Description
Die vorliegende Erfindung bezieht sich auf ein Verfahren zum Einkapseln eines Kernmaterials, wie Gewebe oder individuelle Zellen, und zwar so, dass das Gewebe bzw. die Zellen innerhalb einer semipermeablen Membran, welche für Nährstoffe, Ionen, Sauerstoff und andere zum Beständighalten des Gewebes und zur Unterstützung seiner normalen metabolischen Funktionen erforderlichen Materialien durchlässig, jedoch für Bakterien, Lymphocyten und grosse Proteine solcher Beschaffenheit, wie sie für immunochemische Ab-stossreaktionen verantwortlich sind, undurchlässig ist, lebensfähig und in geschützter Form gehalten werden. Das vorliegende Verfahren erlaubt die Produktion beispielsweise eines Insulin erzeugenden Systems oder eines anderen Hormon produzierenden Systems, weil es das Einkapseln von Pankreas-B-Zellen, -A-Zellen, intakten Langerhans-Inseln aus der Bauchspeicheldrüse von Säugetieren und Menschen und anderen Geweben oder Gewebefraktionen, welche Hormone absondern, erlaubt. Die Kapseln lassen sich in einem Kulturmedium suspendieren und sondern im Verlaufe einer bestimmten Zeit Hormon ab. Die Kapseln können auch als künstliches Pankreas, welches beispielsweise durch Injektion in ein an Diabetes leidendes Säugetier oder einen an Diabetes leidenden Menschen implantiert werden kann, verwendet werden und funktionieren dann in vivo, wobei sie je nach der vorhandenen Zuckerkonzentration Insulin und andere Hormone absondern.
Es darf angenommen werden, dass Verfahren zum Einkapseln von lebendem Gewebe unter Aufrechterhaltung der Lebensfähigkeit desselben bisher nicht bekannt sind. Versuche zur Erreichung dieses Zieles waren bisher vergeblich,
weil die für die Kapselmembranbildung erforderlichen Bedingungen sich lebenden Systemen feindlich gegenüberstanden. Die amerikanische Patentschrift Nr. (Serial Nr. 606 166), von welcher in der vorliegenden Beschreibung ebenfalls die Rede sein wird, offenbart eine Technik zum Einkapseln von labilen biologischen Materialien in einer semipermeablen Membran. Diese Technik ermöglicht beispielsweise das Einkapseln von Enzymen in einer Membran, aus welcher das Enzym nicht entweichen kann, während andererseits dem Enzymsubstrat freier Durchlass gewährt wird. Wenngleich diese Patentschrift Reaktionsbedingungen offenbart, welche die Behandlung von biologischen Materialien ermöglichen, ist doch daraufhinzuweisen, dass jene Patentschrift nirgends das Einkapseln von lebendem Gewebe anregt.
Eingekapselte lebende Zellen, Organellen oder Gewebe besitzen manche interessante Verwendungszwecke. So kann beispielsweise das in einer semipermeablen Membran eingekapselte lebende Material in einer permanenten sterilen Umgebung gehalten und gegen eine direkte Kontaktnahme mit grossen, potentiell zerstörenden Molekeltypen abgeschirmt werden, während andererseits niedrigmolekularen Gewebenährstoffen und metabolischen Produkten freier Durchlass gewährt wird. Das Entwickeln einer solchen Einkapselungs-methode dürfte somit zu Systemen für das Freisetzen wertvoller Hormone, wie z.B. Insulin, führen. In solchen Systemen würde das für die Erzeugung des Materials verantwortliche Gewebe von Säugetieren in solcher Weise eingekapselt, dass ein freier Durchlass der Nährstoffe und der metabolischen Produkte durch die Membran gewährleistet, andererseits aber der Durchlass von Bakterien verhindert würde.

Könnte man die Membrandurchlässigkeit steuern, so würde es möglich, einerseits auch künstliche Organe einzubeziehen, welche in Säugetieren oder Menschen, z.B. Diabetikern, implantiert werden könnten, ohne dass ein Abstossen erfolgen würde, und andererseits einen gesteuerten Hormonaus-stoss, z.B. das Infreiheitsetzen von Insulin, das durch Gluco-sekonzentration eingeleitet wird, zu erzielen.
Es wurden verschiedene Versuche unternommen, um für die Implantation in Säugetieren oder Menschen geeignete künstliche Organe zu erzeugen, indem man eine mechanische semipermeable Trennwand, wie z.B. eine Millipore-Diffu-sionskammer oder eine Kapillarrohrkammer, um das einem Donator entnommene Gewebe vorsah. Solche künstliche Organe erfordern normalerweise eine chirurgische Implantation. Hinzu kommt, dass die Schutzmechanismen der menschlichen oder tierischen Körper das Implantat isolieren, indem durch fibroblastisches Überwachstum Poren verstopft werden.
Die vorliegende Erfindung bezieht sich daher in erster Linie auf ein Verfahren zum Einkapseln von Kernmaterialien, wie z.B. lebendem Gewebe, individuellen Zellen oder biologisch aktiven Materialien, in einer semipermeablen Membran; das Verfahren ist im Anspruch 1 definiert.
