Den foreliggende opfindelse angår en analogifremgangsmåde til fremstilling af .hidtil ukendte aminderivater .ai di-Q-(n-høiere alkyl- og alkenyl)-glyceroler eller'deres farma ceutisk acceptable syreadditionssalte> hvilke forbindelser er nyttige til bekæmpelse af virale infektioner i pattedyr. Af særlig interesse er 1,3-di-0-(n-hexadecyl)-2-0-(3-aminopropyl)-glycerol og dens farmaceutisk acceptable syreadditionssalte.
Virusinfektioner, som angriber pattedyr, herunder mennesker, er normalt smitsomme sygdomme, som kan forårsage stor menneskelig lidelse og økonomisk tab. Uheldigvis er opdagelsen af antivirale forbindelser meget mere kompliceret og besværlig end opdagelsen af 2 149022 antibakterielle og antifungale midler. Dette skyldes delvis den tætte strukturelle lighed mellem vira og strukturen af visse essentielle cellulære forbindelser, såsom ribonucleinsyre og desoxyribo-nucleinsyre. Imidlertid er beskrevet adskillige ikke-virale "antivirale forbindelser", dvs.forbindelser "som kan danne enten en beskyttende eller terapeutisk virkning til en klar påviselig fordel for den virusangrebne vært, eller et materiale, som væsentligt kan forøge antistofdannelsen, forbedre antistofaktiviteten, forbedre ikke-speci-fik modstand, bevirke hurtigere helbredelse eller hæmme symptomer", [Herrman et.al., Proc. Soc. Exptl. Biol. Med., 103, 625, (I960)]. Listen af angivne antivirale midler omfatter for at nævne et fåtal, interferon og syntetiske materialer, såsom amantadinhydrochlorid, pyrimidiner, biguanider, guanidin, pteridiner og methisazon. På grund af det temmelig snævre område af virale infektioner, der kan behandles med hver af de i handelen tilgængelige antivirale midler på nuværende tidspunkt, er nye syntetiske antivirale midler altid velkomne som potentiel værdifuld forøgelse af den medicinske teknologis hjælpestoffer.
Pattedyrs celler producerer som svar på virale infektioner et stof, der muliggør, at cellerne kan modstå formering af et antal vira. De virushæmmende eller virusinterfererende stoffer kaldes "interferoner". Interferonerne er glycoproteiner, der kan adskille sig i deres fysisk-kemiske egenskaber, men som alle udøver samme biologiske egenskaber, nemlig hæmning af et stort antal ikke-beslæg-tede vira, ikke har toksisk eller anden skadelig effekt på cellerne og er artsspecifikke (Lockart, Frontiers of Biology, Vol. 2, "Interferons", udgivet af Finter, W.B. Saunders Co., Philadelphia, 1966, side 19-20).
Ingen praktisk, økonomisk fremgangsmåde er endnu blevet udviklet til fremstilling af exogene interferoner til klinisk rutinebrug overfor virale infektioner. Et alternativ til fremstilling af interferoner har derfor været forsøgt, hvilket omfatter administrering tildet dyr, som skal beskyttes eller behandles, af et ikke-viralt stof, der stimulerer eller inducerer frembringelse af interferon i cellerne. Interferon fremstillet på denne måde betegnes "endogen" interferon.

De hidtil ukendte forbindelser fremstillet ved fremgangsmåden ifølge denne opfindelse har formlerne R1-0-CH- I Δ o CH-O-Y-NHR i 2 1 R -0-CH2 eller 3 chjO-y-^nhr·3

Forbindelserne med formlen I eller ll udyiser antf-yiral virkning overfor en stor mængde vira in vivo i pattedyr og in vitro i cellekulturer fra pattedyr. I det mindste en væsentlig mængde af denne virkning resulterer fra de nævnte forbindelsers evne til at inducere interferonproduktion i cellerne, d.v.s. endogen interferon.
Med "farmaceutisk acceptable" syreadditionssalte menes de salte, som er ikke-toksiske" ved de indgivne doser. De farmaceutisk acceptable syreadditionssalte, der kan anvendes, omfatter sådanne vandopløselige og vanduopløselige salte, såsom hydrochlorider, hydrobromider, fosfater, nitrater, sulfater, acetater, hexafluor-fosfater, citrater, gluconater, benzoater, propionater, butyrater, sulfosalicylater, maleater, laurater, malater, fumarater, succina-ter, oxalater, tartrater, amsonater (4,4'-diamino- stilben-2,2'-di-sulfonater), pamoater (l,l'-methylen-bis-2-hydroxy-3-naphthoater), stearater, 3-hydroxy-2-naphthoater, p-toluen-sulfonater, methansul-fonater, lactater og suraminsalte.

