリル:0.01NHClの
    1:1混合物を用いるシリカゲルのカラムクロマ
    トグラフイーにより精製し、これによつて抗生物
    質A/16686塩酸塩を回収し、これを交互結合し
    たデキストランゲルのカラムを用いるクロマトグ
    ラフイーにより、脱塩し、そして必要に応じて、
    抗生物質A/16686塩酸塩を普通の方法により対
    応する遊離塩基または他の生理学的に許容しうる
    酸付加塩に変えることを特徴とする特許請求の範
    囲第3項記載の方法。 6 抗生物質A/16686遊離塩基およびその生理
    学的に許容しうる酸付加塩からなる群より選ばれ
    る抗生物質を活性成分として含有することを特徴
    とする抗バクテリア剤。