Die temporären Kapseln könne aus einem freie saure Gruppen enthaltenden, wasserlöslichen Polysaccharid gebildet werden, welches sich bei Änderung der Bedingungen im Medium durch Zufuhr mehrwertiger Kationen unter Bildung einer die Form nicht verändernden Masse gelieren lässt. Es enthält also eine Vielzahl von Gruppen, welche sich leicht unter Bildung von anionischen Gruppen ionisieren lassen. Die Anwesenheit solcher Gruppen im Polymer erlaubt eine Quervernetzung der Oberflächenschichten der Kapsel unter Bildung einer «permanenten» Membran, sofern eine Behandlung mit Polymeren, die mehrere reaktionsfähige Gruppen mit entgegengesetzter Ladung enthalten, erfolgt.
Das für die Bildung von temporären Kapseln zur Zeit bevorzugte Material ist entweder ein natürliches oder ein synthetisches Polysaccharid solcher Art, welches sich durch Behandlung mit mehrwertigen Kationen, wie z. B. Ca++, unter Bildung einer die Form beibehaltenden Masse gelieren lässt.
Bevorzugtes Polysaccharid sind Alkalimetallalginate. Andere wasserlösliche, verwendbare gummiartige Materialien sind Guargummi, Gummi-arabikum, Carragen, Pektin, Tragantgummi, Xanthangummi oder saure Fraktionen davon.
Die bevorzugte Methode für die Bildung der Tröpfchen besteht darin, dass man die das gummiartige Material, Nährstoffe und Gewebe enthaltende Suspension durch ein vibrierendes Kapillarrohr einzwängt, welches sich im Zentrum eines durch rasches Rühren einer ein mehrwertiges Katjon enthaltenden Lösung erzeugten Wirbels angeordnet ist. Die aus der Spitze des Kapillarrohrs ausströmenden Tröpfchen kommen unmittelbar mit der Lösung der mehrwertigen Kationen in Berührung und liefern ein Gel in Form von sphäroidisch geformten Körpern.
Zur Bildung einer permanenten semipermeablen Membran rings um die temporären Kapseln bedient man sich der Vernetzung der Oberflächenschichten des gelierten gummiartigen, freie saure Gruppen aufweisenden Polysaccharids mit Polymeren, welche mit sauren Gruppen zu reagieren vermögende, reaktionsfähige Gruppen, wie z.B. Amin- oder Imingruppen, enthalten. Dies erfolgt mit Vorteil in einer verdünnten Lösung des ausgewählten Polymers. Je niedriger das Molekulargewicht des Polymers ist, umso grösser erfolgt im allgemeinen das Eindringen in die Oberfläche der temporären Kapseln. Je grösser das Eindringen ist, umso weniger wird die erzielte Membran durchlässig sein. Permanente Vernetzungen werden zufolge der Salzbildung zwischen den mit

Säuren reaktionsfähigen Gruppen des vernetzenden Polymers und den sauren Gruppen des Polysaccharidmaterials erzielt. Die Semipermeabilität lässt sich in gewissen Grenzen dadurch steuern, dass man das Molekulargewicht des vernetzenden Polymers, dessen Konzentration und die Dauer der Reaktion richtig wählt. Mit Erfolg verwendete, vernetzende Polymere sind Polyäthylenimin und Polylysin. Das Molekulargewicht kann je nach dem Ausmass der erforderlichen Permeabilität zwischen 3000 und 100 000 oder mehr schwanken. Gute Resultate erzielt man unter Verwendung von Polymeren mit einem durchschnittlichen Molekulargewicht im Bereiche von ungefähr 35 000.
Die Kapseln können so gestaltet werden, dass sie dank einer besonderen Auswahl des vernetzenden Polymers eine gute in vivo-Lebensdauer aufweisen. Proteine oder Polypep-tidvernetzer, wie z.B. Polylysin, werden in vivo leicht angegriffen, wodurch die Membran relativ rasch zerstört wird. Durch Säugetiere oder Menschen nicht leicht verwertbare Vernetzungsmittel, wie z.B. Polyäthylenimin, erlauben eine längere Lebensdauer der Membranen. Durch Auswahl des vernetzenden Polymers oder durch gleichzeitige oder nachträgliche Vernetzung mit zwei oder mehreren solcher Mate- -rialien ist es möglich, die Lebensdauer des implantierten Gewebes nach dem gesteckten Ziele zu erreichen.
Es ist ferner auch bei gewissen zur Bildung der temporären Kapseln verwendeten Materialien möglich, den Massentransfer innerhalb der Kapsel nach erfolgter Bildung der permanenten Membran dadurch zu verbessern, dass man die Bedingungen, unter welchen das Material flüssig ist, wiederum herstellt, beispielsweise durch Entfernen des mehrwertigen Kations. Dies kann durch Ionenaustausch, z.B. durch Eintauchen in einen mit Phosphat gepufferten salzhaltigen Puffer oder Citratpuffer geschehen. In gewissen Fällen, z.B. dann, wenn man das eingekapselte Gewebe konservieren will oder wenn die temporär gelierte Kapsel permeabel ist, kann es von Vorteil sein, das eingekapselte gummiartige Material in verntztem, geliertem Zustande zu belassen.
