Stand der Technik
Die Verbindungen dieser Erfindung sind neu Benzothia-zolin-Derivate, worin der Substituent in 3-Stellung an dem Benzothiazolinring, Niederalkanoyl ist und derjenige in 2-Stellung Phenyl bedeutet, das durch eine Ethergruppe in beliebiger Lage wieder substituiert ist. 2-Phenylbenzothiazolin-Derivate wurden von H. Breuer et al. (U.S. Pat. Nr. 3 720 683), H.D. Cossey et al. (J. Chem. Soc., 1965,954) und P.J. Palmer et al. (J. Med. Chem., 14,248) beschrieben. Das U.S. Patent von Breuer et al. bezieht sich auf 2-Phenylbenzo-

thiazolin-Derivate, in welchen der Phenylring durch ein Alkyl usw. substituiert ist, aber nicht durch eine Ethergruppe, und die pharmakologischen Wirkungen sind entzündungshemmend und anti-mikrobiell.
3 Cossy et al. und Palmer et al. beschreiben 2-Phenylben-zothiazolin-Derivate, in welchen der Phenylring durch eine Ethergruppe substituiert ist, aber der Substituent in 3-Stel-lung ist nicht Niederalkanoyl, aber Wasserstoff, Alkyl,
Benzyl usw., und die pharmakologische Wirkung ist anti-mikrobiell.
Darstellung der Erfindung
Die Erfindung bezieht sich auf Benzothiazolin-Derivate der Formel (1)

Die Verbindungen dieser Erfindung sind Benzothiazolin-Derivate, bei welchen der Substituent in 3-Stellung an dem 25 Benzothiazolinring Niederalkanoyl und derjenige in 2-Stel-lung Phenyl ist, welcher wieder durch eine Ethergruppe in beliebiger Stellung substituiert ist. Die Verbindungen dieser Erfindung sind nicht nur neu in ihrer chemischen Struktur, aber ebenfalls wurden sie zur Behandlung von kardiovaskulären 30 Krankheiten nützlich gefunden und eine derartige Wirkung auf kardiovaskuläre Krankheiten wurde nicht bei bekannten Benzothiazolin-Derivaten gefunden.
Kardiovaskuläre Krankheiten sind Angina cordis, Arhythmia, Thrombose usw., und ß-Blocker, Inhibitoren der 35 Blutplättchen-Aggregation, Calciumantagonisten usw. werden als therapeutisches Mittel verwendet.
In pharmakologischen Tests wurde bewiesen, dass die Verbindung dieser Erfindung eine überlegene Blutplättchen-Anti-Aggregationswirkung und Calcium-Antagonisations-40 Wirkung aufweisen, so dass sie für kardiovaskuläre Krankheiten nützlich sind.
Bevorzugte Verfahren zur Herstellung der erfindungsge-mässen Verbindungen sind nachfolgend zusammengestellt: Die Reaktion der Verbindung der Formel (II) mit der 45 Verbindung der Formel (III),

Die Verfahren werden ausführlich wie folgt beschrieben: Die Reaktion des Hydroxy-Derivates der Formel (II) mit dem Halogenid der Formel (III) benötigt keine spezifischen Bedingungen und bekannte Methoden, die allgemein für eine s Reaktion eines Hydroxy-Derivates mit einem Halogenid benutzt werden, können verwendet werden, aber vorzugsweise wird die Reaktion in einem organischen Lösungsmittel (z.B. Man kann Methoden, die allgemein für eine Reaktion eines Amin-Derivates mit einem Halogenid, einem Carbonsäure-Derivat, Aldehyde-Derivat oder Epoxy-Derivat verwendet werden, anwenden.
Die Verbindungen dieser Erfindung können in Säure-Salze umgewandelt werden. Die genannten Salze werden gewöhnlich durch allgemeine Methoden unter Verwendung anorganischer Säuren oder organischer Säuren erhalten. Beispiele für pharmazeutisch annehmbare Salze dieser Verbindungen sind Chlorwasserstoffsäuresalze, Schwefelsäuresalze, Phosphorsäuresalze, Milchsäuresalze, Maleinsäuresalze, Fu-marsäuresalze, Methansulfonsäuresalze, p-Toluolsulfonsäu-resalze, Oxalsäuresalze usw.
Die Verbindungen der Formel (I) haben Stereoisomere, da eines oder mehrere asymmetrische Kohlenstoffatome vorhanden sind, und diese Isomeren werden von dieser Erfindung ebenfalls erfasst.
Beispiele werden nachfolgend gezeigt, und die Verbindungen dieser Erfindung sind in Tabellen I-XI zusammengestellt. Die Zuordnungen der NMR-Spektren in Tabelle X wer-55 den gemäss den Zahlen der Formel (IX) gemacht.

