Un anticuerpo de la invención preferentemente comprende una o más regiones determinantes de complementariedad (CDR), preferentemente por lo menos la región variable de CDR3, de la región variable de cadena pesada (VH) y/o de la región variable de cadena ligera (VL) de un anticuerpo monoclonal contra CLD18, preferentemente de un anticuerpo monoclonal contra CLD18 descrito en la presente memoria, y preferentemente comprende una o más de las regiones determinantes de complementariedad (CDR), preferentemente por lo menos la región variable de CDR3, de las regiones variables de cadena pesada (VH) y/o de las regiones variables de cadena ligera (VL) indicadas en la presente memoria. En una forma de realización, dicha región o regiones determinantes de complementariedad (CDR) se seleccionan de entre un conjunto de regiones determinantes de complementariedad, CDR1, CDR2 y CDR3 indicadas en la presente memoria. En una forma de realización particularmente preferente, un anticuerpo de la invención preferentemente comprende las regiones determinantes de complementariedad CDR1, CDR2 y CDR3 de la región variable de cadena pesada (VH) y/o de la región variable de cadena ligera (VL) de un anticuerpo monoclonal contra CLD18, preferentemente de un anticuerpo monoclonal contra CLD18 indicado en la presente memoria, y preferentemente comprende las regiones determinantes de complementariedad CDR1, CDR2 y CDR3 de las regiones variables de cadena pesada (VH) y/o de las regiones variables de cadena ligera (VL) indicadas en la presente memoria.
Un anticuerpo de la invención que comprende una o más CDR, un conjunto de CDR o una combinación de conjuntos de CDR tal como se indica en la presente memoria, comprende dichas CDR conjuntamente con sus regiones de marco intermedias. Preferentemente, la parte también incluye por lo menos aproximadamente 50% de la primera o cuarta región de marco o de ambas, siendo el 50%, el 50% C-terminal de la primera región de marco y el 50% N-terminal de la cuarta región de marco. La construcción de anticuerpos de la presente invención preparados mediante técnicas de ADN recombinante puede resultar en la introducción de residuos en el lado N-terminal o Cterminal de las regiones variables codificadas por conectores introducidos para facilitar la clonación u otras etapas de manipulación, incluyendo la introducción de conectores para unir regiones variables de la invención a secuencias proteicas adicionales, incluyendo cadenas pesadas de inmunoglobulina, otros dominios variables (por ejemplo en la producción de diacuerpos) o marcajes de proteína.
Un anticuerpo de la invención que comprende una o más CDR, un conjunto de CDR o una combinación de conjuntos de CDR tal como se indica en la presente memoria, comprende dichas CDR en una región de marco de anticuerpo humano.
La referencia en la presente memoria a un anticuerpo que comprende con respecto a la cadena pesada del mismo una cadena particular, o una región o secuencia particular, preferentemente se refiere a una situación en la que todas las cadenas pesadas de dicho anticuerpo comprenden dicha cadena, región o secuencia particular. Esto resulta aplicable, análogamente, a la cadena ligera de un anticuerpo.
La presente memoria describe asimismo a ácidos nucleicos que comprenden genes o secuencias de ácidos nucleicos codificantes de anticuerpos o partes de los mismos, por ejemplo una cadena de anticuerpo, tal como se indica en la presente memoria. Los ácidos nucleicos pueden encontrarse comprendidos en un vector, por ejemplo un plásmido, cósmido, virus, bacteriófago u otro vector utilizado, por ejemplo, convencionalmente en ingeniería genética. El vector puede comprender genes adicionales, tales como genes marcadores que permiten la selección del vector en una célula huésped adecuado y bajo condiciones adecuadas. Además, el vector puede comprender elementos de control de la expresión que permiten la expresión correcta de las regiones codificantes en huéspedes adecuados. Dichos elementos de control son conocidos por el experto en la materia y entre ellos pueden incluirse un promotor, un casete de corte y empalme y un codón de inicio de la traducción.
