Claims (1)
- 【特許請求の範囲】 1.式(I−1)ないし(I−4)からなる式(I): [式中、 XはOまたはS; R1は水素、アミノ、ハロ、C1-6アルキル、ヒドロキシまたはC1-6アルコキシ ; R2は水素、ハロ、C1-6アルキル、C1-6アルコキシ、ニトロ、アミノまたはC1 -6 アルキルチオ; R3は水素、ハロ、C1-6アルキル、C1-6アルコキシまたはアミノ; R4は水素またはC1-6アルキルを意味する] [式中、 XはOまたはS; Aは単結合、−CH2−またはCOを表すか、またはAは(CH2)a−E−(C H2)b(ここに、aおよびbの一方は0であり、他方は0または1であり、Eは O、SまたはNHである); R1は水素、アミノ、ハロ、C1-6アルキル、ヒドロキシまたはC1-6アルコキシ ; R2は水素、ハロ、C1-6アルキル、C1-6アルコキシ、ニトロ、アミノまたはC1 -6アルキルチオ; R3は水素、ハロ、C1-6アルキル、C1-6アルコキシまたはアミノ; R4は水素またはC1-6アルキルを意味する] [式中、 XはOまたはS; Aは単結合、−CH2−またはCOを表すか、またはAは(CH2)a−E−(C H2)b(ここに、aおよびbの一方は0であり、他方は0または1であり、Eは O、SまたはNHである); fおよびgは、共に、水素であるか、または一緒になって結合を形成し; R1は水素、アミノ、ハロ、C1-6アルキル、ヒドロキシまたはC1-6アルコキシ ; R2は水素、ハロ、C1-6アルキル、C1-6アルコキシ、ニトロ、アミノまたはC1 -6 アルキルチオ; R3は水素、ハロ、C1-6アルキル、C1-6アルコキシまたはアミノ; R4は水素またはC1-6アルキルを意味する] [式中、 XはOまたはS; R1は水素、アミノ、ハロ、C1-6アルキル、ヒドロキシまたはC1-6アルコキシ ; R2は水素、ハロ、C1-6アルキル、C1-6アルコキシ、ニトロ、アミノまたはC1 -6 アルキルチオ; R3は水素、ハロ、C1-6アルキル、C1-6アルコキシまたはアミノ; R4’およびR”は、独立して、水素またはC1-6アルキルを意味する]; 式(I−1)ないし式(I−4)中: YはOまたはNH; Zは式(a)、(b)または(c): (式中、 n1は0、1、2、3または4;n2は0、1、2、3または4;n3は2、3、 4または5; qは0、1、2または3;pは0、1または2;mは0、1または2; R5は水素、C1-12アルキル、アラルキルであるか、またはR5は(CH2)z− R10であり、ここにzは2または3であって、R10はシアノ、ヒドロキシル、C1-6 アルコキシ、フェノキシ、C(O)C1-6アルキル、COC6H5、CONR11 R12、NR11COR12、SO2NR11R12またはNR11SO2R12から選択され、 ここに、R11およびR12は水素またはC1-6アルキルであり; および R6、R7およびR8は、独立して、水素またはC1-6アルキル;および R9は水素またはC1-10アルキルを意味する) で示される基である化合物またはその医薬上許容される塩、および5−HT4レ セプターアンタゴニスト活性を有する医薬の製造における、式(I)の化合物ま たはその医薬上許容される塩、あるいはCO−Y結合が複素環式バイオアイソス ターにより置換されている式(I)の化合物の使用。 2. 式(I−1)中: R1が水素またはアミノであり、R2が水素またはハロであり、R3が水素また はハロであって、R4が水素である; 式(I−2)中: R1が水素またはアミノであり、R2が水素またはハロであり、R3が水素また はハロであって、R4がしばしば水素である; 式(I−3)中: R1が水素またはアミノであり、R2が水素またはハロであり、R3が水素また はハロであって、R4が水素である; 式(I−4)中: R1が水素またはアミノであり、R2が水素またはハロであり、R3が水素また はハロであって、R4’およびR4”が水素である請求項1記載の化合物。 