Claims (47)
1. Humanizovana forma anti-c-met antitela glodara sadrži: (a) najmanje jednu HVR sekvencu odabranu iz grupe koju čine: (i) HVR-L1 sadrži sekvencu A1-A17, gde A1-A17 predstavlja KSSQSLLYTSSQKNYLA, (ii) HVR-L2 sadrži sekvencu B1-B7, gde B1-B7 predstavlja VVASTRES, (iii) HVR-L3 sadrži sekvencu C1-C9, gde C1-C9 predstavlja QQYYAYPWT, (iv) HVR-40H1 sadrži sekvencu Dl-DIO, gde D1-D10 predstavlja GYTFTSYWLH, and (v) HVR-H2 sadrži sekvencu E1-E18, gde E1-E18 predstavlja GMIDPSNSOTRFNPNFKD; i (b) varianta HVR-H3 sadrži sekvencu TYRSYVTPLDY, SYRSYVTPLDY, TYSSYVTPLDY, SYSSY-VTPLDY, TYSSYVTSLDY ili TYSSYVTALDY gde je monovalentni afinitet pomenutog antitela za humani c-met veći nego monovalentni afinitet antitela glodara koje sadrži varijabilnu sekvencu lakog lanca prikazanu kao SEQ ID NO: 9 i varijabilnu sekvencu teškog lanca prikazanu kao SEQ ID NO: 10 i ovde pomenuto antitelo inhibira c-met-zavisnu ćelijsku proliferaciju bolje od referentnog antitela koje sadrži himerično anti-c-met antitelo koje sadrži varijabilnu sekvencu lakog lanca i teškog lanca prikazanu kao SEQ ID NO: 509 i 10 kako sledi,
2. Antitelo iz patentnog zahteva 1, gde je najmanje deo okvir sekvence humana konsenzus okvir sekvenca. , sadrži varijantu HVR-55L

8. Antitelo iz patentnog zahteva 1, gde je monovalentni afinitet antitela za humani c-met najmanje 3-ostruko veći od monovalentnog afiniteta antitela glodara sadrži varijabilnu sekvencu lakog lanca i teškog lanca prikazanog kao SEQ ID NO: 9 i 10 na SI. 7.
9. Humanizovano antitelo iz patentnog zahteva 1, gde je antitelo glodara produkovano pomoću hibridoma ćelijske linije čuvane kao American Type Culture Collection Accession Number ATCC sa oznakom HB-11895 (hybridoma 5D5. 11. 6).

Antitelo iz patentnog zahteva 1, gde humanizovano antitelo inhibira vezivanje humanog faktora rasta hepatocita za njegov receptor bolje nego referentno antitelo koje podrazumeva himerično anti-c-met antitelo koje sadrži variabilnu sekvencu lakog lanca i teškog lanca prikazanu kao SEQ ID NO: 9 i 10 kako sledi.
17. Antitelo iz patentnog zahteva 16, gde antitelo inhibira vezivanje sa vrednosti IC50 koja je manja od polovine one koju ima himerično antitelo.
18. Antitelo iz patentnog zahteva 17, gde je IC50 određen preko opsega koncentracija antitela od oko 0. 01 nM do oko 1000 nM.
19. Antitelo iz patentnog zahteva 1, gde humanizovano antitelo inhibira aktivaciju receptora humanog faktora rasta hepatocita (HGF) bolje nego referentno antitelo koje podrazumeva himerično anti-c-met antitelo koje sadrži varijabilnu sekvencu lakog lanca i teškog lanca prikazanu kao SEQ ID NO: 9 i 10 kako sledi.
20. Antitelo iz patentnog zahteva 19, gde antitelo inhibira aktivaciju receptora sa vrednosti IC50 koja je manja od polovine one koju ima himerično antitelo.
