Description
Verfahren zur Herstellung von Bornanocarbonsäureamiden
Die Erfindung betrifft ein Verfahren zur Herstellung neuer Bornanocarbonsäureamide der Formel

in der die Reste R identisch sind und Wasserstoff oder Methyl darstellen; R1 Wasserstoff; R2 Hydroxy oder R1 und R2 das Sauerstoffatom einer Ketogruppe, die auch ketalisiert sein kann; und R3 Phenyl, das durch ein Halogenatom, eine niedere Alkyl-, Alkoxy-, Alkylthio-, Acylamino-, Acylamino - niederalkyl-, eine Cyano-, Carbamoyl- oder Diacylimidogruppe substituiert sein kann; oder Cycloalkyl oder einen 5- oder 6gliedrigen N-verknüpften stickstoffhaltigen Heterocyclus darstellt, und deren Salzen.
Unter niederen Alkylgruppen, die in durch das Symbol R3 repräsentierten Resten enthalten sind, sollen geradkettige oder verzweigte Alkylgruppen mit 1-6 Kohlenstoffatomen verstanden werden. Beispiele dafür sind Methyl, Äthyl, Propyl, Isopropyl, Butyl, Pentyl, Hexyl und deren Isomeren. Ein durch das Symbol R3 dargestelltes Halogenphenyl ist vorzugsweise Chlorphenyl.
Eine durch das Symbol R3 dargestellte Acylamino-phenylgruppe kann aliphatische oder aromatische Acylreste enthalten. Beispiele für solche Gruppen sind Acetaminophenyl oder Benzoylaminophenyl. Eine Diacylimidophenylgruppe ist beispielsweise die Phthalimidophenylgruppe. Ein Acylaminoalkylphenylrest ist z. B.
ein Benzamidoäthylphenylrest, der im Benzamidorest durch Niederalkoxy- und/oder Halogen substituiert ist.
Beispiele für 5- oder 6gliedrige stickstoffhaltige heterocyclische Ringe sind Piperidin und Pyrrolidin. Beispiele für Cycloalkyl sind Cyclopentyl und Cyclohexyl.
Als Ketale kommen beispielsweise Dialkylketale, wie das Dimethyl- oder Diäthylketal; oder Alkylenketale, wie das Äthylenketal in Betracht.
Die Verbindungen der Formel I werden erfindungsgemäss dadurch hergestellt, dass man ein entsprechendes reaktionsfähiges Campher-3-carbonsäure- bzw. Nor campher-3 -carbons äurederivat, in dem die ringständige Ketogruppe auch ketalisiert sein kann, mit einem Sulfonsäureamid der Formel R3-SO2-NH2 II oder einem Alkalisalz davon umsetzt. Gewünschtenfalls wird das Reaktionsprodukt in ein Salz übergeführt.
Die Umsetzung einer Verbindung der Formel mit einem der genannten Campher- bzw. Norcampherderivate wird zweckmässig so vorgenommen, dass man den Reaktionspartner in einem geeigneten organischen Lösungsmittel, gegebenenfalls unter Erwärmen, zusammenbringt. Geeignete Lösungsmittel sind z. B. Kohlenwasserstoffe, wie Benzol; halogenierte Kohlenwasserstoffe, wie Chloroform; oder Dimethylformamid.
Die erfindungsgemäss erhältlichen Verbindungen der Formel I können in verschiedenen Konfigurationen, die von der Stereochemie der bicyclischen Ausgangsmaterialien abhängt, vorliegen, z. B. als Racemat oder in optisch aktiver Form. Bevorzugte Verbindungen der Formel I sind solche, in denen R3 Alkylphenyl, Alkylthiophenyl oder Acylaminoalkylphenyl darstellt und davon insbesondere diejenigen, die sich konfigurativ vom DL- oder D-Campher bzw. vom Borneol oder Isoborneol ableiten.
