Description

Przedmiotem wynalazku jest sposób wytwarzania optycznie czynnej pochodnej kwasu (2R) lub (2S) 3-(3-{egzobicyklo[ 2.2.1 ]hept-2- yloksy} -4-metoksyfenylo)-glutarowego mającej zastosowanie jako związek pośredni do wytwarzania czynnego farmaceutycznie (2R) i (2S) 5-(3-{egzo-bicyklo [2.2.1 ]hept-2-yloksy} -4-metoksyfenylo) -3,4,5,6-tetrahydropirymidyn-2(1H)-onu.
Związki wytwarzane sposobem według wynalazku określane są wzorami 1 i 2. Wzór 1 odpowiada enancjomerowi (2S) a wzór 2 enancjomerowi (2R). Wytwarzane z nich związki o wzorach 3 i 4 są szczególnie cennymi przedstawicielami związków, ujawnionych szeroko przez Saccomano i wsp. w opublikowanym, międzynarodowym zgłoszeniu patentowym WO 87/06576, mających zastosowanie jako antydepresanty. Chociaż w odnośniku tym ujawniono szczegółowo racemiczny 5-(3-{(2S)-egzo-bicyklo[2.2.1] hept-2-yloksy}-4-metoksyfenylo)3,4,5,6-tetrahydropiiymidyn-2( 1H)-on, to nie opisano w nim szczegółowo optycznie czynnych odmian tego związku ani konkretnych sposobów ich otrzymywania. Związki o wzorach 3 i 4 są także szczególnie cenne w leczeniu astmy i niektórych chorób skórnych.
Sposób według wynalazku wytwarzania optycznie czynnej pochodnej kwasu (2R) lub (2S)
3- (3-{egzo-bicyklo[ 2.2.1 ] hept-2-yloksy} - 4-metoksyfenylo)glutarowego o określonech wyżej wzorach 1 lub 2 polega na reakcji optycznie czynnego 3-(egzo-bicyklo[ 2.2.1 ]- hept-2-yloksy)4- metoksebenzaldehydu o wzorze 5 enancjomer 2S, lub o wzorze 6 enancjomer 2R, z co najmniej dwoma równoważnikami molowymi kwasu 2-cyjanooctowego w pirydynie, w obecności katalitycznych ilości piperydyny, w temperaturze w zakresie około 25 - 100°C, z wytworzeniem optycznie czynnego 3-(3-{egzo-bicyklo [2.2.1 ] hept-2-yloksy}-4-metoksyfenylo)pentanodinitrylu, przedstawionego wzorem 1 lub wzorem 2.
Otrzymaną sposobem według wynalazku pochodną kwasu glutarowego przekształca się w glutaramid, który z kolei cyklizuje się działaniem molowego nadmiaru tetraoctanu ołowiu w pirydynie z utworzeniem optycznie czynnego 5-{3-{egzo-bicyklo[ 2.2.1]hept-2-yloksy }-4-metoksyfenelo]-3,4,5,6-tetrahydropirymidyn-2-(1H)-onu o wzorze 3 lub o wzorze 4.
Wyjściowym związkiem stosowanym w sposobie według wynalazku jest optycznie czynny 3-(egzo-bicyklo[ 2.2.1 ]hept-2-yloksy)-4- metoksybenzaldehyd o wzorze 5 lub o wzorze 6, który otrzymuje się z kolei z optycznie czynnych endo-bicyklo[ 2.2.1 ]heptan-2-oli (endo-norborneoli) otrzymanych z rozdzielonego enzymatycznie endo-bicyklo [2.2.1 ]heptan-2-olu przez częściową transtryfikację endo-bicyklo[2.2.1 ]heptan-2-olu maślanem 2,2,2-trichloroetylu prowadzoną w prawie bezwodnym, obojętnym dla reakcji rozpuszczalniku organicznym, w obecności lipazy trzustkowej ssaka i oddzielenie z otrzymanej mieszaniny nieprzereagowanego (-)- (2S)-endo-bicyklo [2.2.1 ]heptan-2-olu [ (2S)-endo-norborneolu] o wzorze 7 i maślanu (2R)165 132
- endo-bicyklo [2.2.1 ]heptan-2-ylu, który hydrolizuje się do enancjomerycznego (+)-(2R)-endo-bicyklo [2.2.1 ]heptan-2-olu [ (2R)-endo-norborneolu ] o wzorze 8. Otrzymane optycznie czynne (2R)- i (2S) -endo- bicyklo[2.2.1]heptan-2-ole poddaje się następnie reakcji z 3- hydroksy-4-metoksybenzaldehydem w obecności trifenylofosfiny i azadikarboksylanu dietylu z utworzeniem, odpowiednio, optycznie czynnych aldehydów 3- {(2S)-egzo-bicyklo [2.2.1] hept-2-yloksy}-4-metoksybenzaldehydu o wzorze 5, i 3-{(2R)-egzo-bicyklo-[2.2.1]hept-2-yloksy }-4-metoksybenzaldehydu o wzorze 6.