Das oben beschriebene Verfahren wurde zum Einkapseln von lebefähigen Langerhans-Inseln angewandt, welche in einem Medium, welches die für die Lebensfähigkeit und das Aufrechterhalten des in vitro Metabolismus des Gewebes erforderlichen Materialien enthält, in Gegenwart von Glucose Insulin ausstossen. Dermassen eingekapseltes Gewebe liess sich während drei Monaten in lebensfähigem Zustande erhalten. Auch Leberzellen wurden eingekapselt und haben sich in einem physiologisch aktiven Zustande gezeigt.
Die Kapseln gemäss vorliegender Erfindung ermöglichen eine Gewebeimplantationsmethode, welche keinen chirurgischen Eingriff erfordert und keine Probleme der Immunreaktion mit sich bringt. Die Kapseln werden an einer geeigneten Stelle des Körpers injiziert und funktionieren solange normal, bis das Gewebe erschöpft ist oder bis die natürlichen Körpervorgänge die Kapseln isolieren, weil die für die Lebensfähigkeit des Gewebes erforderlichen Substanzen nicht mehr zur Verfügung stehen. In diesem Zeitpunkt kann man, da für das Implantat kein chirurgischer Eingriff erforderlich war, frisches Gewebe durch eine weitere Injektion leicht zuführen. Der menschliche oder tierische Körper kann daher, solange erwünscht, mit der spezifischen Funktion versehen werden.
Gemäss einer bevorzugten Ausführungsform der Erfindung werden tierische oder menschliche Langerhans-Inseln oder -Inselpräparate, welche die gewünschten Mengen an A-, B- und/oder D-Zellen aus den Inseln enthalten, in mit Poly-iysin und/oder Polyäthylenimin vernetzten Alginatmembra-nen eingekapselt. Diese lassen sich periodisch beispielsweise in die Bauchhöhle eines an Diabetes leidenden menschlichen

oder tierischen Körpers injizieren und funktionieren als künstliche Bauchspeicheldrüse.
Ein erstes Ziel der vorliegenden Erfindung ist daher die Schaffung eines Verfahrens zum Einkapseln von lebenden Zellen, Organellen oder Gewebe in eine Membran, welche für Nährmittel und andere für das Aufrechterhalten der Lebefähigkeit und des Metabolismus erforderliche Substanzen und für metabolische Produkte permeabel ist, für Bakterien und für ein Molekulargewicht über einem bestimmten Wert aufweisende Substanzen jedoch undurchlässig ist, um auf diese Weise für das immunologische Abstossen fremden Gewebes verantwortliche Mittel auszuschliessen. Andere Ziele der Erfindung umfassen die Schaffung von eingekapseltem lebendem Gewebe, welches für die Erzeugung von Hormonen, wie z.B. Insulin, und für die Bewirkung komplexer chemischer Veränderungen im in vivo-Gewebe brauchbar ist. Auf diese Weise kann man ein Insulin erzeugendes System, ein Körperflüssigkeiten entgiftendes System und eingekapselte Aktivkohle herstellen.
Die beiliegende Zeichnung veranschaulicht in schemati-scher Weise eine bevorzugte Methode für das Einkapseln von für das erfindungsgemässe Verfahren verwendbarem lebendem Gewebe, sowie die in Frage kommende Mikrokap-sel.
Die einzukapselnden Gewebe, Organellen oder Zellen werden in an sich bekannter Weise in fein verteilter Form hergestellt und in einem für die Aufrechterhaltung und das Fördern der vor sich gehenden metabolischen Prozesse des in Frage stehenden Gewebes geeigneten, wässrigen Medium suspendiert. Für diesen Zweck geeignete Medien sind im Handel zugänglich. Der mittlere Durchmesser des einzukapselnden Materials kann weitgehend zwischen weniger als 1 Mikron und mehreren Millimetern schwanken. Langerhans-Inseln von Menschen und Tieren besitzen im allgemeinen einen Durchmesser von 140 bis 200 Mikron. Gewünschtenfalls kann man selbstverständlich individuelle Zellen, wie z.B. Pankreas-B-Zellen, -A-Zellen, -D-Zellen oder verschiedene Mischungen davon, ganze Langerhans' Inseln, individuelle Hepatozyten, Organellen oder andere Gewebeeinheiten einkapseln. Auch Mikroorganismen sowie nichtlebende Mate- * rialien, wie z.B. biologische Materialien, können eingekapselt werden.