El ácido nucleico puede estar unido funcionalmente a las secuencias de control de la expresión anteriormente indicadas, permitiendo la expresión en células eucarióticas o procarióticas. Los elementos de control que garantizan la expresión en las células eucarióticas o procarióticas son bien conocidos por el experto en la materia.
Los métodos de construcción de las moléculas de ácidos nucleicos, para la construcción de vectores que comprenden las moléculas de ácidos nucleicos anteriormente indicadas, para la introducción de los vectores en células huésped apropiadamente seleccionados, o para provocar o conseguir la expresión, son bien conocidos de la técnica.
Otras características y ventajas de la presente invención resultarán evidentes a partir de la descripción detallada y reivindicaciones siguientes.

La fig. 29 muestra la expresión de CLD18 en estómago normal. El análisis inmunohistoquímico con anticuerpo específico de CLD18 de estómago de ratón revela un patrón de expresión conservado. Aunque los epitelios superficiales y las criptas más profundas expresaban CLD18 en la superficie celular, la región central del cuello era negativa para CLD18.

Descripción detallada de la invención
Los anticuerpos descritos en la presente memoria pueden ser anticuerpos monoclonales aislados que se unen específicamente a un epítopo presente sobre CLD18. Entre los anticuerpos monoclonales aislados comprendidos dentro de la presente invención se incluyen los anticuerpos IgA, IgG1-4, IgE, IgM e IgD. En una forma de realización, el anticuerpo es un anticuerpo IgG1, más particularmente un IgG1, kappa o IgG1, isotipo lambda. En otra forma de realización, el anticuerpo es un anticuerpo IgG3, más particularmente un isotipo kappa de IgG3 ó IgG3, isotipo lambda. En todavía otra forma de realización, el anticuerpo es un anticuerpo IgG4, más particularmente un isotipo kappa de IgG4 ó IgG4, isotipo lambda. En todavía otra forma de realización, el anticuerpo es un anticuerpo IgA1 ó IgA2. En todavía otra forma de realización, el anticuerpo es un anticuerpo IgM.
En una forma de realización, la invención se refiere a anticuerpos que se unen específicamente a células que expresan CLD18, y que preferentemente: (i) se unen a células que expresan CLD18A2, e (ii) no se unen a células que no expresan CLD18A2 pero que expresan CLD18A1. Los anticuerpos de la invención preferentemente: (i) median en la eliminación de las células que expresan CLD18A2, e (ii) no median en la eliminación de las células que no expresan CLD18A2 pero que expresan CLD18A1.
En otra forma de realización, la invención se refiere a anticuerpos que: (i) se unen a células tumorales que expresan CLD18, (ii) no se unen a células que expresan CLD18 de mucosa de estómago normal, y/o (iii) no se unen a células que expresan CLD18 de tejido pulmonar no canceroso.
La invención incluye además anticuerpos que: (i) median en la eliminación de células tumorales que expresan CLD18, (ii) no median en la eliminación de células que expresan CLD18 de mucosa de estómago normal, y/o (iii) no median en la eliminación de células que expresan CLD18 de tejido pulmonar no canceroso.
En formas de realización particulares, los anticuerpos de la invención (i) se unen a un epítopo sobre CLD18A2 que no se encuentra presente sobre CLD18A1, preferentemente SEC ID nº 21, nº 22 y nº 23, (ii) se unen a un epítopo localizado sobre CLD18A2-bucle1, preferentemente SEC ID nº 28, (iii) se unen a un epítopo localizado sobre CLD18A2-bucle2, preferentemente SEC ID nº: 31, (iv) se unen a un epítopo que comprende CLD18A2-bucle1 y CLD18A2-bucleD3, (v) se unen a un epítopo no glucosilado localizado sobre CLD18A2-bucleD3, preferentemente SEC ID nº: 29, o (vi) se unen a un epítopo presente en CLD18 humano y de ratón (SEC ID nº 2, SEC ID nº 8 y SEC ID n 35, SEC ID nº 37, respectivamente).