Syreadditions salte af baserne med formlerne I eller II kan fremstilles ved konventionelle fremgangsmåder, såsom ved blanding af aminforbindelsen i et passende opløsningsmiddel med den ønskede syre og udvinding af saltet ved fordampning eller udfældning efter tilsætning af et opløsningsmiddel, hvori saltet er uopløseligt. Hydrochloridsalte kan let fremstilles ved genindvinding af hydrogenchlorid gennem en opløsning af aminforbindelsen i et organisk opløsningsmiddel. Som det kan ses af de følgende eksempler har mange af de isolerede hydrochlorid- eller di-hydrochloridsalte af baserne med formlerne I eller II tendens til at indeholde en betydelig mængde vand. Det vides ikke, hvorvidt dette 7 149022 observerede "fangne" vand er vilkårligt indesluttet under krystallisationen eller svarer til dannelse af virkelige molekylære hydrater eller stammer fra tilstedeværelsen af et andet fænomen. I hvert tilfælde kan saltene indeholdende "fanget" vand let præpareres og administreres uden forudgående dehydratisering.
Udgangsmaterialerne l,2-di-0-(n-højere alkyl)-glyceroler kan fremstilles ved den af Kates, M. et al., Biochemistry, 2_, 394 (1963) angivne metode. Udgangsmaterialerne 1,3-di-O-(n-højere alkyl)-glyceroler kan fremstilles ved den af Damico, R., et al. J, Lipid Res., <?, 63 (1967) angivne metode. Udgangsmaterialerne 1,2- og 1,3-di-O-(n-højere alkenyl)-glyceroler kan fremstilles ved den af Bauman, W.J. og Mangold, Η. K., j. Org. Chem., 31, 498 (1966) angivne metode.
Den anti-virale virkning af forbindelserne fremstillet ifølge opfindelsen blev bestemt ved anvendelse af to uafhængige fremgangsmåder. Ved den første blev prøveforbindelsen administreret til mus ad intra-peritoneal vej 18 til 24 timer før indgift af en letal dosis af encephalomyocarditis (EMC) virus. Overlevelsesdata er opført lo dage efter indgift af virus og sammenlignet med data fra ubeskyttede dyr. Fremgangsmåden, ved hvilken medikamentet indgives 18 til 24 timer før og på et væsentligt adskilt sted fra virusinjektionen, er beregnet på at eliminere lokale effekter mellem medikament og virus og identificere udelukkende de forbindelser, som giver et systemisk anti-viralt svar.
Ved den anden fremgangsmåde behandles monolag af humane nasale polypceller, dyrket på mikrotiterplader med prøveforbindelsen ca.
18 timer før behandling med en letal dosis af vesikular stomatitis-virus (VSV). Prøveforbindelsen vaskes bort fra monolaget før virusbehandling. Kulturvæske ekstraheres fra pladerne efter en inkubationsperiode, titreres for mængden af infectiøs virus tilstedeværende i mikrotiterplader af L-929 muse-fibroblaster. Sammenligning gøres med virusdata fra kulturvæske ekstraheret fra ubeskyttede polypceller.
Yderligere blev mange af foibindelseme fremstillet ifølge opfindelsen afprøvet med hensyn til deres evne til at forøge den kendte anti-virale virkning af polyinosin:polycytidylinsyre. Endelig blev visse af forbindelserne afprøvet med hensyn til deres evne til at inducere cirkulerende interferon i mus efter parenteral administrering ved anvendelse af den af Hoffman, W.W., et al., Antimicrobial Agents and Chemotherapy, 3, 498-501 (1973) beskrevne fremgangsmåde.

Parenteral, topisk eller intranasal administrering af de i det foregående nævnte beskrevne aminer til pattedyr· før udsættelse af pattedyret for en infectiøs virus giver hurtig modstand mod virusen. Fortrinsvis bør administreringen finde sted fra ca. 2 til ca. 1 dag før udsættelse for virus, men dette vil variere noget med den specielle dyreart og med den specielle infec-tiøse virus.
