Description
Die Erfindung betrifft ein Verfahren zur Herstellung neuer substituierter Purine, von denen insbesondere die 9-(2-Hydr-oxyäthoxymethyl)-Derivate, wie Adenin, Guanin, Thioguanin und 2,6-Diaminopurin, von Bedeutung sind.
ls Über die Synthese verschiedener acyclischer Purin-Nucleo-side wurde von Schaeffer et al. in J. Med. Chem. 14 (1971) 367 in einer Studie über die Wechselwirkung zwischen Adenosin-desaminase und dem Enzymsubstrat berichtet, wobei insbesondere 9-(2-Hydroxyäthoxymethyl)-adenin erwähnt und 20 über die Messung von dessen Substrataktivität mit Adenosin-desaminase berichtet wurde.

worin X Sauerstoff oder Schwefel und R1, R2, R3, R4, R5 und R6 verschiedenartige Substituenten darstellen, eine Antivirus-40 Wirkung zeigen und insbesondere gegenüber den verschiedenen Arten von DNA- und RNA-Viren wirksam sind, wie sowohl in vitro- als auch in vivo-Versuche zeigten. Diese Verbindungen sind insbesondere gegen die folgenden Viren wirksam: Cyto-megalovirus, Adenovirus, insbesondere Adenovirus 5, Rhino-45 virus, Mengovirus und Sindbisvirus, ferner ganz besonders gegen Kuhpocken- und Herpes-Viren, einschliesslich H. simplex, H. zoster und H. varicella, bei Säugetieren, welche Krankheiten, wie Keratitis herpetica bei Kaninchen und Encephalitis herpetica bei Mäusen, hervorrufen. Darüberhinaus sind die so Verbindungen zur Behandlung von Mononucleosis infectiosa nützlich.

f) Rs nur dann eine substituierte Carbamoylgruppe der Formel NH-CO-Z, worin Z der Gruppe -CH(NH2)CH2-CöH5 entspricht; bedeutet, wenn R1 Dialkylamino darstellt; ausgenommen, dass R2 nicht Wasserstoff bedeutet, wenn Rs Hydroxy und R1 Monoalkylamino darstellen; R1 nur dann Chlor bedeutet, wenn R2 nicht Wasserstoff darstellt; R1 nicht Amino oder Monoalkylamino bedeutet, wenn Rs Benzyloxy und R2, R3 und R6 Wasserstoff darstellen; sowie deren Säureadditionssalze, insbesondere deren pharmazeutisch annehmbaren Salze.
Aufgrund ihrer starken Wirkung sind diejenigen Verbindungen der Formel I besonders bevorzugt, worin X Sauerstoff;
R1 Amino, Hydroxy oder Alkylthio ;
R2 Amino;
R5 Hydroxy, Benzoyloxy, Carboxypropionyloxy, Acetoxy oder Hydroxyalkyl; und R3, R4 und R6 jeweils Wasserstoff bedeuten, vorausgesetzt, dass R5 nur dann Hydroxyalkyl bedeutet, wenn R1 Hydroxy darstellt, und dass R5 nur dann Acetoxy bedeutet, wenn R1 Amino darstellt.
Verbindungen der Formel A, worin
X Schwefel;
R1 Amino oder Monoalkylamino; und R2, R3, R4, Rs und R6 jeweils Wasserstoff bedeuten, haben sich ebenfalls als hochwirksam erwiesen.
Als Halogensubstituent wird Chlor bevorzugt. Alkyl bedeutet im vorliegenden Fall allgemein eine Gruppe mit 1 bis 12, vorzugsweise 1 bis 8, Kohlenstoffatomen.
Als Säureadditionssalze, die für therapeutische Anwendungen bevorzugt sind, kommen insbesondere solche in Betracht, die sich von pharmazeutisch annehmbaren organischen oder anorganischen Säuren, wie Milchsäure, Essigsäure, Äpfelsäure, p-Toluolsulfonsäure, Salzsäure oder Schwefelsäure, herleiten.
Gegenstand der Erfindung ist das in Patentanspruch 1 definierte Verfahren.
