Claims (1)
- 【特許請求の範囲】 1.ドセタキセルおよびドセタキセル誘導体から選択されるタキソイド系の治療 剤、1種以上の不飽和リン脂質および小量の1種以上の負電荷リン脂質を含んで 成ることを特徴とする、安定で、かつドセタキセルおよびドセタキセル誘導体か ら選択されるタキソイド系の活性成分について高度に濃縮されている製薬組成物 。 2.ドセタキセルおよびドセタキセル誘導体から選択されるタキソイド系の治療 剤を3〜15mg/ml含んで成ることを特徴とする請求の範囲第1項に記載の製薬 組成物。 3.ドセタキセルおよびドセタキセル誘導体から選択されるタキソイド系の治療 剤が一般式: 式中、 −記号R1およびR2は、それぞれ水素原子を表すか、あるいは基R1およびR2 の一方は水素原子を表し且つ他方はヒドロキシル、アシルオキシまたはアシルカ ルボニルオキシ基を表すか、あるいはR2は水素原子を表し且つR1はメチル基の 炭素原子とα-位で結合を形成してシクロプロパン環を形成し、 −記号R3およびR4の一方は水素原子を表し且つ他方はヒドロキシル基を表わ すか、あるいはR3およびR4は一緒になってオキソ基を形成 し、 −記号R5およびR6は、それぞれ水素原子を表すか、あるいは記号R5および R6の一方は水素原子を表し且つ他方はヒドロキシル、アシルオキシ、アシルカ ルボニルオキシまたはアルコキシメチルカルボニルオキシ基を表すか、あるいR5 およびR6は一緒になってオキソ基を形成し、 −記号R7はアルコキシ、アルケニルオキシまたはシクロアルキルオキシ基を 表し、そして −R8は直鎖状もしくは分枝状のアルキル、直鎖状もしくは分枝状のアルケニ ルまたは直鎖状もしくは分枝状のアルキニル基または3〜6個の炭素原子を含有 するシクロアルキル基を表すか、あるいは場合によってはハロゲン原子およびア ルキル、アルコキシ、ジアルキルアミノ、アシルアミノ、アルコキシカルボニル アミノおよびトリフルオロメチル基から選択される同一または異なる1つ以上の 原子または基により置換されてもよいフェニル基、あるいは窒素、酸素および硫 黄原子から選択される同一または異なる1つ以上のヘテロ原子を含む5-員の芳 香族複素環式基を表し、 ここでアルキル基および他の基のアルキル部分は、直鎖もしくは分枝鎖中に1 〜8個の炭素原子を含み、そしてアルケニルまたはアルキニル基は2〜8個の炭 素原子を含むものとする、 の生成物から選択されることを特徴とする請求の範囲第1または2項のいずれ かに記載の製薬組成物。 4.ドセタキセルおよびドセタキセル誘導体から選択されるタキソイド系の治療 剤が、R2が水素原子を表し、R1が水素原子またはヒドロキシ ル基を表すか、あるいはR1がメチル基の炭素原子とα位で単結合を形成し、R3 およびR4が一緒になってオキソ基を形成し、R5が水素原子を表し、そしてR6 が水素原子またはヒドロキシル、アセチルオキシまたはメトキシアセチルオキシ 基を表すか、あるいはR5およびR6は一緒になってオキソ基を形成し、R7がt- ブトキシ基を表し、そしてR8がイソブチル、イソブテニル、ブテニル、シクロ ヘキシル、フェニル、2-フリル、3-フリル、2-チエニル、3-チエニル、2-チアゾ リル、4-チアゾリルまたは5-チアゾリル基を表す請求の範囲第2項に定義された 生成物から選択されることを特徴とする請求の範囲第1、2または3項に記載の 製薬組成物。 5.治療剤がドセタキセルであることを特徴とする請求の範囲第1ないし第4項 のいずれか1項に記載の製薬組成物。 6.治療剤が4α,10β-ジアセトキシ-2α-ベンゾイルオキシ-5β,20-エポキシ-1 β-ヒドロキシ-7β,8β-メチレン-9-オキソ-19-ノル-11-タキセン-13α-イル(2R ,3S)-3'-t-ブトキシカルボニルアミノ-2'-ヒドロキシ-3'-フェニルプロピオネー トであるとを特徴とする請求の範囲第1ないし第4項のいずれか1項に記載の製 薬組成物。 7.