Ved indgift af de pågældende forbindelser anvendes de lettest og mest økonomisk i dispergeret form i en acceptabel bærer.
Når det siges, at forbindelserne er dispergeret, betyder det, at partiklerne kan være af molekulær størrelse og holdes i sand opløsning i et passende opløsningsmiddel, eller at partiklerne kan.være af colloidal størrelse og dispergeret i en væskefase i form af en suspension eller en emulsion. Betegnelsen "dispergeret" betyder også, at partiklerne kan være blandet med og spredt i en fast bærer, således at blandingen er i form af et pulver eller støv.
Denne betegnelse omfatter også blandinger, der egner sig til anvendelse som sprøjtemidler, omfattende opløsninger, suspensioner eller emulsioner af forbindelserne fremstillet ifølge opfindelsen.
Administreret parenteralt (subcutant, intramuskulært, intra-peritonealt) anvendes forbindelserne i en mængde på fra ca. 1 mg/kg legemsævgt til ca. 200 mg/kg legemsvægt. Det foretrukne område er fra ca. 5 mg/kg til ca. 100 mg/kg legemsvægt, og det mest foretrukne område fra ca. 5 mg til ca. 50 mg/kg legemsvægt. Dosis afhænger naturligvis af det pattedyr, som skal behandles, og den specielle aminforbindelse, som er anvendt, og bestemmes af den person, som er ansvarlig for administreringen. Sædvanligvis administreres små doser i begyndelsen med gradvis . forøgelse af dosis, indtil det optimale dosisniveau er bestemt for det specielle subjekt, som er under behandling.
' Ved anvendelse af den intranasale administrationsvej kan anvendes en hvilken som helst praktisk fremgangsmåde til kontakt mellem det anti-virale middel og dyrets åndedrætsveje. Effektive fremgangsmåder omfatter administrering af midlet ved hjælp af intranasale eller nasapharyngeale dråber og ved inhalation ved hjælp af en nebulizer eller en aerosol. Sådanne administreringsfremgangsmåder er af praktisk betydning, fordi de tilveje-bringer en let, sikker og effektiv metode. Til intranasal administrering af midlet, sædvanligvis i en 9 149022 acceptabel bærer, er en koncentration af midlet mellem 1,0 mg/ml og 100 mg/ml tilfredsstillende. Koncentrationer i området på fra ca. 30 til 50 mg/ml muliggør administrering af passende volumen .
Til topisk anvendelse benyttes de anti-virale forbindelser mest passende i en bærer for at muliggøre let og sikker anvendelse og bedre absorption. Også her er koncentrationer i området fra ca. 1,0 mg/ml til ca. 250 mg/ml tilfredsstillende. Generelt vil man ved de to foregående metoder anvende en dosis i området fra ca. 1,0 mg/kg til ca. 250 mg/kg legemsvægt og fortrinsvis fra ca.
5,0 mg /kg til ca. 50 mg/kg.
Opfindelsen forklares nærmere ved hjælp af følgende eksempler.
Eksempel 1 1,3-Di-0-(n-hexadecyl)-2-0-(3-aminopropyl)-glycerol-hydrochlorid.