65 Aber Protonen (aromatische), die nicht zugeordnet werden, werden als aromatische H (Ar-H) bezeichnet.
Das Proton der -OCH3-Gruppe wird als -OCH3(P) zugeteilt, wenn die Gruppe die Gruppe in C2'-C6'-Stellung der For-
mei (IX) gebunden ist, und wird als -OCH3(A) bezeichnet, wenn die Gruppe an die Phenylgruppe, die der Substituent des Aminanteils der Seitenkette ist, gebunden ist.
Beispiel 1
3-Acetyl-2-[ 3-t-butylamino -2- hydroxypropoxy ) phenyl] ben-zothiazolin-Hydrochlorid ( Verbindung Nr. 86)
1,31g Acetyl-2-[2,3-epoxypropoxy) phenyljbenzothiazo-lin und 4,21 ml t-Butylamin werden in 10 ml Ethanol gelöst und man erhitzt die Lösung eine Stunde lang am Riickfluss. Nach dem Abkühlen gibt man 2 ml 2N HCl/Ethylacetat zu der Reaktionsmischung und die Lösung wird im Vakuum konzentriert. Zu dem Rückstand gibt man Ether, um Kristalle herzustellen. Die Kristalle werden durch Filtration gesammelt und ergeben 1,3 g (82%) der Titelverbindung. Die physikalischen Daten sind in Tabelle VII gezeigt. Die Verbindungen, die in der Tabelle VII gezeigt sind, werden durch eine ähnliche Methode, die weiter oben beschrieben ist, unter Verwendung entsprechender Amin-Deri-vate hergestellt.
Beispiel 2
3-Acetyl-2-[2- ( 3-t-butylamino -2- hydroxypropoxy )phenyl] benzothiazolin-Hydrochlorid (VerbindungNr. 86)
1,64 g 3-Acetyl-2-[2-(2,3-epoxypropoxy)phenyl] benzo-thiazolin, 1,82 g 3-Acetyl-2-[2-(3-chlor -2- hydroxyprop-oxy)benzothiazolin und 10 ml t-Butylamin werden in 30 ml Ethanol gelöst und man erhitzt die Lösung 3 Stunden lang am Rückfluss. Die Lösung wird im Vakuum konzentriert und der Rest in Ethylacetat gelöst. Die Lösung wird mit gesättigter Natriumchloridlösung gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Zu dieser Lösung werden 2N HCl/Ether gegeben, um Kristalle zu produzieren. Die Kristalle werden filtriert, um 3,5 g (80%) der Titelverbindung zu geben.
Die physikalische Konstante der Kristalle ist die gleiche wie in der Verbindung, die nach Beispiel 1 erhalten wurde.
Beispiel 3
3-Acetyl-2-[2-(3-dimethylaminopropoxy) phenyl] benzothia-zolin (VerbindungNr. 7)
Zu der Suspension von 0,26 g Natriumhydrid in wasserfreiem DMF gibt man tropfenweise in einer Stickstoffatmosphäre bei Zimmertemperatur die Lösung von 1,36 g 3-Acet-yl-2-(2-hydroxyphenyl)benzothiazolin in wasserfreiem DMF. Nach der Zugabe wird die Reaktionsmischung 20 Minuten lang bei Zimmertemperatur gerührt. Zu der Reaktionsmischung gibt man die Lösung von 0,61 g 3-Dimethylami-nopropylchlorid in 10 ml wasserfreiem DMF. Die Reaktionsmischung wird zwei Stunden lang bei 80 °C gerührt und mit n-Hexane nach dem Abkühlen gewaschen und in Eiswasser gegossen. Das abgetrennte Öl wird mit Ethylacetat extrahiert. Die organische Schicht wird mit N-NaOH-Lösung gewaschen, Wasser und gesättigter Natriumchloridlösung, in der angegebenen Reihenfolge und über wasserfreiem Magnesiumsulfat getrocknet. Ethylacetat wird im Vakuum entfernt, um 1,0 g (56 %) der Titelverbindung zu ergeben.
Physikalische Daten sind in Tabelle I gezeigt. Die Verbindungen, die in Tabellen I, II und III gezeigt sind, werden durch ein ähnliches Verfahren, das weiter oben beschrieben ist, unter Verwendung entsprechender Benzo-thiazolin-Derivate und substituierten Aminoalkyl-Haloge-nide hergestellt.
Beispiel 4
3-Acetyl-2[2-(3-aminopropoxy) phenyl] benzothiazolin-Hy-drochlorid (Verbindung Nr. 1 )
Durch Einsetzen von 0,83 g 3-Aminopropylbromid für