En formas de realización particularmente preferentes, los anticuerpos de la invención se unen a un epítopo de CLD18A2 que no se encuentra presente sobre CLD18A1.
Entre los anticuerpos de la invención se incluyen algunos anticuerpos totalmente humanos. Dichos anticuerpos pueden ser producidos en un animal transgénico no humano, por ejemplo en un ratón transgénico, capaz e producir múltiples isotipos de anticuerpos monoclonales humanos de CLD18 mediante recombinación V-D-J e intercambio de isotipos. Dicho animal transgénico también puede ser un conejo transgénico para la producción de anticuerpos policlonales, tal como se da a conocer en la patente US nº 2003/0017534.
La unión de un anticuerpo de la invención al antígeno CLD18 puede mediar en la destrucción de las células que expresan CLD18 (por ejemplo, una célula tumoral), por ejemplo mediante la activación del sistema de complemento. La destrucción de las células que expresan CLD18 puede producirse mediante uno o más de los mecanismos siguientes: citotoxicidad dependiente de complemento (CDC) de las células que expresan CLD18; apoptosis de las células que expresan CLD18; fagocitosis de célula efectora de las células que expresan CLD18; o citotoxicidad celular dependiente de anticuerpo de célula efectora (ADCC) de las células que expresan CLD18.
Con el fin de que la presente invención se entiende con mayor facilidad, en primer lugar se definen determinados términos. Se proporcionan definiciones adicionales durante toda la descripción detallada.
Definición de términos
El término "CLD18" se refiere a la claudina-18 e incluye cualesquiera variantes, incluyendo CLD18A1 y CLD18A2, conformaciones, isoformas y homólogos específicos de CLD18 que se expresan naturalmente en las células o que se expresan en células transfectadas con el gen CLD18. Preferentemente, "CLD18" se refiere a CLD18 humano, en particular CLD18A2 (SEC ID nº 1, nº 2) y/o CLD18A1 (SEC ID nº 7, nº 8), más preferentemente CLD18A2.
El término "CLD18A1" incluye las variantes modificadas post-traduccionalmente, isoformas y homólogos específicos de CLD18A1 humano naturalmente expresados por las células o que se expresan sobre células transfectadas con el gen CLD18A1.

regiones variables de las cadenas pesadas y ligeras contienen un dominio de unión que interactúa con un antígeno. Las regiones constantes de los anticuerpos pueden mediar en la unión de la inmunoglobulina a tejidos o factores del huésped, incluyendo diversas células del sistema inmunológico (por ejemplo células efectoras) y el primer componente (C1q) del sistema clásico del complemento.
La expresión "anticuerpo humanizado" se refiere a una molécula que presenta un sitio de unión a antígeno que se deriva sustancialmente de una inmunoglobulina procedente de una especie no humana, en la que la estructura restante de inmunoglobulina de la molécula se basa en la estructura y/o en la secuencia de una inmunoglobulina humana. El sitio de unión a antígeno puede comprender dominios variables completos fusionados con dominios constantes o únicamente las regiones determinantes de complementariedad (CDR) injertadas en las regiones de marco apropiadas en los dominios variables. Los sitios de unión a antígeno puede ser de tipo salvaje o modificados con una o más sustituciones de aminoácido, por ejemplo modificados para ser más similares a inmunoglobulinas humanas. Algunas formas de anticuerpos humanizados conservan la totalidad de las secuencias de CDR (por ejemplo un anticuerpo de ratón humanizado que contiene la totalidad de las seis CDR del anticuerpo de ratón). Otras formas presentan una o más CDR que se encuentran alteradas respecto al anticuerpo original.