A. 1,3-Di-(n-hexadecyl)-2-0-(2-cyanoethyl)-glycerol.
En blanding af l,3-di-0-(n-hexadecyl)-glycerol (80 g, 148 mmol) acrylonitril (1,49 kg, 28,1 mol) og vandig 2N natriumhydroxid (1,2 1) blev opvarmet til 50°C. Tetrabutylammoniumhydroxid (19,2 g 40 vægtprocent vandig opløsning, 29,15 mmol) blev langsomt tilsat, hvorved temperaturen af den exotherme reaktionsblanding steg til ca 80 til 90°C. Reaktionsblandingen blev så omrørt i 20 min. uden ydre opvarmning, efterfulgt af afkøling til 20°C og tilsætning af vand (1,0 1). Et fast materiale, en blanding af ikke-reageret og cyanoethyleret l,3-di-0-(n-hexadecyl)-glycerol blev isoleret og behandlet igen med frisk acrylonitril (1,49 kg, 28,1 mol), vandig 2N natriumhydroxid (1,2 1) og tetrabutylammoniumhydroxid (19,2 g 40 vægtfs vandig opløsning, 29,15 mmol) i 20 min. under omrøring ved 50°C, efterfulgt af afkøling og tilsætning af vand (1,0 1). Den resulterende l,3-di-0-(n-hexadecyl)-2-0-(2-cyanoethyl)-glycerol i form af et fast stof blev filtreret, vasket med vand, derefter methanol og acetonitril og tørret [82 g, 93% udbytte, smp. 45-46°C

En blanding af l,3-di-0-(n-hexadecyl)-2-0-(2-cyanoethyl)-glycerol (20,5 g, 35,5 mmol), tetrahydrofuran (200ml), ethanol (10 ml) og Raneynikkelkatalysator 3 g, blev mættet med ammoniakgas ved 0-5°C og derpå hydrogeneret (3,5 ato) i en Paar-hydrogenator i 3 timer ved stuetemperatur. Blandingen blev så filtreret, katalysatoren vasket med tetrahydrofuran (50 ml) og det totale filtrat fordampet i vakuum til en olie. Denne fremgangsmåde blev gentaget 3 gange mere med friske reaktanter og katalysator til dannelse af en totalmængde på 77 g olie. Olien blev opløst i ether (500 ml) og opløsningen vasket med 2 vægt% vandig ammoniumhydroxidopløsning (500 ml), tørret med MgSO^, filtreret og inddampet i vakuum til dannelse af et fast stof. Eksempel 8 1,3-Di-O-(n-hexadecyl)-2-0-(4-aminobutvl)-glycerol—hydrochlorid.
A. 1,3-Di-O-(n-hexadecyl)-2-0-(3-hydroxypropyl)-glycerol.
Boranmethylsulfid (BMS) kompleks (6,5 ml, 68,5 mmol) blev sat ved 0-5°C til en opløsning af l,3-di-0-(n-hexadecyl)-2-0-allyl-glyce-rol (10,82 g, 18,6 mmol, fremstillet som angivet i eksempel 11A) i hexan (190 ml), og den resulterende opløsning blev omrørt i 3 timer ved stuetemperatur. Reaktionsopløsningen blev så igen afkølet til 0-5°C, og ethanol (17,3 ml) blev dråbevis tilsat til spaltning af resterende BMS. Reaktionsopløsningen blev så behandlet med vandig 3N natriumhydroxid (13 ml) og 30 vægt% vandig hydrogenperoxid (11 ml) og omrørt i 16 timer under tilbagesvaling, afkølet og udhældt i isvand indeholdende natriumbisulfit. Isvandopløsningen blev omrørt, indtil den gav negativ stivelses-iodidprøve for peroxider, og derpå ekstraheret med ether (3 x 200 ml). B. 1,3-Di-O-(n-hexadecyl)-2-0-[3-(p-tosyloxy)propyl]-glycerol.
1,3-Di-O-(n-hexadecyl-2-O-(3-hydroxypropyl)-glycerol (8,0 g, 13,4 mmol) blev sat ved 10°C til en opløsning af p-toluensulfonylchlorid (5,25 g, 27,5 mmol) og pyridin (10 ml i 200 ml methylenchlorid), og blandingen blev omrørt i 60 timer ved stuetemperatur. Vand (200 ml) blev så tilsat, methylenchlorid og den vandige fase blev adskilt, sidstnævnte blev ekstraheret med methylenchlorid (2 x 150 ml) . 1,3-Di-0-(n-hexadecyl)-2-0-[3-(p-tosyloxy)propyl]-glycerol (3,0 g, 4,0 mmol) blev opløst i en opløsning af natriumcyanid (0,5 g, 10 mmol) i Ν,Ν-dimethylformamid (50 ml) og den resulterende opløsning blev omrørt i 16 timer ved 80°C, afkølet, fortyndet med vand (100 ml) og ekstraheret med ether (3 x 150 ml). Den samlede etherekstrakt blev vasket med IN saltsyre (3 x 75 ml), derpå med mættet vandig natriumhydrogencarbonatopløsning (3 x 75 ml), så vand (75 ml) og til slut mættet vandig natriumchloridopløsning (75 ml) og derpå tørret (MgSO^), filtreret og inddampet i vakuum til dannelse af et voksagtigt fast stof, der blev anvendt til næste trin uden yderligere rensning [2,0 g, 83%'s udbytte, ir (CHCl^) 2250 cm-1].