Die Ausgangsverbindungen der Formel II, worin M und/oder G Halogen bedeuten, können beispielsweise durch Halogenie-rung entsprechender Verbindungen erhalten werden. Ferner können Hydroxygruppen durch Hydrolyse, Amin- oder Mono-oder Dialkylaminogruppen durch Aminolyse; Mercapto- oder Alkylthiogruppen durch Schwefelung entsprechender Vorstufen in die Verbindung der Formel II eingeführt werden.
Diese Methoden sind sämtlich bekannt und können der Veröffentlichung Heterocyclic Compounds-Fused Pyrimidines,
Part II Purines, herausgegeben von D. J. Brown, (1971) Wiley-Interscience, entnommen werden.
Die Ausgangsverbindungen der Formel II können auch hergestellt werden, indem man ein Purin der Formel III

Für A kommen beispielsweise reaktive Reste organischer oder anorganischer Säuren, z. B. Halogen oder Carboxylat, in Betracht. Bedeutet Q eine abspaltbare Gruppe, so ist diese bevorzugt eine Acylgruppe.
Die bevorzugte Methode zur Herstellung der Verbindungen der Formel II besteht in der Kondensation von Purinen, die in 2- und 6-Stellung die gewünschten Substituenten tragen, mit einem acyl- oder aralkyl-blockierten 2-Halogenalkoxy-äthanol, z. B. 2-Benzoyloxyäthoxymethylchlorid, in einem stark polaren Lösungsmittel, wie Dimethylformamid oder Hexamethylphosphoramid, in Gegenwart einer Base, wie Triäthylamin oder Kaliumcarbonat. Die Reaktion wird vorzugsweise bei Raumtemperatur durchgeführt. Dabei wird allgemein ein längerer Zeitraum, z. B. von mehreren Tagen, benötigt, um vernünftige Ausbeuten zu erhalten.
Eine andere Möglichkeit besteht darin, die Kondensation durch Einwirkung von Wärme, z. B. in der Schmelze, zu bewirken. Dabei wird das gewünschte Produkt direkt erhalten.
Für diese Art der Reaktion erhitzt man ein geeignet substituiertes Purin zusammen mit einem Acyloxyalkoxymethyl-carboxylat, z. B. 2-Oxa-l,4-butandiol-diacetat, in Gegenwart einer katalytischen Menge einer stärken Säure, wie Schwefelsäure. Im allgemeinen sind Temperaturen oberhalb 100 °C erforderlich, sie sollten jedoch 200 °C nicht überschreiten,

um die Gefahr einer Zersetzung auf ein Minimum zu reduzieren.
Zweckmässigerweise wählt man die Temperatur so, dass das Gemisch der Reaktionspartner zusammenfliesst, d. h. schmilzt, bevor sich diese zersetzen.
Die Schmelzreaktion kann auch durchgeführt werden, indem man ein 9-Acylpurin mit einem entsprechenden Alkoxymethyl-carboxylat oder -halogenid unter im wesentlichen gleichen Bedingungen, jedoch möglicherweise etwas niedrigeren Temperaturen, reagieren lässt.
Ausserdem kann die Schmelzreaktion auch unter Verwendung eines entsprechenden Diesters, z. B. 2-Acetoxyäthoxy-methylacetat, durchgeführt werden.
Die erfindungsgemäss erhältlichen Verbindungen können zu pharmazeutischen Zubereitungen oder Präparaten verarbeitet werden. Hierfür besonders geeignet sind solche Verbindungen, bei denen R6 eine Alkylgruppe mit 1 bis 8 Kohlenstoffatomen bedeutet, ferner solche Verbindungen, bei denen die Substituenten einen Alkylanteil mit 1 bis 4 Kohlenstoffatomen aufweisen. Die Wirkstoffe können, insbesondere in Form ihrer pharmazeutisch annehmbaren Säureadditionssalze, mit geeigneten, in der Pharmazie üblichen, Trägerstoffen verarbeitet werden. Bevorzugt werden die Präparate in Form von Einzeldosiszubereitungen angeboten.
Dieser oben verwendete Ausdruck «wirksame Einzeldosis» bedeutet eine vorausbestimmte gegen Viren wirksame Menge, die genügt, um in vivo den Virusorganismus zu bekämpfen. Pharmazeutisch annehmbare Trägerstoffe sind Stoffe, welche sich zur Darreichung des Medikamentes eignen. Sie können fest, flüssig oder gasförmig sein, ferner sollen sie inert und medizinisch annehmbar und mit den Wirkstoffen verträglich sein.