3.CO−Y−Zに結合する基が本明細書に記載のいずれかの実施例に含まれ るものである請求項1または2記載の化合物。 4.Zが式(a)で示されるものであり、(CH2)n1がアザサイクルの炭素 原子で結合している請求項1〜3のいずれか1つに記載の化合物。 5.ZがN−置換4−ピペリジルメチルである請求項4記載の化合物。 6.N−置換基がC2またはそれ以上のアルキルであるか、または所望により 置換されていてもよいベンジルである請求項5記載の化合物。 7.(1−ブチル−4−ピペリジニルメチル)−4−アミノ−5−クロロベン ゾ[b]フラン−7−カルボキシアミド、 (1−ブチル−4−ピペリジニルメチル)ベンゾチオフェン−7−カルボキシラ ート、 (1−ブチル−4−ピペリジニルメチル)−5−クロロベンゾ[b]フラン−7 −カルボキシラート、または (1−ブチル−4−ピペリジニルメチル)−5−クロロベンゾ[b]フラン−7 −カルボキシアミドである化合物。 8.1−ブチルピペリジン−4−イルメチルジベンゾフラン−4−カルボキシ ラート、 (1−ブチル−4−ピペリジニルメチル)−9H−キサンテン−4−カルボキシ ラート、 (1−ブチル−4−ピペリジニルメチル)−9−オキソ−9H−キサンテン−4 −カルボキシラート、 (1−ブチル−4−ピペリジニルメチル)−10−フェノキサジン−4−カルボ キシラート、 (1−ブチル−4−ピペリジニルメチル)−1−アミノ−2−クロロジベンゾフ ラン−4−カルボキシアミド、 (1−ブチル−4−ピペリジニルメチル)−1−アミノ−2−クロロジベンゾフ ラン−4−カルボキシラート、または (1−ブチル−4−ピペリジニルメチル)−2−クロロジベンゾフラン−4−カ ルボキシラートである化合物。 9.(1−ブチル−4−ピペリジニルメチル)−1−アミノ−2−クロロ−5 a,6,7,8,9,9a−テトラヒドロジベンゾフラン−4−カルボキシラー ト、 (1−ブチル−4−ピペリジニルメチル)−1−アミノ−2−クロロ−5a,6 ,7,8,9,9a−テトラヒドロジベンゾフラン−4−カルボキシアミド、 (1−ブチル−4−ピペリジニルメチル)−2−クロロ−シス−5a,6,7, 8,9,9a−ヘキサヒドロジベンゾフラン−4−カルボキシラート、または (1−ブチル−4−ピペリジニルメチル)−2−クロロ−6,7,8,9−テト ラヒドロジベンゾフラン−4−カルボキシラートである化合物。 10.10−(1−ブチルピペリジン−4−イルメチル)−8−クロロ−3,4 ,5,6−テトラヒドロ−2,6−メタノ−2H−1−ベンゾキサシンカルボキ シラートである化合物。 11.医薬上許容される塩の形態である請求項7〜10のいずれか1つに記載の 化合物。 12.YがNHである請求項7〜10のいずれか1つに記載の化合物。 13.適当な酸誘導体を適当なアルコールまたはアミンと反応させることからな る、請求項1に記載のエステルまたはアミド化合物(YはOまたはNH)の製法 。 14.請求項1〜12のいずれか1つに記載の化合物と、医薬上許容される担体 とからなる医薬組成物。 15.活性治療物質として用いるための請求項1記載の化合物。 16.5−HT4レセプターアンタゴニストとして用いるための医薬の製造にお ける請求項1記載の化合物の使用。 17.胃腸障害、心臓血管系障害およびCNS障害の治療または予防において、 5−HT4レセプターアンタゴニストとして用いる請求項16記載の使用。