Die Dauer der Lebefahigkeit solcher lebenden Bestandteile hängt unter anderem von der Zugänglichkeit der erforderlichen Nährstoffe, dem Sauerstofftransfer, der Abwesenheit toxischer Substanzen im Medium und vom pH-Wert des Mediums ab. Bisher war es nicht möglich, solche lebende Materialien in einer physiologisch verträglichen Umgebung bei gleichzeitiger Einkapselung beständig zu erhalten. Das Problem lag darin, dass die für die Membranbildung erforderlichen Bedingungen für das Gewebe tödlich oder schädlich waren. Es bestand keine Methode zur Membranbildung bei Anwesenheit eines in gesundem Zustande lebensfähigen Gewebes. Es wurde nun festgestellt, dass gewisse wasserlösliche Substanzen, welche mit dem lebenden Gewebe physiologisch verträglich sind und unter Bildung einer die Form beibehaltenden, kohärenten Masse in einen wasserunlöslichen Zustand übergehen können, zur Bildung einer «temporären Kapsel» oder einer Schutzschicht um die Gewebepartikel verwendet werden können. Solche Materialien werden vorzugsweise bei niedriger Konzentratin dem Gewebekulturmedium zugesetzt. Die Lösung wird hierauf zu Tropfen verformt, welche neben dem Aufrechterhaltungsmedium Gewebe enthalten, worauf die Lösung unmittelbar in einen wasserunlöslichen Zustand übergeführt und mindestens an der Oberflächenschicht geliert wird. Hierauf werden die die Form beibehaltenden temporären Kapseln mit einer permanenten, semipermeablen Membran ausgerüstet. In jenen Fällen, in denen das zur Bildung der temporären Kapseln verwendete Material dieszulässt, kann der Kapselinhalt nach der Bildung der permanenten Membran erneut verflüssigt werden. Dies geschieht durch erneutes Zustandebringen solcher Bedingungen im Medium, bei welchen das Material löslich ist.
Die zur Bildung der temporären Kapseln zu verwendenden Materialien sind wasserlösliche, natürliche oder synthetische Polysaccharidmassen. Solche Materialien sind im Handel zugänglich. Dabei handelt es sich vorwiegend um aus vegetabilischen Materialien extrahierte Substanzen, wie sie häufig als Zusatzstoffe zu verschiedenen Nahrungsmitteln eingesetzt werden. Natriumalginat gilt als besonders bevorzugtes wasserlösliches Material. Andere verwendbare, gummiartige Materialien sind Guargummi, Gummi-arabi-kum, Carrageenan, Pektin, Traganthgummi, Xanthangum-mi oder deren saure Fraktionen.
Diese Materialien enthalten glucosidvernetzte Saccharid-ketten. Sie enthalten freie saure Gruppen, die häufig in Form von Salzen mit Alkalimetallionen, z. B. Natrium, vorhanden sind. Wird ein mehrwertiges Ion, z.B. Calcium oder Strontium, durch ein Alkalimetallion ausgetauscht, so werden die flüssigen, wasserlöslichen Polysaccharidmoleküle vernetzt, und zwar unter Bildung eines wasserunlöslichen, seine Form beibehaltenden Gels, das nach der Entfernung der Ionen durch Ionenaustausch oder mit Hilfe eines Sequistriermittels wiederum löslich gemacht werden kann. Im wesentlichen lassen sich beliebige mehrwertige, Salze bildende Ionen einsetzen. Vorzugsweise wird man physiologisch verträgliche Ionen, wie z.B. Calcium, verwenden. Dadurch wird das Halten des Gewebes im lebenden Zustande begünstigt. Für emp-; findlichere Materialien können auch andere mehrwertige Kationen eingesetzt werden.
Eine typische Zubereitung aus Gewebe-Gewebemedium-Gummimasse besteht aus gleichen Volumina Gewebe in seinem Medium und einer 1 bis 2%-igen Lösung des gummiartigen Materials in physiologischer Kochsalzlösung. Bei Verwendung von Natriumalginat wird mit Erfolg eine 1,0 bis 1,5%-ige Lösung verwendet.
In der nächsten Stufe des Einkapselungsverfahrens wird die das Gewebe enthaltende gummiartige Lösung zu Tröpfchen gewünschter Grösse verformt. Hierauf werden die Tröpfchen unmittelbar zu die Form beibehaltenden sphärischen oder spheroidischen Massen geliert. Eine für die Durchführung dieser Arbeitsstufen verwendbare Vorrichtung ist in der beiliegenden Zeichnung BC veranschaulicht. Ein eine wässrige Lösung mit mehrwertigem Kation, z.B. eine 1,5%-ige Calciumchloridlösung, enthaltender Becher 10 wird mit einem magnetischen Rührstab 11 und einem Rührwerk 12 ausgerüstet. Der Rührmechanismus wird so betätigt, dass ein Wirbel 14 mit einem hohlen Zentrum 16 entwickelt wird. Ein Kapillarrohr 18 mit bestimmtem Innendurchmesser wird innerhalb des Hohlbereiches 16 des Wirbels angeordnet und mit einem Vibrator 20 ausgerüstet. Die das Gewebe und das gelöste, gummiartige Material enthaltende Suspension wird durch die Kapillare zugeführt. Die Wirkung der Oberflächenspannung, welche die Bildung von verhältnismässig grossen Tropfen mit sich bringen würde, wird mit Hilfe des Vibrators so verringert, dass Tröpfchen, wie durch 22 erläutert, von einer mit dem Innendurchmesser des Kapillarrohres vergleichbaren Grösse aus der Spitze des Kapillarrohres weggeschüttelt werden. Diese Tröpfchen kommen unmittelbar mit der Lösung in Berührung, wobei sie Calciumionen absorbieren. Daraus ergibt sich eine Quervernetzung des Gels und die Bildung einer die Form behaltenden, hoch viskosen, schützenden Temporärkapsel, welche das suspendierte Gewebe und sein Medium enthält. Die Kapseln werden in der Lösung als separate Phase gesammelt und durch Ansaugen abgetrennt.