La expresión "anticuerpo quimérico" se refiere a aquellos anticuerpos en los que una parte de cada una de las secuencias de aminoácidos de las cadenas pesadas y ligeras es homóloga respecto a las secuencias correspondientes en anticuerpos derivados de una especie particular o pertenecientes a una clase particular, mientras que el segmento restante de las cadenas es homólogo respecto a las secuencias correspondientes de otra especie. Típicamente, la región variable de tanto las cadenas ligeras como las pesadas imita las regiones variables de anticuerpos derivados de una especie de mamífero, mientras que las partes constantes son homólogas respecto a las secuencias de anticuerpos derivadas de otra especie. Una clara ventaja de dichas formas quiméricas es que la región variable puede derivarse convenientemente de fuentes actualmente conocidas utilizando células B o hibridomas fácilmente disponibles de organismos huésped no humanos en combinación con regiones constantes derivadas de, por ejemplo, preparaciones de células humanas. Aunque la región variable presenta la ventaja de su fácil preparación y de que la especificidad no resulta afectada por el origen, siendo humana la región constante, es menos probable que induzca una respuesta inmunológica de un sujeto humano al inyectar los anticuerpos que con la región constante de origen no humano. Sin embargo, la definición no se encuentra limitada al presente ejemplo particular.
La expresión "parte de unión a antígeno" de un anticuerpo (o simplemente "parte de unión"), tal como se utiliza en la presente memoria, se refiere a uno o más fragmentos de un anticuerpo que conservan la capacidad de unirse específicamente a un antígeno. Se ha demostrado que la función de unión a antígeno de un anticuerpo puede llevarse a cabo con fragmentos de un anticuerpo de longitud completa. Entre los ejemplos de fragmentos de unión comprendidos dentro de la expresión "parte de unión a antígeno" de un anticuerpo se incluyen: (i) fragmentos Fab, fragmentos monovalentes que consisten de los dominios VL, VH, CL y CH, (ii) fragmentos F(ab')2, fragmentos bivalentes que comprenden dos fragmentos Fab unidos mediante un puente disulfuro en la región bisagra, (iii) fragmentos Fd que consisten de los dominios VH y CH, (iv) fragmentos Fv que consiste de los dominios VL y VH de un único brazo de un anticuerpo, (v) fragmentos dAb (Ward et al., Nature 341:544-546, 1989), que consisten de un dominio VH, (vi) regiones determinantes de complementariedad (CDR) aisladas, y (vii) combinaciones de dos o más CDR aisladas que opcionalmente pueden unirse mediante un conector sintético. Además, aunque los dos dominios del fragmento Fv, VL y VH, se encuentran codificados por genes separados, pueden unirse, utilizando métodos recombinantes, mediante un conector sintético que les permite prepararlos en forma de una sola cadena proteica en la que las regiones VL y VH se emparejan formando moléculas monovalentes (conocidas como Fv de cadena sencilla (scFv), ver, por ejemplo, Bird et al., Science 242:423-426, 1988, y Huston et al., Proc. Natl. Acad. Sci. USA 85:5879-5883, 1988). Dichos anticuerpos de cadena sencilla también se pretende que se encuentren comprendidos dentro de la expresión "parte de unión a antígeno" de un anticuerpo. Un ejemplo adicional son las proteínas de fusión de dominio de unión-inmunoglobulina, las cuales comprenden: (i) un polipéptido dominio de unión que se encuentra fusionado con un polipéptido región bisagra de inmunoglobulina, (ii) una región constante CH2 de cadena pesada de inmunoglobulina fusionada con la región bisagra, e (iii) una región constante CH3 de cadena pesada de inmunoglobulina fusionada con la región constante CH2. El polipéptido dominio de unión puede ser una región variable de cadena pesada o una región variable de cadena ligera. Las proteínas de fusión de dominio de unióninmunoglobulina se dan a conocer en mayor detalle en las patentes US nº 2003/0118592 y nº 2003/0133939. Estos fragmentos de anticuerpo se obtienen utilizando técnicas convencionales conocidas por el experto en la materia, y los fragmentos se criban para su utilidad de la misma manera que los anticuerpos intactos.