Diese pharmazeutischen Zubereitungen können parenteral, oral, als Zäpfchen oder als Scheideneinlage verabreicht werden, ferner lokal als Salbe, Crème, Aerosol oder Puder angewendet werden, oder als Augen- oder Nasentropfen verabreicht werden. Die jeweilige Zubereitungsform hängt davon ab, ob eine innerliche oder äusseriiche Virusinfektion behandelt werden soll.
Bei innerlichen Infektionen werden die Zubereitungen für gewöhnlich oral oder parenteral in Dosen verabreicht, welche für die freie Base berechnet sind, nämlich 0,1 bis 250, vorzugsweise 1,0 bis 50, mg/kg Körpergewicht des Säugetiers. Beim Menschen werden sie als Einzeldosis, und zwar mehrmals täglich, in einer Menge von 1 bis 250 mg/Einzeldosis verabreicht.
Für die orale Darreichung geeignete Zubereitungen können feine Pulver oder Granulate, Verdünnungsmittel, Dispersions-mittel und/oder oberflächenaktive Mittel enthalten und in einer Mixtur, entweder in Wasser oder in Sirup, in Kapseln oder Sachets in trockenem Zustand oder in einer nicht-wässri-gen Lösung oder Suspension, welche Suspendierungsmittel enthalten, vorliegen. Ferner können die Zubereitungen Tabletten sein, welche Bindemittel oder Gleitmittel enthalten oder aber Suspensionen in Wasser oder Sirup.
Ist es wünschenswert oder notwendig, so können in den Zubereitungen auch Geschmacksmittel, Konservierungsmittel, Suspensionsmittel, Eindickungsmittel oder Emulgatoren enthalten sein. Eine bevorzugte Darreichungsform sind Tabletten und Granulate, wobei diese auch überzogen sein können. Für die parenterale Darreichung oder für die Darreichung in Tropfenform, beispielsweise bei Augeninfektionen, können die Verbindungen in wässriger Lösung in einer Konzentration von ungefähr 0,1 bis 10, vorzugsweise 0,1 bis 7, am meisten bevorzugt in einer Konzentration von 0,2, Gew./Vol./ %, vorliegen. Die Lösungen können Antioxydantien, Puffer und/ oder andere Zusätze enthalten.
Andererseits können die vorliegenden Verbindungen bei der Infektion von Augen oder anderen äusserlichen Geweben, beispielsweise im Mund oder auf der Haut, als äusseriiche Salbe oder Crème an dem infizierten Körperteil des Patienten angewendet werden. Die Verbindungen können in einer Salbe, beispielsweise mit einer wasserlöslichen Salbengrundlage,
oder in einer Crème, beispielsweise mit einer Öl-in-Wasser-Crèmegrundlage, in einer Konzentration von ungefähr 0,1 bis 10, vorzugsweise 0,1 bis 7, am besten jedoch 1, Gew./Vol/% verabreicht werden.
Besonders bevorzugt aufgrund ihrer äusserst starken Wirkung gegen Viren, insbesondere gegen Herpes-Viren, sind 9-(2-Hydroxyäthoxymethyl)-guanin (R1 = OH, R2 = NH2) und 2-Amino-9-(2-hydroxyäthoxymethyl)-adenin. Darüberhinaus besitzen auch die nachfolgenden Verbindungen eine sehr starke Wirkung gegen die Herpes-Viren und Kuhpocken-Viren:
9-(2-Benzoyloxyäthoxymethyl)guanin,
9-(3-Hydroxypropoxymethyl)guanin,
2-Amino-6-methylthio-9-(2-hydroxyäthoxymethyl)purin,
9-{2-(3-Carboxypropionyloxy)äthoxymethyl}guanin,
9-(2-Acetoxyäthoxymethyl)-2,6-diaminopurin,
9-Äthylthiomethyladenin und das 9-Äthylthiomethyl-6-
methylaminopurin.
Die nachfolgenden Beispiele dienen zur Erläuterung der Erfindung.
