Opis wynalazku
Wynalazek dotyczy pochodnych sulfonowanych aminokwasów i zawierających je kompozycji farmaceutycznych stanowiących inhibitory metaloproteinazy.
Matryca pozakomórkowa składa się z kolagenu, proteoglikanu, itd. Jej funkcją jest wspieranie tkanek; odgrywa ona rolę w utrzymywaniu funkcji komórki, np. propagacji, różnicowania, adhezji lub tym podobnych. Metaloproteinazy matrycowe (MMP), takie jak żelatynaza, stromelizyna, kolagenaza itp. odgrywają ważną rolę w degradacji matrycy pozakomórkowej, i enzymy te działają na rzecz wzrostu, przebudowy tkanki, itd. w warunkach fizjologicznych. Zatem, uważa się, że te enzymy uczestniczą w postępie różnego typu chorób wiążących się z rozkładem i zwłóknieniem tkanek, takich jak zapalenie kości i stawów, reumatoidalne zapalenie stawów, owrzodzenie rogówki, zapalenie ozębnej, przerzut i nacieczenie nowotworu i infekcja wirusowa (np. infekcja HIV). Obecnie nie jest jasne, który enzym istotnie uczestniczy w powyższych chorobach, lecz uważa się, że enzymy te uczestniczą co najmniej w rozkładzie tkanki. Ujawniono jako inhibitory metaloproteinazy pochodne aminokwasów, np. pochodne aminokwasów kwasu hydroksamowego (JP-A-6-2562939), pochodne aminokwasów kwasu karboksylowego i/lub ich pochodne kwasu hydroksamowego (WO95/35276) itd.
O ile jest możliwe inhibitowanie aktywności MMP, sądzi się, że inhibitory MMP przyczyniają się do poprawy stanu i profilaktyki powyższych chorób spowodowanych lub związanych z aktywnością MMP. Stąd tez od dawna było pożądane opracowanie inhibitorów MMP.
W powyższej sytuacji, twórcy wynalazku ustalili, że pewien rodzaj pochodnych sulfonamidowych wykazuje silną aktywność inhibitowania MMP.

Korzystnie konfiguracja asymetrycznych atomów węgla związanych z R1 jest konfiguracją R.
Przedmiotem wynalazku jest również kompozycja farmaceutyczna zawierająca pochodną o wzorze I, w którym R4 oznacza wiązanie, -CH=CH- lub -ChC-, a pozostałe podstawniki są takie jak określono dla wzoru I.
Kompozycja ta jest przeznaczona do inhibitowania metaloproteinazy, zwłaszcza do inhibitowania kolagenazy typu-IV.
Związki według wynalazku można zsyntetyzować z odpowiednich α-aminokwasów przedstawionych wzorem (XV) za pomocą następujących 6 metod syntetycznych. Do roztworu 500 mg (1,4 mmol) związku (XVII-2) otrzymanego w przykładzie 1 w 12 ml bezwodnego tetrahydrofuranu dodano 387 mg (2 x 1,4 mmol) sproszkowanego węglanu potasu, 319 mg (1,5 x 1,4 mmol) kwasu 4-metoksyfenyloborowego i 81 mg (0,05 x 1,4 mmol) tetrakis-(trifenylofosfino) palladu. Uzyskaną mieszaninę mieszano w atmosferze argonu przez 48 h w 75°C. Mieszaninę reakcyjną rozcieńczono octanem etylu. Warstwę organiczną przemyto 1N HCl, 5% NaHCO3 aq. i wodą, osuszono nad Na2SO4 i zatężono pod obniżonym ciśnieniem. Pozostałość poddano chromatografii kolumnowej na żelu krzemionkowym. Frakcje wymywane n-heksanem / octanem etylu = 3/1 zebrano i powtórnie wykrystalizowano z n-heksanu uzyskując 447 mg żądanego związku (XIX-1). Wydajność 83%. T. topn. 122-123°C.

Do roztworu 390 mg (1,01 mmol) związku (XIX-1) w 8 ml tetrahydrofuranu i 8 ml metanolu dodano 5,1 ml 1N NaOH i uzyskaną mieszaninę mieszano w 60°C przez 6 h. Mieszaninę reakcyjną zatężono pod obniżonym ciśnieniem, by usunąć rozpuszczalnik organiczny. Uzyskaną pozostałość rozcieńczono octanem etylu. Mieszaninę zakwaszono wodnym roztworem kwasu cytrynowego i wyekstrahowano octanem etylu. Warstwę organiczną przemyto solanką, osuszono Na2SO4 i zatężono pod obniżonym ciśnieniem uzyskując 373 mg związku (Ia-3-1). Wydajność 100%. T. topn.: 174 - 176°C; IR(KBr, ν max cm-1): 1735, 1503, 1343, 1163.