Przekształcenie optycznie czynnych (2R)- i (2S) -endo-norborneoli w odpowiednie aldehydy o wzorach 5 i 6 przeprowadza się sposobem ujawnionym w zgłoszeniu nr WO 87/06576.
Zgodnie z wynalazkiem aldehydy o powyższych wzorach 5 i 6 kondensuje się z co najmniej dwoma równoważnikami molowymi kwasu cyjanooctowego, otrzymując dinitryl przedstawionym wzorem 1 lub wzorem 2. Kondensację dogodnie przeprowadza się w pirydynie, która ponadto służy jako katalizator zasadowy, w obecności aminy drugorzędowej, korzystnie niezatłoczonej sterycznie, jak piperydyna lub pirolina, w ilości ogólnie zapewniającej nadmiar molowy. Chociaż początkowe etapy reakcji mogą być przeprowadzone w temperaturach pokojowych, reakcję (w tym dekarboksylację) najlepiej kończy się w temperaturze leżącej w zakresie około 80 - 110°C.
Następujący przykład ilustruje sposób według wynalazku, przy czym wstępne etapy dotyczą wytwarzania związków wyjściowych.
Przykład. (+)-(2R)- i (-)-(2S)-endo-Norborneol [ (2R)- i (2S) - endo-Bicyklo[2.2.1]heptan-2-ol d,1-Endonorborneol (5,0g, 44,6 mmoli) i maślan trichloroetylu (5,1g 23,2 mmoli) rozpuszczono w 40 ml eteru dietylowego. Dodano sita molekularne 4.10'1 m (4g) i mieszaninę mieszano w temperaturze pokojowej. Świńską lipazę trzustkową (Sigma, typ II, surowa) dodano porcjami w ilościach 0,5g, 1,0g, 1,0g, 1,0g i 0,5g po odpowiednio 0, 20,43, 50 i 67 godzinach. Reakcję monitorowano za pomocą *H-NMR i przy stopniu przereagowania w przybliżeniu 50% (92h) mieszaninę przesączono przez ziemię okrzemkową i odparowano pod próżnią bez ogrzewania. (Alkohol łatwo sublimuje). Surową pozostałość chromatografowano na żelu krzemionkowym stosując gradientowy system eluenta 2 - 25% eter/heksan, otrzymując 2,9g (15,9 mmola) maślanu (2R)- endonorbomylu w postaci przezroczystego oleju i 1,8g (16,0 mmola) (2S)-endonorborneolu w postaci białego osadu; [alfa]o = - 2,03°; e.e. 87,2% [za pomocą Ή-NMR pochodnej, estru kwasu (S)-alfa-metoksy-alfa-(triffuorometylo)fenylooctowego (MTPA)]. Ponieważ specyficzna skręcalność jest tak niska, wartości e.e. oznaczone za pomocą nMr są bardziej wiarygodną miarą czystości optycznej.
