Abstract
Selon l'invention, l'il-18 participe à la fois dans l'immunité innée et acquise. La bioactivité de l'il-18 est régulée négativement par la protéine de liaison à l'il-18 (il18bp), un inhibiteur naturel et hautement spécifique. Cette protéine soluble forme un complexe avec l'il-18 libre, empêchant son interaction avec le récepteur de l'il-18, neutralisant et inhibant ainsi son activité biologique. La présente invention concerne des molécules de liaison, en particulier des anticorps ou des fragments de ceux-ci, qui se lient à l'il-18 et ne se lient pas à l'il-18 liée à il18bp (complexe il-18/il-18bp). Hormis son rôle physiologique, l'il-18 a été présentée comme médiant une variété de maladies auto-immunes et inflammatoires. Les molécules de liaison des inventions peuvent être utilisées en tant que molécules thérapeutiques pour le traitement de maladies auto-immunes et inflammatoires associées à l'il-18 ou en tant qu'outils de diagnostic pour la caractérisation, la détection et/ou la mesure de l'il-18 non liée à il18bp en tant que composante du groupe total d'il-18.
