Description
本発明はシアノコバラミン水溶液の安定化法に
関する。 シアノコバラミンは悪性貧血、神経疾患、肝硬
変症、眼性疲労などに効果が認められ、医薬品と
して使用されている。しかしながら、その水溶液
はPH４−６ではかなり安定であるが、これを涙液
に近いPHすなわちPH7.0−7.5にすると安安性が悪
くなる。 コバラミン関連化合物の安定化剤としては従来
デキストラン、プロピレングリコール、フマール
酸、グリセリン、ソルビトール、クエン酸ナトリ
ウム、グルタミン酸ナトリウムなどが知られてい
るが、いずれも未だ充分満足すべきものではな
い。 本発明者らはさらにすぐれた安定化剤を求めて
研究を続けた結果、タウリンに顕著なシアノコバ
ラミン水溶液の安定化作用のあることを発見し
た。 第１および第２表は生理食塩水にシアノコバラ
ミン0.005W／Ｖ％、8.4ミリモル濃度のリン酸緩
衝液とさらにタウリン、グリシンまたはグリタミ
ン酸ナトリウムをそれぞれ所定濃度で加え最終的
にPH7.5（第１表）および（第２表）に調整し、
100℃、24時間の安定性を試験した結果を示す。
シアノコバラミンは日本薬局方の規定に従い
361nmの吸光度によつて定量した。 第１表において、安定化剤無添加の場合シアノ
コバラミンの残存率は50％以下であるがグルタミ
ン酸ナトリウム、グリシンまたはタウリンの添加
でそれぞれ残存率は上昇した。そして、就中タウ
リンの効果は著しく、タウリン0.1％の添加はグ
リシン約0.2％、グルタミン酸ナトリウム約１％
添加に匹敵する。第２表の結果もほぼ同様であ
る。

くは0.1−1W／Ｖ％である。 シアノコバラミン水溶液としてはシアノコバラ
ミンの薬効を期待しうる任意の濃度の水溶液が用
いられ、本発明の目的を阻害しない限り塩類、緩
衝剤、粘性剤または殺菌剤その他の薬剤を含有し
ていてもよい。 本発明の安定化剤はシアノコバラミン水溶液の
PHが7.0−7.5の範囲にあつても前記のように優れ
た効果を発揮するが、所望により他のPH範囲にお
いて使用してもよい。 実施例 １ （％） シアノコバラミン 0.01 タウリン 0.5 塩化ナトリウム 0.5 塩化カリウム 0.15 ホウ酸 0.5 ヒドロキシエチルセルローズ 0.1 リン酸一水素ナトリウム 0.2 塩化ベンザルコニウム 0.005 ホウ砂 0.03 精製水 を加えて 全量 100とする。 （PH7.2に調整する） 以上を無菌的に混合溶液とし、点眼瓶に入れて
４ケ月、40℃で遮光下に保存したのち測定したと
ころシアノコバラミンの残存率は92.9％であつ
た。 実施例 ２ （％） シアノコバラミン 0.02 タウリン 0.5 塩化ナトリウム 0.8 ホウ酸 0.5 ホウ砂 適量 塩化ベンザルコニウム 0.005 精製水を加えて 全量 100とする。 （PH7.2に調整する） 以上の混合溶液を無菌的に調整し、点眼瓶に充
填し、例１同様に保存、側定したところ同様のシ
アノコバラミン残存率を示した。

- 【特許請求の範囲】 １ タウリンを添加することを特徴とするシアノ
コバラミン水溶液の安定化法。 ２ タウリンの添加割合がシアノコバラミン水溶
液に対し0.05W／Ｖ％以上、好ましくは0.1−
1W／Ｖ％である特許請求の範囲第１項記載の安
定化法。 ３ シアノコバラミン水溶液のPHが6.5−7.5、好
ましくは7.0−7.5である特許請求の範囲第１項記
載の安定化法。