Beispiel 1
2-Chlor-9-(2-hydroxyäthoxymethyl)-adenin
2,6-Dichlorpurin wurde mit l-Benzoyloxy-2-chlormethoxy-äthan kondensiert, wobei im wesentlichen nach der von Schaeffer et al. (1. c.) beschriebenen Methode gearbeitet wurde. Man erhielt 2,6-Dichlor-9-(2-benzoyloxy-äthoxymethyl)-purin einer Ausbeute von41% d. Th.; Schmelzpunkt 121 bis 125°C. Die so erhaltene Verbindung wurde mit Ammoniak und Methanol im Bombenrohr auf 95 °C erhitzt. Dabei erfolgte eine gleichzeitige Ammonolyse der 6-Chlor- und der Benzoyl-oxygruppe. Man erhielt die Titelverbindung einer Ausbeute von 94% d. Th.
Schmelzpunkt 188 bis 190°C nach Umkristallisieren aus Isopropanol.
Beispiel 2 9-(2-Hydroxyäthoxymethyl)guanin (I; R1 = OH, R2 = NH2)
0,97 g festes Natriumnitrit wurde bei Zimmertemperatur unter Rühren während eines Zeitabschnittes von einer Stunde zu einer Lösung von 0,5 g 2-Chlor-9-(2-hydroxyäthoxymethyl)-adenin in 10 ml Eisessig hinzugefügt. Das Reaktionsgemisch wurde während zusätzlicher 4V2 Stunden gerührt. Der weisse Feststoff wurde durch Filtrieren entfernt, mit kalter Essigsäure ausgewaschen und sodann gut mit kaltem Wasser verrieben, um vorhandenes Natriumacetat zu entfernen. Das feste Produkt wurde zurückgehalten. Das vereinigte Essigsäurefiltrat und die Waschflüssigkeiten wurden bei vermindertem Druck bei 40 °C eingedampft und der ölige Rückstand mit kaltem Wasser verrieben. Das resultierende feste Material wurde mit dem oben isolierten Feststoff vereinigt und die vereinigten Feststoffe getrocknet und aus Äthanol umkristallisiert, wobei 0,25 g 2-Chlor-9-(2-hydroxyäthoxymethyl)hypoxanthin resultierten.
Schmelzpunkt: >310°C. Elementaranalyse und NMR-Spektrum stimmten mit dieser Struktur überein.
Eine Mischung von 0,375 g 2-Chlor-9-(2-hydroxyäthoxy-methyl)hypoxanthin und 80 ml Methanol, die mit wasserfreiem Ammoniak gesättigt war, wurde in einem Bombenrohr bei 125 °C 5 Stunden lang erhitzt. Das Bombenrohr wurde in s

einem Eisbad gekühlt und das Reaktionsgemisch entfernt. Das Lösungsmittel und der überschüssige Ammoniak wurden bei vermindertem Druck bei 50 °C entfernt. Nach dem Verreiben des Rückstandes mit kaltem Wasser zur Entfernung des gebildeten Ammoniumchlorids wurde der restliche Feststoff getrocknet und aus Methanol umkristallisiert, wobei 0,24 g reines 9-(2-Hydroxyäthoxymethyl)guanin resultierten. Schmelzpunkt: 256,5 bis 257°C.