El término "epítopo" se refiere a un determinante proteico capaz de unirse a un anticuerpo, en el que la expresión "de unión" en la presente memoria preferentemente se refiere a una unión específica. Los epítopos habitualmente consisten de agrupamientos superficiales de moléculas químicamente activos, tales como aminoácidos o cadenas laterales sacáridas y habitualmente presentan características específicas de estructura tridimensional, así como características de carga específicas. Los epítopos conformacionales y no conformacionales se distinguen en que la unión a los primeros, pero no a los segundos, se pierde en presencia de solventes desnaturalizantes.

Tal como se utiliza en la presente memoria, un "anticuerpo heterólogo" se define en relación a un organismo transgénico que produce este tipo de anticuerpo. Esta expresión se refiere a un anticuerpo que presenta una secuencia de aminoácidos o una secuencia de ácidos nucleicos codificante que corresponde a la presente en un organismo que no consiste del organismo transgénico, y que se deriva generalmente de una especie que no es la del organismo transgénico.
Tal como se utiliza en la presente memoria, un "anticuerpo heterohíbrido" se refiere a un anticuerpo que presenta cadenas ligeras y pesadas de diferentes orígenes de organismo. Por ejemplo, un anticuerpo que presenta una cadena pesada humana asociada a una cadena ligera murina es un anticuerpo heterohíbrido.
Los anticuerpos indicados en la presente memoria preferentemente han sido aislados. Un "anticuerpo aislado" tal como se utiliza en la presente memoria pretende referirse a un anticuerpo que se encuentra sustancialmente libre de otros anticuerpos que presentan diferentes especificidades antigénicas (por ejemplo un anticuerpo aislado que se une específicamente a CLD18 se encuentra sustancialmente libre de anticuerpos que se unen específicamente a antígenos diferentes de CLD18). Un anticuerpo aislado que se une específicamente a un epítopo, isoforma o variante de CLD18 humano puede, sin embargo, presentar reactividad cruzada con otros antígenos relacionados, por ejemplo de otra especie (por ejemplo homólogos específicos de CLD18). Además, un anticuerpo aislado puede encontrarse sustancialmente libre de otros materiales celulares y/o compuestos químicos. En una forma de realización de la invención, una combinación de anticuerpos monoclonales "aislados" se refiere a anticuerpos que presentan diferentes especificidades y que se combinan en una composición bien definida.
Según la invención, la expresión "de unión" preferentemente se refiere a "de unión específica". Tal como se utiliza en la presente memoria, "de unión específica" se refiere a la unión de un anticuerpo a un antígeno predeterminado. Típicamente, el anticuerpo se une con una afinidad correspondiente a una KD de aproximadamente 1x10-7 M o inferior, y se une al antígeno predeterminado con una afinidad correspondiente a una KD que es por lo menos dos órdenes de magnitud inferior a su afinidad para la unión a un antígeno no específico (por ejemplo BSA, caseína) diferente del antígeno predeterminado o de un antígeno estrechamente relacionado.
El término "KD" (M), tal como se utiliza en la presente memoria, pretende referirse a la constante de equilibrio de disociación de una interacción particular de anticuerpo-antígeno.
Tal como se utiliza en la presente memoria, "isotipo" se refiere a la clase de anticuerpo (por ejemplo IgM o IgG1) que se encuentra codificada por genes de región constante de cadena pesada.
Tal como se utiliza en la presente memoria, "intercambio de isotipo" se refiere al fenómeno por el que la clase, o isotipo, de un anticuerpo cambia de una clase de Ig a una de las otras clases de Ig.
La expresión "de origen natural" tal como se utiliza en la presente memoria aplicada a un objeto se refiere al hecho de que un objeto puede encontrarse en la naturaleza. Por ejemplo, un polipéptido o secuencia polinucleótido que se encuentra presente en un organismo (incluyendo virus), que puede aislarse a partir de una fuente en la naturaleza y que no ha sido modificada deliberadamente por el hombre en el laboratorio es de origen natural.