Gemäss einer Ausführungsform des Verfahrens wird eine kleine Menge des Polymers der für das permanente Quervernetzen des gummiartigen Materials verwendeten Art gleichzeitig mit den mehrwertigen Ionen und der Lösung (oder einer das verwendete gummiartige Material zu gelieren vermögenden anderen Lösung) einbezogen. Daraus ergibt sich die Bildung von permanenten Vernetzungen. Kapseln dieser Art besitzen gewisse Vorteile, wenn es darum geht, das Gewebe möglichst beständig zu erhalten.
Bei der nächsten Arbeitsstufe wird eine semipermeable Membran um die Oberfläche der temporären Kapseln gebildet.
Die in der beiliegenden Zeichnung als Stufe D wiedergegebene Methode zur Bildung der Membran ist die permanente Vernetzung der Oberflächenschichten der Tröpfchen, indem man sie mit einer wässrigen Lösung eines Polymers behandelt, welches Gruppen enthält, die mit den sauren Gruppen in den Gelmolekülen zu reagieren vermögen. Gewisse langkettige quaternäre Ammoniumsalze lassen sich unter gewissen Umständen für diesen Zweck verwenden. Verwendet man saure gummiartige Materialien, so kann es sich um Polymere handeln, welche mit Säuren reaktionsfähige Gruppen enthalten, wie z.B. Polyäthylenimin und Polylysin. In diesen Fällen werden die Polysaccharide durch Reaktion der Carboxylgruppen mit den Amingruppen quervernetzt. Die Durchlässigkeit lässt sich durch Wahl des Molekulargewichtes des Vernetzungspolymers steuern. So wird beispielsweise eine Lösung eines Polymers mit niedrigem Molekulargewicht in einer bestimmten Zeitdauer stärker in die temporären Kapseln eindringen als ein Polymer mit hohem Molekulargewicht. Das Eindringungsausmass des vernetzenden Mittels entspricht der erreichbaren Permeabilität. Im allgemeinen werden die Porengrössen mit zunehmendem Molekulargewicht und geringerer Durchdringung grösser sein. Im allgemeinen lassen sich Polymere mit einem Molekulargewicht von 3000 bis 100 000 Dalton oder mehr je nach der Reaktionsdauer, der Konzentration der Polymerlösung und dem gewünschten Durchlässigkeitsausmass verwenden. Erfolgreiche Bedingungen bestehen in der Verwendung von Polylysin mit einem durchschnittlichen Molekulargewicht von ungefähr 35 000 Dalton bei einer Reaktionsdauer von 2 Minuten und unter Rühren einer physiologischen Kochsalzlösung, welche 0,0167% Polylysin enthält. Für das Steuern der Durchlässigkeit geeignete, optimale Reaktionsbedingungen für ein gegebenes System lassen sich leicht empirisch bestimmen.
Die Wahl des Vernetzungsmittels bzw. der Vernetzungsmittel bestimmt gleichfalls die in vivo-Verweilzeit der Kapseln. Im oben beschriebenen System umfasst die permanente Kapselmembran ein entweder mit einem Polypeptid und/ oder Protein, z.B. Polylysin, oder einer synthetischen Substanz, wie z.B. Polyäthylenimin, vernetztes Polysaccharid (eine leicht injizierbare Substanz). Die Polymeren schwanken je nach dem, wie sie in vivo digeriert werden können. Gewisse Polymere, wie z.B. Protein, lassen sich leicht digerieren, während andere sich leicht zersetzen und noch andere unverändert bleiben. Das erfindungsgemässe Verfahren macht sich eine Quervernetzung mit einem oder mehreren Polymeren zunutze, um auf diese Weise Kapseln zu erzeugen, welche einen bestimmten Auflösungsgrad in vivo aufweisen, wobei dieser Auflösungsbereich im allgemeinen zwischen einigen Stunden oder Tagen liegt, um endgültig zu sein. Nachstehend findet sich ein Beispiel darüber, wie Kapseln erzeugt werden, welche mindestens 2 Monate innerhalb der Bauchhöhle von Ratten intakt bleiben. Die Erfindung sei aber keineswegs auf diese bestimmten Kapselmembranen oder auf Kapseln mit einer derartigen in vivo-Laufzeit eingeschränkt. Die optimale in vivo-Lebensdauer der Mikrokapseln hängt nämlich vom beabsichtigten Zweck und Gebrauch und von der Stelle der Implantation ab. Der Fachmann weiss, wie er Mikrokapseln empirisch mit einer bestimmten in vivo-Lebensdauer herstellen kann.