Beispiel 3
2-Amino-9-(2-hydroxyäthoxymethyl)adenin (I; R1 = R2 = NHa)
Eine Mischung aus 4,4 g 2-Chlor-9-(2-hydroxyäthoxy-methyl)adenin und 40 ml flüssigem Ammoniak wurde in einem Bombenrohr 19 Stunden lang auf 120°C erhitzt. Das Reaktionsgemisch wurde aus dem Bombenrohr entfernt und der überschüssige Ammoniak im Vakuum entfernt. Der Rückstand wurde zwischen Äther und Wasser verteilt und die wässrige Schicht noch zweimal mit Äther und einmal mit Chloroform ausgewaschen. Die wässrige Lösung wurde sodann mit einem Überschuss eines stark basischen Ionenaustauscherharzes behandelt, um das Ammoniumchlorid in Ammoniak überzuführen, und das Ganze im Vakuum zur Trockne eingedampft. Der Rückstand wurde in der minimalen Menge Äthanol gelöst, sodann mit 10 g «Florisil» versetzt, und das Gemisch zur Trockne eingedampft. Der Rückstand in Chloroform wurde auf eine vorbereitete «Florisil»-Säule gebracht und mit 5 % Methanol/95 % Chloroform (Vol./Vol.) eluiert. Es wurden Fraktionen von je 30 ml gesammelt und mittels Dünnschichtchromatographie (Süicagelplatten, entwickelt in 10% Methanol/ 90% Chloroform) auf das Vorhandensein des Reaktionsproduktes geprüft. Die ersten 40 Fraktionen enthielten Ausgangsmaterial aber kein Reaktionsprodukt und wurden verworfen. Die anschliessenden Fraktionen, welche das gewünschte Produkt enthielten, wurden vereinigt und zur Trockne abgedampft. Der Rückstand wurde aus Äthanol (mit Aktivkohlebehandlung) und aus n-Propanol umkristallisiert. Es resultierten 0,5 g (12% der theoretischen Ausbeute) 2-Amino-9-(2-hydroxyäthoxymethyl)adenin.
Schmelzpunkt: 183 bis 184°C.
«Florisil» ist ein aktiviertes Magnesiumsüikat, welches häufig bei der Säulenchromatographie verwendet wird.
Beispiel 4
Herstellung von 2,6-Diazido-9-(2-benzoyl-oxyäthoxymethyl)purin 10 g 2,6-Dichlor-9-(2-benzoyloxyäthoxymethyl)purin, 3,5 g Natriumazid und 54 ml eines Äthanol/Wasser-Gemisches (1:1, Vol./Vol.) wurden unter Rühren 31h Stunden lang bei 110 bis 120° C unter Rückfluss und Rühren erhitzt. Die Dünnschichtchromatographie des Reaktionsgemisches zeigte eine vollständige Umsetzung. Beim Abkühlen über Nacht verfestigte sich das öl und wurde filtriert, mit Äthanol und Wasser ausgewaschen und aus Äthanol umkristallisiert. Die Verbindung wurde mit quantitativer Ausbeute erhalten und wies einen Schmelzpunkt bei 124 bis 125°C auf. Die Verbindung ist in wässriger Base labil und sonnenlichtempfindlich.
Beispiel 5 9-Äthylthiomethyladenin Ein Reaktionsgemisch, welches 5,53 g Chlormethyl-äthyl-sulfid, 7,73 g 6-Chlorpurin und 5,57 g Triäthylamin in 50 ml Dimethylformamid enthielt, wurde 3 Tage lang bei Zimmertemperatur stehen gelassen. Sodann wurde abfiltriert und das Filtrat eingedampft. Der halbfeste Rückstand wurde in ungefähr 80 ml Chloroform gelöst, auf eine Säule aus 360 g «Florisil» (aktiviertes Magnesiumsilikat) in Chloroform gegeben

und mit Chloroform eluiert. Das anfängliche Eluat (zirka 500 ml) wurde verworfen; die nächsten 2,8 Liter wurden aufgefangen und eingedampft. Sodann wurden ungefähr 50 ml Ligroin zu dem öligen Rückstand gegeben, die Mischung abgekühlt und das resultierende weissliche kristalline 6-Chlor-9-äthylthiomethylpurin abfiltriert und mit Ligroin ausgewaschen. Ausbeute: 3,3 g.
Schmelzpunkt: 78 bis 810 C.
Die Umkristallisierung aus Äther/Ligroin ergab weisse Nadeln.
Schmelzpunkt: 81 bis 82,5 °C.
1,5 g 6-Chlor-9-äthylthiomethylpurin und 20 ml flüssiger Ammoniak wurden in ein Bombenrohr gefüllt und über Nacht auf 60 °C erwärmt. Nach Abdampfen des Ammoniaks erhielt man einen Rückstand, wplcher mit kaltem Wasser versetzt, anschliessend filtriert und mit kaltem Wasser ausgewaschen wurde. Man erhielt 1,17 g 9-Äthylthiomethyladenin als weissen Feststoff, Schmelzpunkt 140 bis 142 °C. Die Umkristallisation aus Äthanol ergab weisse Platten, Schmelzpunkt 142 bis 143 °C.