Lithiumaluminiumhvdrid (800 mg, 21 mmol) blev sat til en opløsning af l,3-di-0-(n-hexadecyl)-2-0-(3-cyanopropyl)-glycerol (2,0 g, 3,3 mmmol) i ether (100 ml), og blandingen blev omrørt i 60 timer ved stuetemperatur. Tilstrækkeligt vand til afslutning af reaktionen blev forsigtigt tilsat, efterfulgt af yderligere 100 ml vand. Den resulterende blanding blev omrørt i yderligere 1 time ved stuetemperatur og derpå ekstraheret med ether (3 x 100 ml). Eksempel 9 1,2-Di-O-(n-hexadecyl)-3-0-(3-aminomethylbenzyl)-glycerol—hydro-chlorid.
A. l,2-D±-0—(n—hexadecyl)-3-0-(3-cyanobenzyl)-glycerol.
Natriumhydrid (1,056 g af 50 vægt% mineraloliedispersion, - 22 .mmol) blev sat til en opløsning af 1,2-di-O-(n-hexadecyl)-glycerol (9,73 g, 18 mmol) i tetrahydrofuran (150 ml), og den resulterende opløsning blev omrørt i 20 min. ved stuetemperatur under nitrogen. m-Cyanobenzylbromid (4,0 g, 20 mmol) blev tilsat, og.reaktionsblandingen omrørt natten over ved stuetemperatur under nitrogen. Vand (200 ml) blev så tilsat forsigtigt, og den resulterende blanding ekstraheret med ethylacetat (3 x 150 ml). Det samlede ethylacetatekstrakt blev tørret over MgSO^, filtreret og inddampet i vakuum til en olie (12 g), som blev renset ved chromatografi på . silicagel (eluering med benzenzhexan) [8,0 g, 68%'s udbytte, olie, ir (CHCI3) 2230 cm"1.

En opløsning af 1,2-di-O-(n-hexadecyl)-3-0-(3-cyanobenzyl)-glycerol (1,0 g, 1,5 mmol) i ether (10 ml) blev langsomt under nitrogen sat til en suspension af lithiumaluminiumhydrid (0,057 g, 1,5 mmol) i ether (40 ml)., og den resulterende blanding blev omrørt i 1 time under tilbagesvaling og. nitrogen og derpå afkølet. 50 ml vand blev forsigtigt tilsat, og blandingen blev ekstraheret med ether (3 x 50 ml). Den samlede etherekstrakt blev tørret over MgSO^, filtreret og inddampet i vakuum til en olie, som blev renset ved chromatografi på silicagel (eluering med benzen:ethanol) og derpå opløst i ethylacetat. A. 1/2-Di-O-(n-hexadecyl)-3-0-(p-tosyl)-glycerol.
På lignende måde som beskrevet i eksempel 8B blev den angivne forbindelse fremstillet ved reaktion mellem l,2-di-0-(n-hexadecyl)-glycerol og p-toluensulfonylchlorid. Rensning skete ved omkrystallisation med ethylacetat [smp. 53-55°C, ir (CHClg) 1360 og 1180 cm .
B. 1,2-Di-O-(n-hexadecyl)-3-0-(4-cyanophenyl)-glycerol.
En blanding af l,2-di-0-(n-hexadecyl)-3-0-(p-tosyl)-glycerol (1,4 g, 2,0 mmol), natrium-4-cyanophenolat (0,5 g, 3,5 mmol) og xylen (100 ml) blev omrørt i 16 timer under tilbagesvaling. Da reaktionen endnu ikke var fuldendt, blev xylenet fjernet ved destillation og erstattet med Ν,Ν-dimethylformamid (100 ml) og den resulterende opløsning blev omrørt i yderligere 16 timer ved 150°C. Reaktionsopløsningen blev så afkølet, fortyndet med vand (100 ml) og ekstraheret med ether (2 x 100 ml). Den samlede ether-ekstrakt blev vasket, først med 3N saltsyre (100 ml), så med 10vægt% - vandig natriumhydrogencarbonatopløsning (100 ml) og derpå vand (100 ml), tørret over MgSO^, filtreret og inddampet i vakuum til en olie, som blev renset ved chromatografi på silicagel (eluering med benzen) [0,65 g, 50%'s udbytte, smp. 53-55°C, ir (CHCl^) 2210 cm"1].
