Abstract

Цей винахід належить до очищення гемоглобіну хроматографією. Спосіб очищення гемоглобіну із сирого розчину, що містить сторонні протеїни, здійснюється шляхом іонообмінної хроматографії. Сирий розчин використовують в концентрації 0,1-20%. Розчин на першій стадії пропускають через колонку, заповнену або аніонообмінником для очищення сирого розчину, в якому сторонні речовини мають кислішу реакцію, ніж гемоглобін, або катіонообмінником для очищення сирого розчину, в якому сторонні речовини мають більш основну реакцію, ніж гемоглобін. Безперервне пропускання сирого розчину на першій стадії здійснюють спочатку до повного насичення колонки гемоглобіном і компонентами з високою спорідненістю до відповідного іонообмінника, а надалі – до перевантаження колонки з подальшим заміщенням на ній гемоглобіну. На другій стадії здійснюють пропускання елюату, одержаного на першій стадії, через аніоно- або катіонообмінник, який не було застосовано на першій стадії. При цьому хроматографію здійснюють також спочатку до насичення колонки гемоглобіном і речовинами з вищою спорідненістю до колонки, а потім до перевантаження і заміщення на ній гемоглобіну.
