Dmax(最大溶解率)為如上逑特定時間内最大溶解率, 且一般D max值宜接近1 D 0 ( Χ )。 ιο (量介值)為製藥物溶解率之介質,®值越小則溶解 率越高,m值越大則溶解率越低。 II (形狀介值)為溶解曲線形狀之介值。當η為1時, Weibull 函數可表為溶解率 U) = X U-exp[-((T-Ti)) /m】},因此與一级動力相同,溶解曲線為線性。當η小 於1時,溶解曲線幾為平行。當η大於1時,溶解曲線 為S形。 T i (位介值)為直到開始溶解之落後時間之介值,依本 發明含大環内酯化合物之徐放性處方亦可由該Weibull 函數而具特色。因此,目的徐放性處方可由調整Dmax (最大溶解率)至80%以上,宜為90%以上,更宜為95%以上 ,η(量介值)定為0.7〜20,宜為1〜12,更宜為1.5〜8, η(形狀介質)為0.2〜5,宜為0.3〜3,更宜為0.5〜1.5, 且Ti(位介值)為0〜12,宜為0〜8,更宜為0〜4。 經濟部智慧財產Α員工消費合作社印製 由ml/η取代上述Weibull函數中m及η介值之值表為活性 成份之最大溶解率中之63.2Χ由處方中釋放(Τ63.2%)。 即Τ 6 3 . 2 % ( h r ) = m 1/η。本發明徐放性處方之釋放特性亦 可由溶解試驗,JP \111方法2(獎法)使用0.0 0 5 %羥丙基 纖維素溶液(調至ΡΗ4 · 5)之試驗溶液評估。依本發明 含大環内酯化合物之徐放性處方,(Τ63.2Χ)—詞其中大 本紙張尺度適用中國國家標準(CNS ) A4規格(210X 297公釐) 570814 Α7 Β7 經濟部智慧財產局員工消費合作社印製 五、發明説明(4 ) 環内酯化合物之63.2%最大溶解量由處方中釋放為〇·7〜 15小時。以往雖已製得含大環内酯化合物之快速釋放處 方,但未曽製得任何徐放性處方,即其T63·2 35為0·7〜15 小時,且此徐放性處方於臨床上極為稱許。本發明首次 達成^ gT63.2 低於〇·?小時,由口投予無法有效徐放 大環内酯化合物。當各方Τ63 · 2Χ高於15小時,活性成份 之釋放會被阻礙而於達到有效血濃度前即失去活性成份 ,因而不適於本發明處方。當Τ63.2 35為1·〇〜12小時, 可得更佳徐放性。Τ63.2 更宜為1.3〜8·2小時,且最佳 徐放性處方為Τ63.2Χ為2〜5小時。 依本發明所用"大環内酯化合物"為具12員以上化合物 之統稱,由鏈徽腸生成大環内酯化合物,如雷帕徽素, 大克力馬斯(FK506),及子囊撤素及其類似物及衍生物 皆為大環内酯化合物。大環内酯化合物之特例可由下式 (I )三環化合物例示。