Odzyskany maślan endo-norbornylu (2,3g, 12,6 mmola), K2CO3 (2,5g . 18,0 mmola) i metanol (65 ml) mieszano przez 64 godziny w temperaturze pokojowej, po czym dokonano podziału pomiędzy eter i wodę. Warstwę organiczną przemyto wodą i solanką, wysuszono Na2 SO4, przesączono i zatężono w próżni, otrzymując 1,8g (11,6 mmola, wydajność 91,9%) (2R)-endonorborneolu; [ alfa ] d = +2,7; e.e. 87,6% (na podstawie 1 H-NMR estru z MPTA).
Te etapy procesu powtórzono, prowadząc wymianę estrową tylko do 44% przemiany, otrzymując (2S)-endonorborneol o niższej czystości optycznej z wydajnością wyższą niż 90%; [ alfa ]d = -0,88; e.e. 71,4 (na podstawie ‘H-NMR, jak powyżej; oraz (2R)-endonorborneol o wyższej czystości optycznej z wydajnością 56,4%; e.e. większe niż 95% (napodstawie JH-NMR, jak powyżej).
3-{ (2S)-egzo-Bicyklo [2.2.1]hept-2-yloksy }-4-metoksybenzaldehyd
Azodikarboksylan dietylu (28,5g 27,7 ml, 0,141 mmola) i trifenylofosfinę (36,9g, 0,141 mola) rozpuszczono w 200 ml tetrahydrofuranu. Do tego roztworu dodano (+)-(2R)-endonorborneol (7,9g, 0,705 mola) w 100 ml tetrahydrofuranu, a następnie 3-hydroksy-4-metoksybenzaldehyd (izowanilina; 24,4g, 0,141 mola) w 100 ml tetrahydrofuranu. Stosując tę samą metodę, (- )-(2S)-endo-norborneol przekształcono w 3- {(2R)-egzo-bicyklo[2.2.1]-hept-2-yloksy}-4-metoksybenzaldehyd, mający z wyjątkiem kierunku skręcalności, te same właściwości fizyczne.
3-(3- {(2S)-egzo-Bicyklo[2.2.1 ]heptt2-y loksy} -metoksyfenylo) pentadinitryl
Otrzymany produkt (8,5g, 0,0346 mola) rozpuszczono w 250 ml pirydyny. Dodano kwas cyjanooctowy (l4,6g, 0,171 mola) i piperydynę (5 ml) i mieszaninę mieszano w temperaturze pokojowej przez 4 godziny, następnie w 60°C przez 2 godziny i na końcu w 100°C przez 24 godziny. Rozpuszczalnik usunięto przez odpędzenie w próżni a pozostałość rozpuszczono w 250 ml octanu etylu, przemyto nasyconym roztworem NaHCO3 i wodą, ponownie odpędzono rozpuszczalnik i krystalizowano z mieszaniny alkohol izopropylowy/eter izopropylowy, otrzymując 5,84g (54%) tytułowego związku; t.t. (temperatura topnienia) 121 - 123°C; [alfa]D =+17,8 (deuterochloroform).
Stosując tę samą metodę otrzymany poprzednio enancjometryczny produkt przekształcono w 3-(3-{(2R)-egzo-bicyklo[2.2.1]hept-2-ytokyy}-4-metokyyfnnyto)-pnntadinitryl, mający z wyjątkiem kierunku skręcalności, te same właściwości fizyczne, jak izomer (2S).

  1. Zastrzeżenie patentowe
    Sposób wytwarzania optycznie czynnej pochodnej kwasu (2R) lub (2S) 3-(3-{egzo-bicyklo [ 2.2.1 ] hept-2-yloksy }-4-metoksyfenylo)glutarowego o wzorze 1 lub o wzorze 2, znamienny tym, że odpowiednio na 3-{(2R)-egzo-bicyklo[ 2.2.1] hept-2- yloksy}-4-metoksybenzaldehyd o wzorze 6 lub na 3-{(2S)-egzo- bicyklo [ 2.2.1] hept-2-yloksy}-4-metoksy benzaldehyd o wzorze 5 działa się co najmniej dwoma równoważnikami molowymi kwasu 2- cyjanooctowego w pirydynie w obecności katalitycznej ilości piperydyny, w temperaturze zawartej w zakresie około 25 - 100°C.