Kolagenazę typu IV (MMP-9) oczyszczono według metod opisanych w: Scott M. Wilhelm i in., J. Biol. Chem., 264, 17213-17221, (1989), SV40-transformed Human Lung Fibroblasts Secrete a 92-kDa Type IV Collagenase Which Is Identical to That Secreted by Normal Human Macrophages; Yasunori Okada i in., J. Biol. Chem., 267, 21712-21719, (1992), Matrix Metalloproteinase 9 (92-kDa Gelatinase / Type IV Collagenase) from HT 1080 Human Fibrosarcoma Cells; Robin V. Ward i in., Biochem. J., (1991) 278, 179-187, The purification of tissue inhibitor of metalloproteinase-2 from its 72 kDa progelatinase complex.
MMP-9 jest wydzielane z linii komórkowej ludzkiego włókniakomięsaka ATCC HT 1080, do jego pożywki hodowli, gdy jest stymulowany przez 12-tetradekanoiloforbolo-13-octan (TPA). Wytwarzanie MMP-9 w tej hodowli zweryfikowano przez zymografię żelatynową jak opisano w: (Hidekazu Tanaka i in., (1993) Biochem. Biophys. Res. Commun., 190, 732-740, Molecular cloning and manifestation of mouse 105-kDa gelatinase cDNA). Pożywkę kondycjonowaną stymulowanego HT 1080 zatężono i oczyszczono za pomocą żelatyno-Sepharose 4B, konkanawalino-A-Sepharose i Sephacryl S-200. Tak otrzymane oczyszczone pro-MMP-9 (92 kDa, żelatynaza B) dała pojedyncze dodatnie pasmo w zymografii żelatynowej. Następnie otrzymano aktywowany MMP-9 przez działanie na pro-MMP-9 trypsyna.
(2) Metody prób inhibitorów kolagenazy typu IV.
Próbę kolagenazy wykonano stosując aktywowany MMP-9 opisany powyżej i substrat dostarczony w zestawie do badania aktywności kolagenazy typu IV (YAGAI, Inc.), według protokołu wytwórcy. Wykonano następujące 4 próby na związek (inhibitor).
(A) substrat (kolagenaza typu IV), enzym (MMP-9), inhibitor (B) substrat (kolagenaza typu IV), inhibitor (C) substrat (kolagenaza typu IV), enzym (MMP-9) (D) substrat (kolagenaza typu IV)
Według protokołu wytwórcy, zmierzono intensywność fluorescencji i wyznaczono procentowe inhibitowanie z następującego równania:
Inhibitowanie (%) = {1- (A - B) / (C - D)} x 100
IC50 oznacza stężenie przy którym procentowe inhibitowanie osiągnie 50%. Wyniki podano w tabeli 9.

HIV) i zarostowego stwardnienia tętnic i anewryzmu arteriosklerotycznego i miażdżycy tętnic, nawrotu zwężenia, posocznicy, szoku septycznego, zakrzepicy tętnicy wieńcowej i anormalnej angiogenezy, zapalenia twardówki, stwardnienia rozsianego, jaskry z otwartym kątem przesączania, retynopatii, retynopatii proliferacyjnej, jaskry nowonaczyniowej, skrzydlika, zapalenia rogówki, pęcherzowego oddzielania się naskórka, łuszczycy, cukrzycy, zapalenia nerek, choroby neurodegeneracyjnej, zapalenia dziąseł, wzrostu nowotworu, angiogenezy nowotworowej, nowotworu oczu i naczyniakowłókniaka, hemonaczyniaka, gorączki, krwotoku, koagulacji, kacheksji i anoreksji i ostrej infekcji, wstrząsu i choroby autoimmunologicznej, malarii, choroby Crohna, zapalenia opon i wrzodu żołądka, ponieważ związek według wynalazku wykazuje silną aktywność inhibitującą względem metaloproteinazy, zwłaszcza MMP.

1. Pochodna sulfonowanych aminokwasów o wzorze I w którym
R1 oznacza C3-C5-alkil, trifluoroetyl, cykloheksylometyl, hydro ksymetyl, benzyloksymetyl, karboksylometyl, karboksyloetyl, fenyl, benzyl ewentualnie podstawiony przez grupę nitrową, atom fluoru,
PL 198 905 B1 lub karboksyl, 1-fenylo-1-hydroksymetyl, fenyloetyl, bifenylometyl, naftylometyl, indolilometyl ewentualnie podstawiony na atomie azotu przez metyl, formyl, acetyl, metylosulfonyl lub etoksykarbonyl, lub w pierścieniu benzenowym przez metyl, metoksyl lub atom fluoru, tiazolilo-metyl, 4-pirydylometyl, benzotiazolilometyl lub benzimidazolilometyl;
R2 oznacza atom wodoru, metyl, 4-aminobutyl lub benzyl;
R4 oznacza wiązanie, -CH2-, -CH=CH-, -ΟξΟ-, -CO-NH-, -N=N-, -NH-, -NH-CO-NH-, -NH-CO-, -SO2NH-, -SO2-NH-N=CH- lub tetrazolodiyl;

Pochodna według zastrz. 1 o wzorze I, w którym R4 oznacza wiązanie, -CH2-, -ΟξΟ-, -N=N-, -NH-, -NH-CO-NH-, lub tetrazolodiyl; pod warunkiem, że R5 oznacza ewentualnie podstawiony ww aryl lub ewentualnie podstawiony ww heteroaryl gdy R4 oznacza tetrazolodiyl, jej postać optycznie czynna, jej farmaceutycznie dopuszczalna sól lub jej hydrat.

Pochodna według zastrz. 1 o wzorze I, w którym R4 oznacza -CO-NH-, -NH-CO-, -SO2NHlub -SO2-NH-N=CH-; pod warunkiem, że R5 oznacza ewentualnie podstawiony ww aryl lub ewentualnie podstawiony ww heteroaryl, gdy R4 oznacza -CO-NH- lub -NH-CO-, jej postać optycznie czynna, jej farmaceutycznie dopuszczalna sól lub jej hydrat.

6. Pochodna według zastrz. 4, albo 5, o wzorze I, w którym R1 oznacza izopropyl, benzyl lub (indol-3-ilo)metyl.
7. Pochodna według zastrz. 4, albo 5, o wzorze I, w którym R5 oznacza fenyl ewentualnie podstawiony jednym lub więcej podstawnikami wybranymi spośród metyloksylu, metylotio, metylu i t-butylu.
8. Pochodna według zastrz. 4, albo 5, o wzorze I, w którym konfiguracja asymetrycznych atomów węgla związanych z R1 jest konfiguracją R.