El término "reorganizado" tal como se utiliza en la presente memoria se refiere a una configuración de un locus de cadena pesada o ligera de inmunoglobulina en el que un segmento V se sitúa inmediatamente contiguo a un segmento D-J ó J en una conformación codificante esencialmente de un dominio VH o VL completo, respectivamente. Puede identificarse un locus génico de inmunoglobulina (anticuerpo) reorganizado mediante comparación con el ADN de línea germinal; un locus reorganizado presentará por lo menos un elemento de homología heptámero/nonámero recombinado.
La expresión "no reorganizado" o "configuración de línea germinal" tal como se utiliza en la presente memoria en referencia un segmento V se refiere a la configuración en la que el segmento V no se ha recombinado de manera que sea inmediatamente contiguo a un segmento D o J.
La expresión "molécula de ácidos nucleicos", tal como se utiliza en la presente memoria, pretende incluir moléculas de ADN y moléculas de ARN. Una molécula de ácidos nucleicos puede ser de cadena sencilla o de doble cadena, aunque preferentemente es ADN de doble cadena.
Los ácidos nucleicos indicados según la invención preferentemente han sido aislados. La expresión "ácido nucleico aislado" se refiere según la invención a que el ácido nucleico: (i) ha sido amplificado in vitro, por ejemplo mediante reacción en cadena de la polimerasa (PCR), (ii) es producido recombinantemente mediante clonación, (iii) ha sido purificado, por ejemplo mediante corte y fraccionamiento mediante electroforesis en gel, o (iv) ha sido sintetizado, por ejemplo mediante síntesis química. Un ácido nucleico aislado es un ácido nucleico que se encuentra disponible para la manipulación mediante técnicas de ADN recombinante.
Los ácidos nucleicos pueden, según la invención, encontrarse presentes solos o en combinación con otros ácidos nucleicos, que pueden ser homólogos o heterólogos. En formas de realización preferidas, un ácido nucleico se

Además, puede desearse según la presente invención modificar las secuencias de aminoácidos indicadas en la presente memoria, en particular las de regiones constantes de cadena pesada humana para adaptar la secuencia a un alotipo deseado, por ejemplo un alotipo observado en la población caucásica. Dichas modificaciones se seleccionan preferentemente de entre el grupo que consiste de las sustituciones de aminoácidos siguientes en SEC ID nº 46 o en posiciones correspondientes dentro de otras regiones constantes de cadena pesada humana: K93R, D235E y L237M. Preferentemente, la totalidad de dichas modificaciones se encuentra incluida en las secuencias de aminoácidos de las regiones constantes de cadena pesada humana.
Según la invención, la expresión "posiciones correspondientes" se refiere a nucleótidos o residuos aminoácidos que, en una alineación de secuencias de dos ácidos nucleicos o secuencias de proteína, se alinean uno con otro.
Preferentemente, el grado de identidad entre una secuencia específica de ácidos nucleicos indicada en la presente memoria y una secuencia de ácidos nucleicos modificada con respecto a dicha secuencia específica de ácidos nucleicos es de por lo menos 70%, preferentemente de por lo menos 75%, más preferentemente de por lo menos 80%, todavía más preferentemente de por lo menos 90% o todavía más preferentemente de por lo menos 95%, 96%, 97%, 98% ó 99%. Preferentemente, las dos secuencias son capaces de hibridarse y formar un dúplex estable una con la otra, llevando a cabo la hibridación preferentemente bajo condiciones que permiten la hibridación específica entre polinucleótidos (condiciones restrictivas). Las condiciones restrictivas se describen en, por ejemplo, Molecular Cloning: A Laboratory Manual, J. Sambrook et al., editores, 2a edición, Cold Spring Harbor Laboratory Press, Cold Spring Harbor, New York, 1989, o en Current Protocols in Molecular Biology, F.M. Ausubel et al., editores, John Wiley & Sons, Inc., New York, y se refieren, por ejemplo, a la hibridación a 65ºC en tampón de hibridación (3,5xSSC, Ficoll al 0,02%, polivinilpirrolidona al 0,02%, albúmina de suero bovino al 0,02%, NaH2PO4 2,5 mM (pH 7), SDS al 0,5%, EDTA 2 mM). SSC es cloruro sódico 0,15 M/citrato sódico 0,15 M, pH 7. Tras la hibridación, la membrana a la que se ha transferido el ADN se lava en, por ejemplo, 2xSSC a temperatura ambiente y después en 0,1-0,5xSSC/0,1xSDS a temperaturas de hasta 68ºC.