C. Titelforbindelsen.
1.2- Di-O-(-n-hexadecyl)-3-0-(4-cyanophenol)-glycerol (0,60 g, 0,93 mmol) blev sat til en suspension af lithiumaluminiumhydrid (0,3 g, 7,9 mmol) i ether (25 ml), og den resulterende blanding blev omrørt i 30 min. ved stuetemperatur, vand (25 ml) blev så forsigtigt tilsat, etherfasen og den vandige fase blev adskilt, og sidstnævnte blev ekstraheret med ether (3 x 25 ml) og ethylacetat (25 ml). De 5 organiske ekstrakter blev samlet, tørret over MgSO^, filtreret og inddampet i vakuum til en olie, som blev opløst i ether. A. 1,2-Di-O-(n-hexadecyl)-3-0-[2-(p-tosyloxy)propyl]-glycerol.
På lignende måde som beskrevet i eksempel 8 A og B blev 1,2-di-0-(n-hexadecyl)-3-0-allyl-glycerol bragt til reaktion med BMS, og de resulterende 2-hydroxypropyl- og 3-hydroxypropvlforbindelser omdannet til deres tilsvarende tosylater. Adskillelse blev ikke foretaget på dette trin. Blandingen af tosylater blev anvendt direkte ved næste trin.
B. 1,2-Di-O-(n-hexadecyl)-3-0-(2-azidopropyl)-glycerol.
Den resulterende blanding af tosylater (3,0 g, 4,0 mmol) blev opløst i N,N-dimethylacetamid (50 ml) og behandlet med en opløsning af natriumazid (0,326 g, 5,0 mmol) i vand (5 ml) i 16 timer ved 90°C. Reaktionsopløsningen blev så afkølet, fortyndet med vand (200 ml) og ekstraheret med ether (2 x 150 ml). Den samlede ether-ekstrakt blev vasket med vand, tørret over MgSO., filtreret og ind- ^ —i dampet i vakuum til en olie (2 g, 81%'s udbytte, ir (CHClg) 2100 cm , som var en blanding af 2-azidopropyl- og 3-azido propylforbindelser, som blev anvendt uden yderligere rensning til næste trin.
C. Titelforbindelsen.
Den resulterende blanding af azider (2 g, 3,2 mmol) blev opløst i ether (100 ml), behandlet med lithiumaluminiumhydrid (0,4 g, 10,5 mmol), og henstod under omrøring i 2 timer ved stuetemperatur. Overskydende hydrid blev ødelagt ved forsigtig tilsætning af ethanol (10 ml) og vand (150 ml), og blandingen blev så ekstraheret med ether (2 x 100 ml). Den samlede etherekstrakt blev tørret over MgSO^, filtreret og inddampet i vakuum til en olie (1,8 g), som blev renset ved chromatografi på silicagel (eluering med benzen: ethanol) og derpå omdannet til hydrochloridsalt ved opløsning og behandling med hydrogenchloridgas. Saltet blev omkrystalliseret 149022 22 fra ethylacetat (0,21 .g, fast stof indeholde ca. 1/2 mol I^O per mol produkt, 10%'s udbytte, smp. 56-58°C, elementær analyse beregnet: '71,03% C, 12,70% H, 2,18% N, fundet: 71,11% C, 12,91% H, 2,16% N).

På lignende måde som beskrevet i eksempel 27 A blev l,2-di-0-(n-octadecyl)-3-0-(2-hydroxypropyl)-glycerol fremstillet ud fra 1,2-di- 0-(n-octadecyl)-3-0-ally1-glycerol. Det angivne produkt blev fremstillet ud fra l,2-di-0-(n-octadecyl)-3-0-(2-hydroxypropyl)-glycerol på lignende måde som beskrevet i eksempel 27 B-C (fast stof indeholdende ca. 1 mol 1^0 per mol produkt, smp. 65-67°C, elementær analyse beregnet: 71,19% C, 12,80% H, 1,98% N, fundet: 71,12% C, 12,52 % H, 1,92% N).
In vivo aktivitet af l,3-di-0-(n-hexadeyl)-2-0-(3-aminopropyl)-glycerol—hydrochlorid overfor EMC-virus.