Beispiel 6 9-(2-Hydroxyäthylthiomethyl)adenin
Nach der von Miles und Owen in J. Chem. Soc. (1952)
Seite 817 beschriebenen Methode wurde 2-Acetoxyäthanthiol hergestellt. Dabei wurden 102,1 g Essigsäureanhydrid während 174 Stunden zu einer Mischung aus 78,1 g 2-Mercaptoäthanol und 3 ml einer 10%igen Schwefelsäurelösung in Essigsäure zugesetzt. Während der Zugabe wurde von aussen gekühlt, um die Reaktionstemperatur unterhalb 40 °C zu halten. Nach Beendigung der Zugabe des Essigsäureanhydrids wurde das Reaktionsgemisch 1 Stunde lang auf 65 °C erhitzt und sodann über Nacht bei Zimmertemperatur stehen gelassen. Sodann wurden 500 ml Äther zu dem Reaktionsgemisch zugegeben und die resultierende Lösung dreimal mit je 100 ml Wasser und einmal mit 100 ml Kochsalzlösung ausgewaschen. Die ätherische Lösung wurde über wasserfreiem Natriumsulfat und Natriumbicarbonat getrocknet. Die Destillation ergab 77,5 g 2-Acetoxyäthanthiol.
Siedepunkt: 57 bis 67 °C bei 10 mm Hg.
In eine Mischung von 24,0 g 2-Acetoxyäthanthiol und 6,0 g Paraformaldehyd wurde 3 Stunden lang Chlorwasserstoffgas bei mässiger Geschwindigkeit unter äusserem Kühlen eingeleitet. Sodann wurden 25 g Calciumchlorid zugesetzt und das Reaktionsgemisch in einem Salz/Eisbad 4 Stunden lang stehen gelassen. 200 ml Methylenchlorid wurden zugesetzt, das Reaktionsgemisch filtriert und das Lösungsmittel unter strengstem Feuchtigkeitsausschluss abgedampft. Der ölige Rückstand wurde destilliert und ergab 18,5 g 2-Acetyloxyäthyl-chlor-methylsulfid.
Siedepunkt: 82 bis 87 °C bei 3 mm Hg.
Auf die von Schaeffer in J. Med. Chem., Band 14 (1971)
Seite 367 beschriebene Weise wurden 3,37 g 2-Acetoxyäthyl-chlormethylsulfid zu einer Mischung aus 3,09 g 6-Chlorpurin, 2,23 g Triäthylamin und 20 ml Dimethylformamid hinzugefügt. Das Reaktionsgemisch wurde bei Zimmertemperatur 90 Stunden lang gerührt und das Lösungsmittel sodann bei vermindertem Druck entfernt. Der ölige Rückstand wurde in 30 ml Chloroform gelöst und in eine Säule aus 200 g «Florisil» in Chloroform eingebracht. Die Eluierung ergab das gewünschte Produkt in 2,1 Liter Eluat, nachdem die anfänglichen 300 ml Eluat verworfen worden waren. Das Lösungsmittel wurde abgedampft und ergab einen öligen Rückstand. Sodann wurde Äther/Petroläther zugesetzt und die Mischung abgeschreckt, wobei 1,4 g 9-(2-Acetoxyäthylthiomethyl)-6-chlorpurin (Schmelzpunkt: 82 bis 87 °C) resultierten. Die Umkristallisation aus Äther ergab weisse Prismen mit einem Schmelzpunkt

von 89 bis 91°C. 1,1 g 9-(2-Acetoxyäthylthiomethyl)-6-chIor-purin und 20 ml Ammoniak wurden in ein Bombenrohr eingebracht und 24 Stunden lang auf 60 °C erhitzt. Das Reaktionsgemisch wurde aus dem Bombenrohr entfernt und der Ammoniak abgedampft. Der Rückstand wurde mit kaltem Wasser verrieben, filtriert und mit zusätzlichem kaltem Wasser ausgewaschen. Man erhielt 0,73 g rohes 9-(2-Hydroxyäthylthio-methyl)adenin mit einem Schmelzpunkt bei 166 bis 169,5 ° C. Nach Umkristallisieren aus Äthanol erhielt man weisse Plättchen mit einem Schmelzpunkt bei 170 bis 172° C.