3-Dimethylaminopropylchlorid in dem Verfahren von Beispiel 3 erhält man 1,2 g der Titelverbindung.

5 Beispiel 5
3-Acetyl-2-[ 2- ( 3-Diethylaminopropoxy ) phenylJbenzothiazo-lin-Hydrochlorid (Verbindung Nr. 10)
Zu der Lösung von 3,48 g 3-Acetyl-2-[2-(3-chlorprop-oxy)- phenyljbenzothiazolin in 20 ml Ethanol gibt man 10,3 io ml Diethylamin und die Mischung wird 2 Stunden lang am Rückfluss erhitzt. Nach Entfernung von Ethanol und der überschüssigen Menge von Diethylamin im Vakuum gibt man Ethylacetat und Wasser zu dem Rückstand. Die organische Schicht wird mit N-Chlorwasserstoffsäure extrahiert, die 15 saure Schicht wird mit N-Natriumhydroxidlösung alkalisch gemacht und mit Ethylacetat extrahiert. Die organische Schicht wird mit gesättigter Natriumchloridlösung gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wird im Vakuum entfernt und das ent-20 standene Öl wird in Methanol gelöst. Zu der Lösung gibt man Chlorwasserstoffsäure in Ethylacetat, um 3,4 g (80%) der Titelverbindung zu ergeben.
Die physikalischen Daten sind in Tabelle I gezeigt.
Die Verbindungen, gezeigt in den Tabellen I, II, III und 25 VIII, werden durch eine ähnliche Methode, die weiter oben beschrieben ist, unter Verwendung entsprechender Benzo-thiazolin-Derivate und Amin-Derivate hergestellt.
Beispiel 6
30 3-Acetyl-2-[2-(3-methylaminopropoxy) phenyl]benzothiazo-lin-Fumarat (VerbindungNr. 2)
Zu der Lösung von 6,55 g 3-Acetyl-2-[2-(3-oxopropoxy) phenyljbenzothiazolin in 100 ml Methanol gibt man 8,10 g Methylamin-Hydrochlorid und 10,0 g eines molekularen Sie-35 bes (3A), zerbrochen in kleine Teilchen. Zu der Suspension gibt man 1,26 g Natriumcyanoborhydrid und die Mischung wird eine Stunde lang bei Zimmertemperatur gerührt. 2N-Chlorwassersäure wird zu der Reaktionsmischung hinzugegeben und Methanol wird im Vakuum entfernt. Die wässrige to Schicht wird mit Ethylacetat gewaschen und mit 2N-Natri-umhydroxidlösung alkalisch gemacht und mit Ethylacetat extrahiert. Die organische Schicht wird mit gesättigter Natriumchloridlösung gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wird im Vakuum « entfernt und das entstandene Öl wird in Methanol gelöst. Zu der Lösung werden Fumarsäure und Ethylacetat zugefügt, um 4,9 g (65%) der Titelverbindung zu ergeben.

3-Acetyl-2-[2-[5-(N-cyclohexyl -N- methylamino)-pentyloxyl -5- methoxyphenyl]benzothiazolin-Fumarat (Verbindung Nr. 21)
Durch Einsetzen von 4,50 g 3-Acetyl-2-[2-(5-brompent-55 yloxy) -5- methoxyphenyljbenzothiazolin für 3-Acetyl -2-[2-(3- chlorpropoxy)phenyl] benzothiazolin und 3,40 g N-Meth-ylcyclohexylamin für Diethylamin im Verfahren von Beispiel 5 erhält man 4,2 g (70%) der Titelverbindung.