Die blutzuckersenkende Wirkung kann mittels der nachstehend beschriebenen Versuchsanordnung ermittelt werden: Als Versuchstiere werden weibliche Hunde, die nach eintägiger Standarddiät 16 Stunden gefastet haben, in Gruppen zu drei Tieren je Präparat und Dosis verwendet. Die Präparate werden in Gelatinekapseln per os verabreicht. Vor der Medikation und in bestimmten Abständen danach werden Blutproben entnommen.
Nach Heparinisierung wird das Plasma abgetrennt und die Glucose darin nach der Glucoseoxydase-Methode (Anal. Biochem. 3 131 (1962) im Technicon-Autoanalyzer bestimmt. Als Standard wird 4 % ige Rinderalbuminlösung mit bekannten Glucosemengen verwendet. Die erhaltenen Ergebnisse werden statistisch ausge wertet und in % Abweichung vom Leerwert umgerechnet. Positive Werte bedeuten Zunahme, negative Werte Abnahme der Plasmaglucosekonzentration.
Mit dieser Versuchsanordnung wurden bei Verabreichung von jeweils 30 ,umol Substanz ermittelt: (1 R)-2-OxoN-(p-tolylsulfonyl)-3-endo-bornano carboxamid: - 25 % (nach 24 Stunden); (1 R)-N- { [p-(2-[5-Chlor-o-anisamido] -äthyi).phenyU sulfonyl}-2-oxo-3 -endo-bornanocarboxamid: -36% (nach 2 Stunden) und -27% (nach 4 Stunden); (lR)-N-{[p-(Methylthio)-phenyl]-sulfonyl}-2-oxo-
3 -endo-bornano-carboxamid: -14% (nach 2 Stunden) und 16% (nach 4 Stunden).
Die Verfahrensprodukte können als Heilmittel in Form pharmazeutischer Präparate Verwendung finden, welche sie oder ihre Salze in Mischung mit einem geeigneten pharmazeutischen, organischen oder anorganischen inerten Trägermaterial enthalten.
In den nachfolgenden Beispielen sind die Tempe raturen in Celsiusgraden angegeben.
Beispiel 1
1,6 g Sulfamylpiperidin und 1,1 g Triäthylamin werden in 10 ml Dimethylformamid gelöst. Dazu lässt man innerhalb 15 Minuten eine Lösung von 2,1 g D-Camphercarbonsäurechlorid in 5 ml Benzol zutropfen. Nach Rühren über Nacht wird das Lösungsmittel am Rotavapor abgedampft. Das resultierende zähflüssige Ö1 wird beim Aufnehmen in etwa 50 ml Wasser kristallin. Man erhält 2 g (1 R)-2-Oxo-N-(piperidinosulfonyl)-3-endobornanocarboxamid vom Schmelzpunkt 1530 (aus Äthanol). [a]D + 79,20 (in Chloroform).
Beispiel 2
In Analogie zu Beispiel 1 erhält man aus D-Campher-3-carbonsäurechlorid und p-Tolylsulfonamid das (1 R)-2-Oxo-N-(p-tolylsulfonyl)-3-endobornanocarbox- amid vom Schmelzpunkt 139-1490 (Zers.) (aus Äthanol/ Wasser; [1Z]D + 94,70 (in Chloroform).
Beispiel 3
In Analogie zu Beispiel 1 erhält man aus D-Campher-3-carbonsäurechlorid und dem Natriumsalz von p-Athylthiobenzolsulfonamid das (1 R)-N-{[p-(iithylthio)-phenyl]-sulfonyl}-
2-oxo-3 -endobornanocarboxamid-Natrium vom Schmelzpunkt 152-1540 (aus Athanol/itther).
Beispiel 4
In Analogie zu Beispiel 1 erhält man aus D-Campher-3-carbonsäurechlorid und dem Natriumsalz von p-(p-Acetaminoäthyl)-benzolsulfonamid das (1 R)-N-{[p-(2-Acetamidoäthyl)-phenyl]-sulfonyl}-
2-oxo-3 -endobornanocarboxamid-Natrium vom Schmelzpunkt 178-1800 (aus Äthanol).