Preferentemente, el grado de similitud, preferentemente de identidad entre una secuencia específica de aminoácidos indicada en la presente memoria y una secuencia de aminoácidos modificada con respecto a dicha secuencia específica de aminoácidos, tal como entre secuencias de aminoácidos que muestran homología sustancial, es de por lo menos 70%, preferentemente de por lo menos 80%, más preferentemente de por lo menos 90% o todavía más preferentemente de por lo menos 95%, 96%, 97%, 98% ó 99%.
La totalidad de las secuencias modificadas anteriormente indicadas se encuentran comprendidas dentro del alcance de la presente invención.

b. Generación de hibridomas productores de anticuerpos monoclonales humanos de CLD18:
Se aislaron esplenocitos de ratón y se fusionaron con PEG a una línea celular de mieloma de ratón utilizando protocolos estándares. A continuación, los hibridomas resultantes se cribaron para la producción de 10 inmunoglobulinas con especificidad para CLD18 utilizando células HEK293 transfectadas con un ácido nucleico codificante de CLD18 humano mediante análisis FACS. Se fusionaron suspensiones de células individuales de linfocitos esplénicas procedentes de ratones inmunizados con células de mieloma de ratón no secretoras P3X63Ag8U.1 (ATCC nº CRL 1597) en una proporción 2:1 utilizando PEG al 50% (Roche Diagnostics, nº CRL 738641). Se sembraron las células en placas a razón de aproximadamente 3x104/pocillo en placas de microtitulación 15 de fondo plano, seguido de la incubación durante aproximadamente dos semanas en medio selectivo que contenía suero de feto bovino al 10%, fusión de hibridoma al 2% y suplemento de clonación (HFCS, Roche Diagnostics, nº CRL 1 363 735) más HEPES 10 mM, 2-mercaptoetanol 0,055 mM, 50 µg/ml de gentamicina y 1xHAT (Sigma, nº CRL H0262). Tras 10 a 14 días, los pocillos individuales se cribaron mediante citometría de flujo para anticuerpos monoclonales anti-CLD18. Los hibridomas secretores de anticuerpos se sembraron en placa nuevamente, se
20 cribaron nuevamente y, en el caso de que todavía fuesen positivos para anticuerpos monoclonales anti-CLD18, se subclonaron mediante dilución limitante. A continuación, los subclones se cultivaron in vitro para generar cantidades reducidas de anticuerpo en medio de cultivo de tejidos para la caracterización. Se seleccionó por lo menos un clon de cada hibridoma, que conservó la reactividad de las células parentales (según FACS). Se generaron 9 bancos de células viables para cada clon y se almacenaron en nitrógeno líquido.

c. Selección de anticuerpos monoclonales de unión a CLD18:
para determinar el isotipo de los anticuerpos, se llevó a cabo un ELISA de los isotipos. Se utilizó el kit monoAB ID de ratón (Zymed, nº CRL 90-6550) o alternativamente el kit de isotipado de anticuerpos monoclonales de ratón IsoStrip 30 (Roche, nº de cat. 1493027) para determinar las subclases de Ig de los anticuerpos monoclonales identificados como reactivos con CLD18.