Beispiel 8
3-Acetyl-2-[ 3-[3-(N-cyclohexyl -N- methylamino)propoxyl] -4- hydroxy-phenyl] benzothiazolin-Fumarat (Verbindung Nr. 37)
654001
3,64 g 3-Acetyl-2-[3-chlorpropoxy) -4- hydroxyphenyl] benzothiazolin und 3,40 g N-Methylcyclohexylamin werden bei 100 °C eine Stunde lang gerührt. Nach Abkühlen auf Zimmertemperatur wird die Reaktionslösung in 50 ml Chloroform gelöst, die Lösung wird mit N-Chlorwasserstoffsäure, N-Natriumhydroxidlösung und gesättigter Natriumchloridlösung in der angegebenen Reihenfolge gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Chloroform wird im Vakuum entfernt und der Rückstand wird durch Silikagel-Kolonnen-Chromatographie gereinigt. Das ölige Produkt wird in 20 ml Ethylacetat gelöst. Zu der Lösung gibt man 0,71 g Fumarsäure in 3 ml Methanol, um 2,78 g (50%) der Titelverbindung zu ergeben.

Die Verbindungen, gezeigt in Tabelle II und III, werden durch eine ähnliche Methode, wie weiter oben beschrieben ist, unter Verwendung entsprechender Benzothiazolin-Derivate und Amin-Derivate hergestellt.
Beispiel 9
3-Acetyl-2-[2-[2-[4-(3,4-dimethoxyphenetyl) -1-piperazin-yljethoxyjphenylbenzothiazolin-Dimaleat (Verbindung Nr. 57)
3,34 g 3-Acetyl-2-[2-(2-chloroethoxy) phenyljbenzothiazolin und 5,01 g 1-(3,4-Dimethoxyphenetyl)piperazin werden bei 110 °C eine Stunde lang gerührt. Nach dem Abkühlen auf Zimmertemperatur wird die Reaktionsmischung in 50 ml Chloroform gelöst, die Lösung wird mit N-Chlorwasserstoff-säure, N-Natriumhydroxidlösung und gesättigter Natriumchloridlösung in der angegebenen Reihenfolge gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Chloroform wird im Vakuum entfernt und das erhaltene Öl wird in 50 ml Ethylacetat gelöst. Zu der Lösung gibt man 2,32 g Maleinsäure in 30 ml Ethylacetat, um 6,24 g (80%) der Titelverbindung zu ergeben.

Die Verbindungen, gezeigt in den Tabellen I, IV, V, VI und VIII, werden durch eine ähnliche Methode, wie weiter oben beschrieben, unter Verwendung entsprechender Benzothiazolin-Derivate und Amin-Derivate hergestellt.
Beispiel 10
3-Acetyl-2-[2-[3-( ethoxycarbonyl) propoxy] phenyl] benzothiazolin (VerbindungNr. 97)
Zu der Suspension von 1,59 g Natriumhydrid in 30 ml wasserfreiem DMF gibt man die Lösung von 8,13g 3-Acetyl-
2-(2-hydroxyphenyl) benzothiazolin in 30 ml wasserfreiem DMF tropfenweise unter Stickstoffatmosphäre bei Zimmertemperatur. Nach der Zugabe wird die Reaktionsmischung 20 Minuten lang bei Zimmertemperatur gerührt. Zu der Reaktionsmischung gibt man die Lösung von 4,95 g Ethyl-4-chlorbutyrat in 15 ml wasserfreiem DMF und rührt 5 Stunden lang bei 70 °C. Nach dem Abkühlen auf Zimmertemperatur wird die Reaktionsmischung in Eiswasser gegossen und mit Ethylacetat extrahiert.
Die organische Schicht wird mit N-Kaliumhydroxidlö-sung, Wasser, N-Chlorwasserstoffsäure und gesättigter Natriumchloridlösung in der angegebenen Reihenfolge gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wird entfernt und das entstandene Öl wird durch Silikagel-Kolonnen-Chromatographie gereinigt, um 8,67 g (75%) der Titelverbindung zu ergeben.

Beispiel 11
3-Acetyl-2-[2-[3-(carboxy) propoxy] phenyl] benzothiazolin ( Verbindung Nr. 96)
Zu der Lösung von 7,71 g 3-Acetyl-2-[2-[3-(ethoxycar-
bonyl)-propoxyl] phenyl] benzothiazolin in 50 ml Methanol gibt man 2N-Natriumhydroxidlösung und die Reaktionsmischung wird 2 Stunden lang bei Zimmertemperatur gerührt. Methanol wird im Vakuum entfernt. Der Rückstand wird mit 5 2N-Chlorwasserstoffsäure angesäuert, um 5,72 g (80%) der Titelverbindung zu ergeben.