Fremstilling af en emulsion skete ved smeltning og blanding af lige dele af den angivne forbindelse, polysorbat 80 og glycerin og påfølgende dispergering af blandingen i varmt vand under voldsom omrøring. Præparatet blev så indstillet til en endelig koncentration på 0,14M natriumchlorid og 0,01M natriumfosfat, pH 7.Yderligere fortyndinger blev gjort med 0,14M natriumchlorid, 0,01M natriumfosfat, pH 7, pufferopløsning.
3 Grupper af 10 hun-albinomus (20-25 g legemsvægt) blev givet 0,5 ml intraperitoneale injektioner indeholdende mængder på 1,5, 5, og 15 mg af den angivne forbindelse/kg legemsvægt. En fjerde kontrolgruppe på 10 mus fik ingen injektion. 18-24 Timer senere fik alle 4 grupper 0,2 ml subeutan injektion indeholdende 20 gange LDgg dosis, som forårsager 50% dødelighed i ubeskyttede mus i 10 dage, af encephalomyocarditis (EMC) virus. Overlevelsesdata blev opsamlet over de næste 10 dage, og den relative overlevelse (Sr) beregnet:

Reduktion af virusudbytte i humane polypceller in vitro med 1,3-di-O-(n-hexadecyl)-2-0-(3-aminopropyl)-glycerol— hydrochlorid.
Et dyrkningsmedium blev fremstillet ved til Eagle's minimale essentielle medium (100 ml) at sætte 100X koncentreret antibiotisk-antimycotisk opløsning (2 ml) 200 mM glutaminopløsning (1 ml) 100X koncentreret opløsning af ikke-essentielle aminosyrer (1 ml) 100 mM natriumpyruvat opløsning (1 ml) og varme-inaktiveret kalvefos ter serum (10%) . Hver fordybning i mikrotiterplader med 96 fordyb-r ninger blev podet med ca. 50.000 menneskelige nasale polypceller suspenderet i 0,2 ml vækstmedium. Pladerne blev så inkuberet i 8-10 dage ved 37°C i en 5% CO2 atmosfære til dannelse af monolag af cellerne.
Efter cellevækstnerioden på 8-10 dage blev sammenløbende monolag på pladerne vasket flere gange med fosfatforpufret saltvand og umiddelbart efter behandlet med 0,2 ml per fordybning af vedligeholdelsesmedium indeholdende 10, 5,0, 1,0, 0,5, 0,1 og 0 yg/ml af den angivne forbindelse. Vedligeholdelsesmediet var identisk med vækstmediet, som er beskrevet i det foregående, med den undtagelse, at mængden af kalvefosterserum er. 5%. Pladerne blev inkuberet i yderligere 18 timer ved 37°C, og monolagene blev derpå vasket flere gange med fosfatforpufret saltvand til fjernelse af den angivne forbindelse, smittet med et materiale indeholdende ca.
1000 gange TCID^q, d.v.s. den dosis, som forårsager 50%'s infektion 1 ubeskyttede kulturer, af vesikular stomatitis-virus (VSV) i en 2 timers adsorptionsperiode ved 37°C, vasket 4 gange med fosfatforpufret saltvand til fjernelse af ikke-adsorberede viruspartikler og forsynet med 0,2 ml per fordybning af vedligeholdelsesmediet. Pladerne blev så inkuberet i 7 timer ved 37°C, og dyrkningsvæsken fra 5-8 reproducerede celler taget fra hver plade, oplagret frosne i prøverør og derpå filtreret for mængden af tilstedeværende infek-tiøs virus i mikrotiterplader af L-929 musefibroblaster. L-929 musekulturerne blev talt mikroskopisk og analyseret ca. 3-4 dage senere med følgende fald i virusudbytte (med hensyn til kontrollen) bestemt for de 5 koncentrationer af den angivne prøveforbindelse:

Virkning af 1,3-Di-O-(n-hexadecyl)-2-0-(3-aminopropyl)-glycerol-hydrochlorid til at inducere cirkulerende interferon.
En blanding af lige vægtmængder af den angivne forbindelse, polysorbat 80 og glycerol blev samlet og derpå homogeniseret i varm 0,14M natriumchlorid indeholdende 0,01M natriumfosfat, pH 7, (PBS).
Den resulterende olie-i-vand-emulsion blev let fortyndet med PBS til administrering.