活性成分の濃度が5から10mg/mlより高い値の間であることを特徴とする 請求の範囲第1ないし第6項のいずれか1項に記載の製薬組成物。 8.不飽和リン脂質が天然、合成または半−合成のリン脂質であることを特徴と する請求の範囲第1ないし第7項のいずれか1項に記載の製薬組成物。 9.不飽和リン脂質が天然のリン脂質であることを特徴とする請求の範 囲第8項に記載の製薬組成物。 10.天然のリン脂質が、植物起源のリン脂質、特にヒマワリまたはダイズ起源 のリン脂質であることを特徴とする請求の範囲第8項または第9項に記載の製薬 組成物。 11.不飽和リン脂質が70〜100%のホスファチジルコリンから成ることを 特徴とする請求の範囲第1ないし第10項のいずれか1項に記載の製薬組成物。 12.植物起源の天然リン脂質が70%より高いレベルの不飽和アシル鎖含むこ とを特徴とする請求の範囲第10項または第11項に記載の製薬組成物。 13.ドセタキセルおよびドセタキセル誘導体から選択されるタキソイド系の治 療剤が、リン脂質の和に対して1〜30重量%の比率で導入されることを特徴と する前記請求の範囲のいずれか1項に記載の製薬組成物。 14.治療剤がリン脂質の和に対して3〜20重量%の比率で導入されることを 特徴とする請求の範囲第13項に記載の製薬組成物。 15.治療剤がリン脂質の和に対して3.5〜10重量%の比率で導入されるこ とを特徴とする請求の範囲第13項または第14項に記載の製薬組成物。 16.負電荷のリン脂質(1つまたは複数)が、ホスファチジルグリセロール、 ホスファチジルセリン、ホスファチジルイノシトール、ホスファチジン酸、また はそれらの誘導体のアルカリ金属塩または四級アンモニウム塩から選択されるこ とを特徴とする前記請求の範囲のいずれか1項に記載の製薬組成物。 17.負電荷のリン脂質(1つまたは複数)が不飽和リン脂質(1つまたは複数 )の総量に対して約0.1〜4重量%の比率で導入されることを特徴とする請求 の範囲第16項に記載の製薬組成物。 18.負電荷のリン脂質(1つまたは複数)が不飽和リン脂質(1つまたは複数 )の総量に対して約0.4〜0.8重量%の比率で導入されることを特徴とする 請求の範囲第16項または第17項に記載の製薬組成物。 19.負電荷のリン脂質(1つまたは複数)が不飽和リン脂質(1つまたは複数 )の総量に対して約0.5重量%の比率で導入されることを特徴とする請求の範 囲第16項ないし第18項のいずれか1項に記載の製薬組成物。 20.液体、凍結または凍結乾燥されることができる前記請求の範囲のいずれか 1項に記載の製薬組成物。 21.凍結防止剤および/または注射すべき最終溶液の等張性を調整するための 薬剤を含むことを特徴とする前記請求の範囲のいずれか1項に記載の製薬組成物 。 22.薬剤が糖、ポリマーおよびアミノ酸から選択されることを特徴とする請求 の範囲第21項に記載の製薬組成物。 23.200nm未満の平均直径の粒子を含むことを特徴とする前記請求の範囲のい ずれか1項に記載の製薬組成物。 24.100nm未満の平均直径の粒子を含むことを特徴とする請求の範囲第23項 前記に記載の製薬組成物。 25.1種以上の不飽和リン脂質、少量の1種以上の負電荷リン脂質、およびド セタキセルおよびドセタキセル誘導体から選択されるタキソイ ド系の活性成分を、アルコールに溶解することにより均一な分散液を生成し、そ してアルコールを全部または一部蒸発させてゲルまたは粘稠な液体を得、このゲ ルまたは液体は水を撹拌しながら加えることにより溶解し、そして次に均一化し 、得られる分散液を次に場合によっては凍結または凍結乾燥することを特徴とす る前記請求の範囲のいずれか1項に記載の製薬組成物の調製法。 26.アルコールがエタノールであることを特徴とする請求の範囲第25項に記 載の方法。 27.得られる分散液を滅菌濾過に供することを特徴とする請求の範囲第25項 または第26項に記載の方法。 28.直ぐに使用できる、安定化された注射可能な溶液を調製するための、請求 の範囲第20項に記載の凍結または凍結乾燥した溶液の使用。