Beispiel 5
In Analogie zu Beispiel 1 erhält man aus D-Campher-3-carbonsäurechlorid und dem Natriumsalz von p-Acetylaminobenzolsulfonamid das (1 R)-N-(N-Acetylsulfanilyl)-2-oxo-3 -endobornano- carboxamid-Natrium vom Schmelzpunkt 2400 (aus Athanol).
Beispiel 6
In Analogie zu Beispiel 1 erhält man aus D-Campher-3-carbonsäurechlorid und dem Natriumsalz von 4 Sulfamoyl-benzoesäureamid das (1 R)-N-[(p-Carbamoylphenyl)-sulfonyl]-2-oxo-
3 -bornanocarboxamid-Natrium als Gemisch der endo- und exo-Isomeren, das mit 1 Mol Athanol und 1 Mol Wasser kristallisiert; das Produkt schmilzt bei 218-22010 (aus Athanol/Wasser).
Beispiel 7
In Analogie zu Beispiel 1 erhält man aus D-Campher-3-carbonsäurechlorid und 4-[ss-(2-Methoxy-5-chlor- benzamido)-äthyl]-benzolsulfonamid das (1 R)-N-{ [p-(2-(5 -Chlor-o-anisamido)-äthyl)-phenyl]- sulfonyl}-2-oxo-3 -endo-bornanocarboxamid, das mit 1 Mol Äthanol und 0,5 Mol Wasser kristallisiert; das Produkt schmilzt bei 108-1100 (aus Äthanol).
Beispiel 8
In Analogie zu Beispiel 1 erhält man aus D-Cam pher-3 -carbons äurechlorid und p-Cyanobenzolsulfamid das (1 R)-N-[(p-Cyanophenyl)-sulfonyl]-2-oxo-3-endobornanocarboxamid vom Schmelzpunkt 163-1660 (aus 22ithanol/Wasser); [a]D + 32,40 (in Äthylacetat).
Claims (1)
- PATENTANSPRUCHVerfahren zur Herstellung von Bornanocarbonsäureamiden der Formel EMI2.1 in der die Reste R identisch sind und Wasserstoff oder Methyl darstellen; R1 Wasserstoff; R- Hydroxy oder R1 und R2 das Sauerstoffatom einer Ketogruppe, die auch ketalisiert sein kann; und R3 Phenyl, das durch ein Halogenatom, eine niedere Alkyl-, Alkoxy-, Alkylthio-, Acylamino-, Acylamino - niederalkyl-, eine Cyano-, Carbamoyl-, oder Diacylamidogruppe substituiert sein kann;oder Cycloalkyl oder einen 5- oder 6gliedrigen N-verknüpften stickstoffhaltigen Heterocyclus darstellt, und deren Salzen, dadurch gekennzeichnet, dass man ein entsprechendes reaktionsfähiges Campher-3-carbonsäure- bzw. Nor-campher-3-carbonsäurederivat, in dem die ringständige Ketogruppe auch ketalisiert sein kann, mit einem Sulfonsäureamid der Formel R3-SO^NHe II oder einem Alkalisalz davon umsetzt.UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man ein D- oder D,L-Campher-3-carbonsäurehalogenid mit einem Alkyl-, Alkylthio- oder Acylaminoalkylbenzolsulfonamid oder einem Alkalisalz davon umsetzt.2. Verfahren nach Patentanspruch und Unteranspruch 1, dadurch gekennzeichnet, dass man ein D Campher-3-carbonsäurehalogenid mit p-Tolylsulfonamid oder einem Alkalisalz davon umsetzt.3. Verfahren nach Patentanspruch und den Unteransprüchen 1 und 2, dadurch gekennzeichnet, dass man ein Reaktionsprodukt der Formel I in ein Salz überführt.