Beispiel 7 9-Äthylthiomethyl-N-6-methyladenin 1,2 g 6-Chlor-9-äthylthiomethylpurin und 20 ml Methylamin wurden in ein Bombenrohr eingefüllt und 24 Stunden lang auf 60 °C erhitzt. Das überschüssige Methylamin liess man dann bei Zimmertemperatur sich verflüchtigen. Der Rückstand wurde mit 10 ml Eiswasser verrieben und filtriert. Durch Umkristallisieren des lohfarbenen Feststoffes aus Äther mit s Kohle («Darco 60») erhielt man 0,64 g 9-ÄthyIthiomethyl-N-6-methyladenin.
Schmelzpunkt: 111,5 bis 113°C.
Beispiel 8
io 2,6-Diamino-9-(2-acetoxyäthoxymethyl)purin
Auf die in Beispiel 9 beschriebene Weise wurde 2,6-Di-chlor-9-(2-acetyloxyäthoxymethyl)purin in 2,6-Diazido-9-(2-acetyloxyäthoxymethyl)purin mit einem Schmelzpunkt von 69 bis 70°C (aus Äthanol/Wasser) übergeführt (Aus-i5 beute 83 %d. Th.)

- 626364PATENTANSPRÜCHE 1. Verfahren zur Herstellung neuer, in 9-Stellung substituierter Purine der Formel ICH-X-CH-CH-R-»5Ifi 1bR1R2 R3R4 R5worinX Schwefel oder Sauerstoff;Wasserstoff, Hydroxy, Alkoxy, Mercapto, Alkylthio, Amino, Monoalkylamino, Dialkylamino oder Azido; Wasserstoff, Halogen, Amino oder Azido;Wasserstoff, Alkyl mit gerader oder verzweigter Kette, Cycloalkyl oder Benzyloxymethyl;Wasserstoff oder Niederalkyl;Wasserstoff, eine Hydroxy-, Amino-, Alkyl-, Hydroxy-alkyl-, Phosphato-, Benzoyloxy-, Benzoyloxymethyl-, Benzyloxy-, Sulfamoyloxy-, Carboxypropionyloxy-, Acetoxy- oder eine substituierte Carbamoylgruppe der Formel NH-CO-Z, wobei Z unsubstituiertes oder ein-oder mehrfach durch Sulfonyl, Amino, Carbamoyl oder Halogen substituiertes Alkyl, Aryl oder Aralkyl; und R6 Wasserstoff oder Alkyl bedeuten, vorausgesetzt, dass R1 nicht Amino oder Methyl-amino ist, falls X Sauerstoff, R2, R3, R4 und R6 Wasserstoff und Rs Wasserstoff, Hydroxy oder Benzyloxy bedeuten, dass R1 nicht Amino oder Äthylamino bedeutet, wenn R2, R3, R4 und R5 Wasserstoff und R6 Methyl darstellen und dass mindestens R1 Amino, Mono-, Dialkylamino der Azido oder R2 Amino oder Azido bedeutet, sowie von deren Säureadditionssalzen, dadurch gekennzeichnet, dass man ein in einem in 9-Stellung substituierten Purin der Formel II(II),CH-X-CH-CH-R"b b b worin M und/oder G Halogen bedeuten und der gegebenenfalls verbleibende Rest M oder G die für R1 bzw. R2 angegebene Bedeutung hat, das für M stehende Halogenatom durch eine Amino-, Monoalkylamino-, Dialkylamino- oder Azido-gruppe und/oder das für G stehende Halogenatom durch eine Amino- oder Azidogruppe ersetzt und, sofern das Reaktionsprodukt eine Base ist, die Verbindung der Formel I gegebenenfalls in ein Säureadditionssalz überführt oder, falls das Reaktionsprodukt ein Salz der Verbindung der Formel I ist, dieses gegebenenfalls in die freie Base oder in ein anderes Salz überführt.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man eine erhaltene Verbindung der Formel I, worin R1 und/oder R2 eine Azidogruppe bedeuten, zur entsprechenden Aminoverbindung reduziert.(I),10
