576832 五、發明說明(1) 〜 發明所屬的技術領域 x 1 本發明是有關具有優異抗菌作用及有經口投與性之啉 羧酸衍生物或其鹽以及含有此等化合物質之抗菌劑。 習知技術 以喹啉羧酸為基本骨幹之化合物,因具有優異之抗菌力 和廣幅度抗菌讀,所以為周知之大多數能作為合成抗菌劑 者。 其中,去N -氧化噁二唑(η 〇 r f u r ο X a c i n e ;特開昭 5 3 - 1 4 1 2 8 6號公報)、稀醇N -氧化σ惡二嗤(e η o x a c i n e ;特開 眧5 5 - 3 1 0 4 2號公報)、全N -氧化σ惡二峻(o f u r o jc a c i n e ;特開 昭 57-46986 號公報)、雙N -氧化 σ惡二嗤(diprc^furoxacine; 特開昭5 8 - 746 6 7號公報)、通N-氧化噁二唑 -- (tosfuroxacine;特開眧60-2 28479號公報)等感染症治療 劑,在臨床上用得很多。 然而,此等化合物在抗菌力、腸吸收性、代謝安定性等 方面並未能充分發揮效率,又,喹啉羧酸系化合物中有特 有之光毒性或細胞毒性之減輕等,因此仍存留有許多改善 課題。再者,最近,對此等藥劑又出現了耐性菌之問題。 發明所欲解決的課題 於是,本發明之目的是提供可適用於臨床上、有優異抗 菌力、腸吸收性、代謝安定性、副作用少之抗…菌劑。 關於如此之實際收況,本發明人為了提供臨床上優異藥 劑而刻意研究之結果,發現下述,一般式(I )

第7頁 576832 五、發明說明(4) 、 本發明之化合物1是可以形成酸加鹽或鹽基加成鹽t兩^* 面之形態,同時,也包含此等鹽與硼化合物所形成之i合 物。 作為酸加成鹽,可列舉如(1 )與鹽酸、硫酸、磷酸等無 機酸之鹽、(2 )與蟻酸、醋酸、檸檬酸、三氯醋酸、三氟 氟酸、富馬酸(fumaric acid)、馬來酸等有機羧酸之鹽、 (3 )與甲石黃酸、苯績酸、對-甲苯石黃酸、三曱基苯續酸、萘 磺酸等磺酸之鹽,或作為鹽基加成鹽,可列舉如(1 ’)與 鈉、鉀等鹼金屬之鹽、(2’)鈣、鎂等鹼土金屬之鹽、(3’) 氨鹽、(4’)與三甲胺、三乙胺、三丁胺、D比哮、N,N -二甲 苯胺、N-甲基哌啶、N-甲基嗎琳、二乙胺、攀己胺、普魯 卡因(procaine)、二〒胺、N -〒-万-苯乙胺、1-埃-費拿命 (ephenamine)、N,N’ -二〒基乙二胺等含氮有機鹽基之_ 鹽。又;作為硼化合物,如氟化硼等鹵化硼、乙酸硼等低 級醯氧化硼等。其中以酸加成‘鹽較佳,尤其以馬來酸鹽、 曱磺酸鹽、對位-曱苯磺酸鹽、鹽酸鹽最好。 本發明之化合物1或其鹽不僅是可成無溶劑和型,也可 以成水和物或溶劑和物存在,因此,本發明之化合物是包 含全部為結晶型及水和甚至於溶劑和物。 本發明之化合物1或其鹽可用任意方法製造,其中一個 製造方法例子列舉如下所示。

第10頁 576832 五、發明說明(7) 、 量,針對化合物(A )而言,為等莫爾以上,尤以等莫t爾# 倍莫爾為佳。 - 又,別的方法有,在化合物(A)中,與N,N -二甲基曱醯 胺乙醛縮二甲醇、N,N -二曱基曱醯胺乙醛縮二乙醇等縮醛 類反應後,與化合物(C )反應也可以導出化合物(D )。與縮 醛之反應中,使用之溶劑只要不影響本反應之任何一種溶 劑都可以,例如前述所列舉之溶劑。本反應通常是在 0〜1 5 0 °C範圍内,最好是在室溫〜1 0 0 °C進行;反應時間通 常是在10分鐘〜48小時之内,最好為1〜10小時。 其次,化合物(D)環化反應而加成成化合物(E)之反應, 是在鹽基性化合物存在下或不存在下於適當$溶劑中進 行,本反應所使用之溶劑,只要不影響本反應之任何一種 溶劑都可以,可列舉如,苯、曱苯、二曱苯等芳香族碳_氫 化合物;二乙醚、四氫呋喃、環氧己烷、一甘醇曱醚、二 甘醇曱醚等醚類;二氣曱烷、氣仿、四氣化碳等鹵化碳氫 類;二曱基曱醯胺、二甲亞楓等非質子極性溶劑;曱醇、 乙醇、丙醇等醇類。又使用之鹽基性化合物則可以列舉如 ,金屬鈉、金屬鉀等鹼金屬類;氫化鈉、氫化鉀等金屬氫 化物;氫氧化鈉、氫氧化鉀、碳酸鈉、碳酸鉀等無機鹽類 ;甲醇鈉、乙醇鈉、第三丁醇鉀等醇鹽類;氟化鈉、氟化 鉀等金屬氟化物;三乙胺、1,8 -二氮雜二環4,0 ]十一 碳烯(DBU)等有機鹽基類。本反應之溫度通常是在0〜200 °C 範圍内,最好是在室溫〜1 8 0 °C進行;反應通常是在5分鐘 〜2 4小時之内完成。鹽基性化合物之使用量,針對化合物