Beispiel 12
3-Acetyl-2-[2-[3-( morpholinocarbonyl) propoxy ] phenyl] benzothiazolin (VerbindungNr. 100)
Zu der Lösung von 1,79 g 3-Acetyl-2-[2-(carboxy)prop-15 oxy] phenyl] benzothiazolin und 0,51 g Triethylamin in 20 ml wasserfreiem Methylenchlorid gibt man tropfenweise unter Rühren bei -13 °C0,68 g Isobutylchlorformiat und rührt zusätzlich 10 Minuten bei der gleichen Temperatur. Zu der Lösung gibt man tropfenweise 0,44 g Morpholin in 5 ml Methyl-20 enchlorid.
Nach der Zugabe wird die Reaktionsmischung eine Stunde lang unter Kühlung mit Eiswasser und eine Stunde lang bei Zimmertemperatur gerührt. Die Reaktionsmischung wird mit 5%iger Zitronensäurelösung, Wasser, N-Kalium-25 hydroxidlösung und gesättigter Natriumchloridlösung in der genannten Reihenfolge gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wird im Vakuum entfernt, um 1,60 g (75%) der Titelverbindung zu geben.

Beispiel 13
35 3-Acetyl-2-[5-methoxy-2-[4-[N-methyl-N- (3,4,5-trimeth-oxaphenethyl) amino] butoxy] phenyl] benzothiazolin-Hy-drochlorid
Zu einer Suspension von 60% Natriumhydrid (4,4 g) in 15 ml wasserfreiem Dimethylformamid gibt man tropfen-4o weise unter Eiskühlung in einer Stickstoffatmosphäre eine Lösung von 3-Acetyl -2- (2-hydroxy -5- methoxyphenyl) benzothiazolin (30,1 g) in wasserfreiem Dimethylformamid (50 ml). Die Mischung wird 10 Minuten lang bei der gleichen Temperatur gerührt. Man gibt tropfenweise 1,4-Dibrombu-45 tan (64,8 g) zu der Mischung und die Reaktionsmischung wird eine Stunde lang bei 50 °C gerührt und in Eiswasser gegossen. Man extrahiert das Produkt mit Ether. Die organische Schicht wird mit normaler Natriumhydroxidlösung gewaschen. Man trocknet die Lösung über wasserfreiem Maso gnesiumsulfat und konzentriert im Vakuum und erhält 24,8 g (65%) 1-Acetyl -2- [2-(4-brombutoxy) -5- methoxyphenyl] benzothiazolin mit einem Schmelzpunkt von 114,5-116 °C.
Zu einer Lösung von l-Acetyl-2-[2-(4-brombutoxy) -5-methoxyphenyl] benzothiazolin, hergestellt nach dem obigen 55 Verfahren, und N-Methyl -3,4,5- trimethoxyphenetylamin (22,0 g) in Ethanol (68 g) gibt man Triethylamin (9,9 g) und die Mischung wird 3 Stunden lang am Rückfluss erhitzt. Man giesst die Reaktionsmischung in Eiswasser und extrahiert mit Chloroform. Die organische Schicht wird mit 2-normaler 60 Chlorwasserstoffsäure und einer gesättigten Natriumchloridlösung gewaschen. Die Lösung wird über wasserfreiem Magnesiumsulfat getrocknet und im Vakuum konzentriert und man erhält 26,1 g (65%) der Titelverbindung.

Pharmakologische Aktivitäten
Calcium-Antagonisten haben nicht nur potentielle heilsame Wirkungen bei der Behandlung von vielen Krankheiten, sie dienen auch als wertvolle Forschungswerkzeuge, um die Exzitations-Kontraktions-Kupplung in verschiedenen Muskeltypen zu klären. (A. Fleckenstein, Ann. Rev. Pharmcol., 17,149-166,1977). Daher wurde die Calcium-antagonistische Aktivität der Verbindungen dieser Erfindung untersucht.