Hunmus (med legemsvægt 20 til 25 g) blev injiceret intraperi-tonealt (0,5 ml) med en mængde af den i det foregående nævnte fortyndede emulsion indeholdende 25 mg af den angivne forbindelse per kg legemsvægt. 8, 12, 16 og 20 timer efter injektion blev prøver af plasma taget fra 4 mus og samlet. Seriefortyndinger i L-15 (Leibovitz) medium indeholdende 5% kalvefosterserum blev inkuberet i mikrotiterplader natten over ved 37°C på sammenløbne monolag af L-929 musefibroblaster. Monolagene blev så vasket med proteinfrit medium, smittet med 10 gange TCID^Q, d.v.s. den dosis, som forårsager 50%'s infektion i ubeskyttede kulturer, af vesikular stomatitis-virus (VSV) i en adsorptionsperiode på 1 time ved 37°C, vasket, igen behandlet med L-15 medium indeholdende 5% kalvefosterserum og derpå inkuberet igen i 48 timer ved 37°C. L-929 kulturerne blev så talt mikroskopisk for viral cytopatologi og analyseret, og med plasma interferonniveauet blev den reciprokke af plasmafortyndingen, som forårsagede 50%’s beskyttelse af L-929 monolagene, bestemt.
Endnu et forsøg efter den i det foregående nævnte fremgangsmåde med undtagelse af, at musene fik 10 mg af den angivne forbindelse per kg legemsvægt, og prøver af peritoneal vask blev taget fra 4 mus og samlet 6, 9, 12, 15 og 18 timer efter injektion. Prøverne blev taget ved at frilægge peritonealmembranen, injicere 1 ml Hank's afbalancerede saltopløsning indeholdende 100 penicillinenheder/ml og 100 yg streptomycin/ml i peritonealhulen, kort massering af abdomen og påfølgende opsugrdng af peritonealvasken.

Forøgelse af den med polyinosin-’po ly cy tidy linsyre [poly-(I:C)]-inducerede cellulære modstand mod virale infektioner af 1,3-di-O-(n-hexadecyl)-2-0-(3-aminopropyl)-glycerol—hydrochlorid.
Vækstmedium blev fremstillet ved til Eagle's minimum essentielle medium (100 mil at sætte 100X koncentreret antibiotisk-antimycotisk opløsning (2 ml), 200 mM glutaminopløsning (1 ml) og varme-inaktiveret kalvefosterserum (5%). Muse-L-929 fibroblaster blev suspenderet i vækstmedium og hver fordybning i mikrotiterplader med 96 fordybninger blev podet med 0,2 ml af suspensionen indeholdende 20.000 til 30.000 celler. Pladerne blev inkuberet i 2-4 dage ved 37°C i en 5%fs C02 atmosfære til dannelse af monolag af cellerne. Pladerne blev vasket 4 gange med fosfatforpufret saltvand umiddelbart før behandling.
Poly-(I:C) blev fremstillet i koncentrationer på 5,0, 1,0, 0,2 og 0,04 yg/ml i det i det foregående beskrevne medium minus kalveserum. 0,1 ml af hver fortynding blev samlet i et dambrædt-arrangement på L-929 cellemonolagene med 0,1 ml fortyndinger indeholdende 20,0, 4,0, 0,8, 0,16 og 0,032 ug af den angivne forbindelse per ml serumfrit medium. Kontrolfordybninger blev givet enten poly (I:C) eller forbindelsen alene. Pladerne blev inkuberet i 6 timer ved 37°C i en 5% CO^ atmosfære, vasket 4 gange med fosfatforpufret saltvand og forsynet med 0,1 ml per fordybning af vækstmedium indeholdende 2% kalvefosterserum. Efter yderligere 18 timers inkubering blev pladerne vurderet for toksicitet og derpå smittet med 0,1 ml per fordybning af vesikular stomatitisvirus (VSV) suspension indeholdende 10-30 g TCID^q (vævskultur inficeret med dosis, som forårsager 50%'s infektion). Pladerne blev inkuberet i yderligere 3-4. dage og derpå vurderet mikroskopisk for cytopatogen effekt (CPE). Celler beskyttet for virusinfektion var frie for CPE. Den minimale beskyttende dosis (MPD) af poly—(I:C) alene blev noteret, og mængden af forbedret eller forøget anti-viral virkning forårsaget ved kombination med den angivne forbindelse opført for hvert fortyndingsniveau og forbindelsen.