In diesem Zeitpunkte der Einkapselung kann man die Kapseln, welche eine permanente semipermeable Membran aufweisen, welche eine gelierte Lösung eines gummiartigen Materials, ein mit dem Gewebe verträgliches Kulturmedium und Gewebepartikel umhüllt, sammeln. Soll das Gewebe in einer Schutzumgebung bloss aufbewahrt werden, so braucht man keine weiteren Schritte einzuleiten. Ist aber innerhalb der Kapseln und durch die Membranen hindurch ein Transfer der Masse vorzunehmen, so ist es vorteilhaft, das Gel wiederum in seine wasserlösliche Form zu verflüssigen. Dies kann dadurch geschehen, dass man die Bedingungen, unter welchen das gummiartige Material eine Flüssigkeit ist, wieder herstellt, indem man die verwendeten Calcium- oder anderen mehrwertigen Kationen entfernt. In den in Gegenwart von mehrwertigen Kationen unlöslichen Gelen kann man das Medium in der Kapsel durch blosses Eintauchen der Kapseln in eine mit Phosphat gepufferte Kochsalzlösung, welche Alkalimetallionen und Wasserstoffionen enthält, wiederum auflösen. Einwertige Ionen führen zu einem Austausch mit den Calciumionen oder anderen mehrwertigen Ionen innerhalb des gummiartigen Materials, wenn, wie dies durch Fig. E der beiliegenden Zeichnung ersichtlich ist, die Kapseln unter Rühren in die Lösung eingetaucht werden. Für den gleichen Zweck kann man auch andere Salze, wie z. B. Natriumeitrat, verwenden.
Je nach der Art der verwendeten Technik für die Herstellung der semipermeablen Membran kann es schliesslich wünschenswert sein, die Kapseln so zu behandeln, dass sie freie Aminogruppen jener Art, welche ansonsten den Kapseln eine Neigung zur Klumpenbildung geben könnten, bindet. Dies kann beispielsweise durch Eintauchen der Kapseln in eine Natriumalginatlösung geschehen.
Die erfindungsgemäss erhältlichen Kapseln ermöglichen es, durch Injektion von eingekapseltem, fein verteiltem Gewebe, von mehrzelligen Fraktionen davon oder von individuellen Zellen in eine bestimmte Stelle eines tierischen oder menschlichen Körpers diesen Körper wenigstens temporär mit der dem Gewebe eigenen physiologischen Funktion zu versehen. Diese Arbeitsweise bringt die beiden folgenden Vorteile mit sich, nämlich man braucht nicht zu einer chirurgischen Implantation zu greifen (obgleich die Kapseln gewünschtenfalls auch chirurgisch implantiert werden können) und man vermeidet mit Erfolg die Probleme der Immunreaktion und der natürlichen physikalischen Abstossung. Die Kapselmembranen werden vorzugsweise aus Substanzen bestehen, welche nach der Verwertung des Gewebes abgestos-sen werden. Wie bereits oben erwähnt worden ist, kann dies durch Verwendung eines Quervernetzers erfolgen, welcher einem in vivo-Zusammenbruch widersteht, wodurch eine wertvolle in vivo-Lebensdauer erreicht wird.
Die nachstehenden Beispiele sollen die Erfindung näher erläutern, ohne sie einzuschränken.
Beispiel 1
Langerhans-Inseln wurden aus Rattenpankreas gewonnen und einer kompletten Gewebekultur (CMRL-1969 Con-naught Laboratories, Toronto, Canada) bei einer Konzentration von ungefähr 103 Inseln pro ml hinzugegeben. Die Gewebekultur enthielt sämtliche für eine kontinuierliche Lebefähigkeit der Inseln sowie der Aminosäuren, welches durch die B-Zellen für die Herstellung von Insulin verwendet wurde, erforderlichen Nährstoffe. Vier Zehntel eines Milliliters der Inselsuspension wurden hierauf zu einhalb Milliliter

1,2%-igem Natriumalginat (Sigma Chemical Company) in physiologischer Kochsalzlösung hinzugegeben.
Hierauf wurden 80 ml einer 1,5%-igen Calciumchloridlö-sung in einen 150 ml Becher, welcher mit einem Rührwerk versehen ist, eingebracht und so gerührt, dass die Bildung eines Wirbels, welcher im Zentrum einen konisch geformten Hohlraum bildet, erzeugt wurde. Hierauf wurde ein Kapillarrohr aus Glas mit allmählich abnehmendem Durchmesser und in einer Spitze mit einem inneren Durchmesser von ungefähr 300 p. endend mit einem Vibrationsgerät (60 Umdrehungen pro Sekunde) ausgerüstet. Die Kapillarspitze wurde hierauf in das Zentrum des Wirbels eingesetzt, das Vibrationsgerät eingeschaltet und die aus Natriumalginat, Kulturmedium und Gewebe bestehende Suspension mit einer Infusionspumpe hindurchgezwängt. Von der Spitze des Kapillarrohrs wurden auf diese Weise Tröpfchen mit einem Durchmesser von 300 bis 400 Mikron ausgestossen und sofort in die Calciumlösung eingeführt.