Pharmakologischer Test I
Die Wirkungspotentiale auf die glatten Muskeln des Uterus, Teania coli und der Pfortader hängen vom Calcium-Ion ab und daher sind diese glatten Muskelpräparate für die Klassierung von Calcium-Antagonisten nützlich. Die Calcium-antagonistische Aktivität der Verbindungen mit der Methode unter Verwendung des Meerschweinchen-Teania coli-Präpa-rates wurde gemessen.
Isolierte Meerschweinchen-Teania coli wurden in 20 ml eines Organ-Bades mit Krebs-Lösung bei 32 °C suspendiert und man leitete 5% Kohlenstoffdioxid in Sauerstoff in Blasenform hindurch. Nach Eintreten des Gleichgewichtes wurde der Muskel mit Ca ++-freier Krebs-Lösung gewaschen und wenn sich der Muskel auf das ursprüngliche Niveau entspannt hatte, wurde er in einer Ca+ +-freien-hohen-K-Krebs-Lösung suspensiert.
Man setzte den Muskel den Testverbindungen 5 Minuten lang aus, bevor man CaCl2 hinzugab, und die Konzentration, hervorgerufen durch CaCl2 (3 x 10_4M), wurde isotonisch aufgezeichnet. Die Calcium-antagonistische Aktivität wurde durch die Konzentration der Testverbindung dargestellt, welche eine 50% Inhibition von durch Ca ++-hervorgerufene Kontraktion (IC50) auslöste.
Wie in Tabelle XI gezeigt ist, haben die Verbindungen dieser Erfindung eine Calcium-antagonistische Aktivität.
Blutplättchen spielen eine wichtige Rolle, nicht nur bei Hämostasen, sondern auch bei Thrombosen. Die Hyperag-gregabilität der Plättchen führt zu einer Erhöhung der Anzahl der zirkulierenden Plättchenaggregate, was zur Entwicklung von Arrythmias des Herzens, Herzversagen oder Myokardinfarkt beiträgt. Diese kardiovaskulären Krankheiten können durch Inhibition der Plättchenaggregation verhindert werden. Der Einfluss der Testverbindungen auf die Plättchenaggregation in vitro wurde daher zusammengestellt und man fand, dass sie eine Anti-Aggregations-Aktivität haben.
Pharmakologischer Test II
Man erhielt Blut aus einem anästhesierten Kaninchen unter Verwendung von 0,1 Volumen von 3,8% Natriumzitrat als Antikoagulans. Plättchenreiches Plasma (PRP) wurde durch Zentrifugieren bei 650 Umdrehungen pro Minute 10 Minuten lang bei Zimmertemperatur isoliert. Nach Vorinkubation von PRP (0,25 ml) mit verschiedenen Konzentrationen der Testverbindungen (14 nl) für 1 Minute bei 37 °C gab man Kollagen (3 jig/ml: endgültige Konzentration) oder ADP (3 |iM: endgültige Konzentration) hinzu, um die Aggregation zu induzieren und die Aggregationsprofile wurden mit einem RIKADENKI-6-Kanal-Aggregometer überwacht. Das Kontrollexperiment enthielt Kochsalzlösung anstelle der Testverbindung.
Die Anti-Aggregationaktivität wurde durch die Konzentration der Testverbindung dargestellt, welche eine 50% Inhibition der Kontrollantwort auslöste.
Wie in der Tabelle XII gezeigt ist, hatten die erfindungsge-mässen Verbindungen eine Anti-Aggregationsaktivität.

Toxizitäts-Test
Die akute Toxizität der Verbindungen dieser Erfindung ist in Tabelle XIII gezeigt.
(Tiere)
Männliche Ratten der Art ddy-SLC (4 Wochen alt, Gewicht 19-21 g) gab man in einen Brutraum von konstanter Temperatur und Feuchtigkeit (23 ± 1 °C, 55 ± 5%) und fütterte sie uneingeschränkt mit einer Kügelchen-Diät und Was-30 ser nach Belieben eine Woche lang. Ratten, die ein normales Wachstum zeigten, wurden für den Test ausgewählt.
(Methode der Verabreichung)
Die Testverbindungen wurden in einer 0,5%igen Tragant-Suspension suspendiert und oral in einer Dosis von 35 0,5 ml/20 g Körpergewicht verabreicht.

Die Verbindungen können entweder oral oder parenteral verabreicht werden. Die Dosierungsformen sind Tabletten, Kapseln, Körnchen, Pulver, Suppositorien, Injektionen usw. Die Dosis wird angepasst, abhängig von dem Symptom, der so Dosierungsform usw., aber üblicherweise beträgt die tägliche Dosis 1 bis 5.000 mg, vorzugsweise 1 bis 1.000 mg, in einer oder wenigen geteilten Dosen.

- 654 001PATENTANSPRÜCHE 1.