Nach Ablauf von 10 Minuten wurde der Rührer abgestellt und die obenauf fliessende Lösung durch Ansaugen entfernt. Die gelierten Kapseln wurden hierauf in einen Becher übertragen, welcher 15 ml einer Lösung enthielt, welche aus 1 Teil einer 2%-igen 2-(Cyclohexylamino)-äthansulfon-säurelösung in 0,6%-igem Natriumchlorid (isotonisch, pH = 8,2), verdünnt mit 20 Teilen 1%-igem Calciumchlorid, bestand. Nach 3 minütigem Eintauchen wurden die Kapseln zweimal in 1 %-iger Calciumchloridlösung gewaschen.
Die Kapseln wurden hierauf in eine 32 ml Lösung eingebracht, welche 1:80 1 %-iges Polylysin (durchschnittliches Molekulargewicht 35 000 atomare Masseneinheiten) in physiologischer Kochsalzlösung enthielt. Nach 3 Minuten wurde die Polylysinlösung dekantiert. Die Kapseln wurden hier- > auf mit l%iger Calciumchloridlösung gewaschen und erneut während 3 Minuten in einer Lösung von Polyäthylenimin (Molekulargewicht 40 000 bis 60 000), erhalten durch Verdünnen einer 3,3%-igen Polyäthyleniminlösung in Mor-pholino-propansulfonsäure-Pufferlösung (0,2 molar, pH = 6) mit genügend 1 %-iger Calciumchloridlösung, um letztendlich eine Polymerkonzentration von 0,12% zu erhalten, suspendiert. Die so erhaltenen Kapseln, welche permanente semipermeable Membranen enthielten, wurden hierauf zweimal mit 1 %-iger Calciumchloridlösung und zweimal mit physiologischer Kochsalzlösung gewaschen und dann mit 10 ml einer 0,12%-igen Alginsäurelösung vermischt.
Diese Kapseln widerstanden einer Klumpenbildung, wobei manche der Kapseln in wahrnehmbarer Weise Langer-hans-Inseln zeigten. Das in den Kapseln vorhandene Gel wurde durch Eintauchen der Kapseln in eine Mischung einer Kochsalzlösung und Citratpufferlösung (pH = 7,4) während 5 Minuten erneut flüssig gemacht. Schliesslich wurden die Kapseln in einem CMLR-69-Medium suspendiert.
Unter dem Mikroskop betrachtet, zeigen diese Kapseln das in der beiliegenden Zeichnung widergegebene Aussehen. Sie besitzen eine äusserst dünne Membran 24, welche eine Insel 26 umschliesst, worin einzelne Zellen 28 festgestellt werden können. Moleküle mit einem Molekulargewicht bis zu ungefähr 100 000 können durch die Membran 24 hindurchgehen. Auf diese Weise können Sauerstoff, Aminosäuren, Nährstoffe und Plasmakomponenten, welche in den Kulturmedien verwendet werden (z.B. fötale Kalbplasmakomponenten), die Inseln erreichen und eine Exkretion vom Insulin ermöglichen.
Beispiel 2
Nach wiederholtem Waschen in physiologischer Kochsalzlösung wurden nach dem Beispiel 1 erhaltene Mikrokap-seln, welche ungefähr 15 Inseln aufwiesen, in 3 ml CMRL-1969 suspendiert. Nach Ablauf von 8 Tagen in Gegenwart von 600 mg/dl Glucose haben die Kapseln 67 Einheiten/ml Insulin in 1 l/2 Stunden abgesondert. Bei einem zweiten Anlauf wurden in der gleichen Zeitspanne 68 Einheiten/ml Insulin erzeugt. Einwöchige Kapseln im gleichen Medium, jedoch in Gegenwart von 100 ml/dl Glucose ergaben im ersten Anlauf einen Ausstoss von 25 Einheiten/ml Insulin in 80 Minuten und im zweiten Anlauf einen solchen von 10 Einheiten/ml.
Beispiel 3
Diabetes-Ratten mit Blutglucosespiegeln im Bereiche von 500 bis 700 mg/dl wurden jeweils mit ungefähr 103 Inseln, welche gemäss Angaben gemäss Beispiel 1 eingekapselt und in physiologischer Kochsalzlösung suspendiert worden waren, behandelt. Die Kapseln wurden durch Injektion in die Bauchhöhle mit Hilfe von 19 Nadeln, die mit einer Spritze versehen waren, eingeführt. Die Blutzuckerspiegel wurden täglich gemessen und waren gleichmässig auf einem Wert von weniger als 300 mg/dl. Nach 2 Monaten getötete Tiere zeigten keine Zeichen irgendeiner toxischen Reaktion an der Implantationsstelle. Die Kapseln wurden aus den getöteten Tieren nach 2 monatiger Lebensdauer in vivo intakt entnommen und zeigten keine Anzeichen der Zersetzung.
Beispiel 4 Einkapselung von Hepatozyten
Es wurde wie beim Beispiel 1 gearbeitet mit dem Unterschied, dass 0,5 ml einer Leberzellensuspension in einer Hank' Lösung anstelle der 0,4 ml Insel-Suspension verwendet wurden. Die Lebefähigkeit des Lebergewebes wurde durch Farbstoffausschlusstechnik (Trypanblauausschluss) demonstriert. Bekanntlich kann Lebergewebe in vitro aus der Umgebung Toxine aufnehmen. Daher werden Toxine mit genügend niedrigem Molekulargewicht, um durch die semipermeable Membran hindurchzugehen, aufgenommen und durch das Gewebe zerstört. Im wesentlichen sind sämtliche durch die Leber behandelten Toxine niedrigmolekulare Materialien. Die Toxine können aber Proteinkomplexe bilden. Die Kapselpermeabilität kann je nach Notwendigkeit schwanken.
Beispiel 5
Die Arbeitsweise von Beispiel 1 wurde wiederholt mit dem Unterschied, dass Aktivkohle in Partikelform direkt in der Natriumalginatlösung suspendiert wurde, wobei die 0,5 ml Gewebesuspension wegfiel und Polylysin mit einem durchschnittlichen Molekulargewicht von 35 000 als Vernetzungsmittel verwendet wurde. Solange die Kohlepartikel kleiner waren als die kleinsten inneren Durchmesser der für die Tröpfchenbildung verwendeten Kapillare ergab sich Kohle von hoher Oberfläche, umgeben durch eine semipermeable Membran. Diese Tröpfchen verhinderten in wirksamer Weise das Entweichen von Kohleteilchen oder -staub, konnten aber zum Absorbieren von Materialien mit mittlerem Molekulargewicht von bis zu ungefähr 2000 Dalton aus der Flüssigkeit, welche die Kapseln passierte, Verwendung finden.
Das vorliegende Verfahren lässt sich auch mit anderen lebenden Zellen, einschliesslich roter Blutkörperchen unter Verwendung von Serum als Medium, Spermienzellen unter Verwendung von Samen als Medium und Baker' Hefe verwenden. Der Fachmann wird es schätzen, dass viele andere Materialien ausser den oben erwähnten eingekapselt werden können und dass die Durchlässigkeit je nach Anwendungsgebiet gesteuert werden kann.

Claims (14)
- 657 7862PATENTANSPRÜCHE1. Verfahren zum Einkapseln eines Kernmaterials innerhalb einer semipermeablen Membran, dadurch gekennzeichnet, dass manA) das Material in einem wässrigen Medium, welches ein wasserlösliches, freie saure Gruppen enthaltendes Polysaccharid enthält, suspendiert;B) die so erhaltene Suspension in Tropfenform überführt;C) die Tröpfchen mit einer Lösung von mehrwertigen Kationen behandelt, um die Tröpfchen unter Bildung von diskreten, die Form beibehaltenden, in Wasser unlöslichen, temporären Kapseln zu gelieren; undD) die Oberflächenschichten der besagten temporären Kapseln permanent vernetzt, um eine semipermeable Membran um die besagten Tröpfchen zu bilden, indem man sie mit einem reaktionsfähige Gruppen, welche mit den sauren Gruppen des besagten Polysaccharids zu reagieren vermögen, enthaltenden Polymeren behandelt, wobei besagtes Polymer ein Molekulargewicht von 3000 Daltons oder mehr hat.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man nach der Bildung der besagten Membranen das gelartige Material erneut auflöst.
- 3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass das wasserlösliche Polysaccharid Natriumalginat und die mehrwertige Kationenlösung eine Calciumlösung sind.
- 4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, dass man eine weitere Arbeitsstufe einsetzt, um die in den Kapseln vorhandenen Calciumionen zu entfernen, um auf diese Weise das gelierte Alginat innerhalb der Membranen der besagten Kapseln erneut aufzulösen.
- 5. Verfahren nach Anspruch 3, dadurch gekennzeichnet, dass das besagte Kernmaterial ein Gewebe eines Säugetieres oder eines Menschen, nämlich Langerhans'-Inseln, Leber und individuellen Zellen davon, ist.
- 6. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass besagtes wässriges Medium ein für das einzukapselnde Kernmaterial physiologisch verträgliches Medium ist.
- 7. Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass besagte Membran für Nährmittel der Zellen und für die Produkte des Zellenmetabolismus permeabel ist.
- 8. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass besagtes Kernmaterial aus lebendem Gewebe oder individuellen Zellen besteht.
- 9. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass das wässrige, insbesondere physiologisch verträgliche Medium ein vollständiges Gewebekulturmedium ist, welches genügt, um das besagte Kernmaterial in vitro aufzubewahren.
- 10. Verfahren nach Anspruch 8 oder 9, dadurch gekennzeichnet, dass besagte Membran für Nährmittel der Zellen und für die Produkte des Zellenmetabolismus permeabel ist.
- 11. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass besagte Membran aus einem Carboxylgruppen enthaltenden Polysaccharid und einem Aminogruppen enthaltenden Polymeren, welche durch Reaktionen zwischen den Carboxylgruppen und den Aminogruppen miteinander quervernetzt sind, besteht.
- 12. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass das besagte vernetzende Polymer Polylysin oder Polyäthylenimin ist.
- 13. Verfahren nach Anspruch 12, dadurch gekennzeichnet, dass das durchschnittliche Molekulargewicht des besagten Polymers bei ungefähr 35 000 Dalton liegt.
- 14. Kapseln, hergestellt nach dem Verfahren gemäss einem der Ansprüche 1 bis 13.
